Treatment of the first leaves of barley seedlings with either 5, 10, 15 or 20 mM salicylic acid, sodium salicylate or acetylsalicylic acid resulted in significant reductions in powdery mildew infection on the upper, second leaves. In general, the greatest reduction in mildew infection on the second leaves was obtained by spraying the first leaves with a 15 mM concentration of the compounds. Although the largest reduction in mildew infection of the upper leaves was obtained when the compounds were applied to the first leaves 1–2 days before inoculation, very substantial reductions in infection were still obtained if the first leaves were treated 12 days before inoculation. The three compounds had little direct effect on mildew infection. When 14C-salicylic acid was fed to first leaves of barley seedlings, uptake was rapid and increased with time. Within 6 h, 0.2% of the salicylic acid appeared in the second leaf and by 24 h after feeding, this had increased to 1.4% (1.1 μmol salicylic acid g-1 fresh wt). The application of various phenolic acids to first leaves also led to reductions in mildew infection on the second leaves. In particular, treatment of the first leaves with 1 mM vanillic acid, isovanillic acid or syringic acid, reduced mildew infection of the second leaves by 81–87%.  相似文献

Six benzylic diamines were synthesised and examined for antifungal activity. Four of the compounds, KB 2, KB 4, KB 5 and KB 6, reduced radial growth of the oat leaf stripe pathogen Pyrenophora avenae, the largest reduction obtained with 25 μM KB 4, which reduced radial growth by 47%. Surprisingly, these four amines had no effect against infection of barley seedlings with the powdery mildew fungus Erysiphe graminis f.sp. hordei. Instead, two different amines, KB 1 and KB 3, reduced powdery mildew infection on barley. The greatest reduction was obtained with 25 μM KB 3, which reduced mildew infection by 69%. All of the amines which exhibited antifungal or fungicidal properties perturbed polyamine formation as measured by the incorporation of labelled ornithine into polyamines.  相似文献

Ethokem (Midkem Agrochemicals, Northampton, UK) and Bond (Newman Agrochemicals, Cambridge, UK) completely inhibited linear growth of Pyricularia oryzae at 1 and 2% concentrations of the compounds and strongly inhibited growth of Pyrenophora avenae in vitro . Vapor Gard (Miller Chemical & Fertilizer Corporation, Hannover, PA, USA) was less effective with relatively little inhibition of P. oryzae , but was a more effective inhibitor of P. avenae . All three compounds decreased cell lengths of both pathogens and increased cell diameters of P. avenae . Vapor Gard and Ethokem did not significantly alter cell diameters of P. oryzae , but a highly significant increase was observed with Bond. Gross changes in hyphal morphology were observed including swollen shortened cells, granulation of the cytoplasm, increased branching and collapsed empty cells. Ethokem and Bond decreased growth rate and inhibited biomass production of P. avenae .  相似文献

D,L-alpha-Difluoromethylornithine (DFMO) is an inhibitor of ornithine decarboxylase, the first enzyme in the polyamine biosynthetic pathway. Exposure of human foreskin fibroblast cells to DFMO before their infection with human strains of cytomegalovirus (CMV) resulted in a reduction in the amount of infectious virus produced. A 3-day exposure to the drug was required to elicit maximal antiviral effect. Cells exposed to DFMO at the time of infection produced normal amounts of infectious virus. Preexposure to the drug for 1, 2, or 3 days before infection resulted in at least 10-, 100-, or 1,000-fold decreases, respectively, in the amount of infectious virus produced. This decrease paralleled the loss of intracellular spermidine and was partially spared by the addition of exogenous putrescine, spermidine, or spermine (10 microM). When added 3 days before infection, DFMO depressed production of herpes simplex virus and simian CMV, as well as wild-type and laboratory prototype strains of human CMV. Although some antiviral effect was observed at a drug concentration of 1 mM, 10 mM gave a stronger effect and was the amount routinely used. At 30 mM DFMO, growth of noninfected cells was slowed but not arrested. Studies to investigate the level at which DFMO interferes with CMV replication showed that DFMO-treated, infected cells (i) exhibit a typical CMV-specific cytopathic effect, (ii) synthesize both viral proteins and viral DNA, (iii) contain at least some capsid forms, and (iv) shed greatly reduced amounts of virus particles into the growth medium. Since CMV virions, like those of herpes simplex virus, contain the polyamines spermidine and spermine, and since DFMO essentially eliminates the pool of intracellular spermidine, the possibility is suggested that this drug may exert its antiviral effect by interfering with virus assembly, perhaps at the level of DNA packaging or capsid envelopment or both.  相似文献

Targeted overexpression of an ornithine decarboxylase (ODC) transgene to mouse skin (the K6/ODC mouse) significantly enhances susceptibility to carcinogenesis. While in most strain backgrounds the predominant tumor type resulting from initiation-promotion protocols is benign squamous papilloma, K6/ODC mice on a FVB/N background develop malignant squamous cell carcinomas (SCCs) rapidly and in high multiplicity after carcinogen treatment. We have investigated the utility of polyamine-based therapy against SCCs in this model using the ODC inhibitor 2-difluoromethylornithine delivered orally. At a 2% concentration in drinking water, DFMO caused rapid tumor regression, but in most cases, tumors eventually regrew rapidly even in the presence of DFMO. The tumors that regrew were spindle cell carcinomas, an aggressive undifferentiated variant of SCC. At 1% DFMO in the drinking water, tumors also responded rapidly, but tumor regrowth did not occur. The majority of DFMO-treated SCCs were classified as complete responses, and in some cases, apparent tumor cures were achieved. The enzymatic activity of ODC, the target of DFMO, was substantially reduced after treatment with 1% DFMO and the high SCC polyamine levels, especially putrescine, were also significantly lowered. Based on the results of BrdUrd labeling and TUNEL assays, the effect of DFMO on SCC growth was accompanied by a significant reduction in tumor proliferation with no increase in the apoptotic index. These results demonstrate that SCCs, at least in the mouse, are particularly sensitive to polyamine-based therapy.  相似文献

The fluorescent hydrazide, Pro-Q Emerald 300 dye, may be conjugated to glycoproteins by a periodic acid Schiff's (PAS) mechanism. The glycols present in glycoproteins are initially oxidized to aldehydes using periodic acid. The dye then reacts with the aldehydes to generate a highly fluorescent conjugate. Reduction with sodium metabisulfite or sodium borohydride is not required to stabilize the conjugate. Though glycoprotein detection may be performed on transfer membranes, direct detection in gels avoids electroblotting and glycoproteins may be visualized within 2-4 h of electrophoresis. This is substantially more rapid than PAS labeling with digoxigenin hydrazide followed by detection with an antidigoxigenin antibody conjugate of alkaline phosphatase, or PAS labeling with biotin hydrazide followed by detection with horseradish peroxidase or alkaline phosphatase conjugates of streptavidin, which require more than eight hours to complete. Pro-Q Emerald 300 dye-labeled gels and blots may be poststained with SYPRO Ruby dye, allowing sequential two-color detection of glycosylated and nonglycosylated proteins. Both fluorophores are excited with mid-range UV illumination. Pro-Q Emerald 300 dye maximally emits at 530 nm (green) while SYPRO Ruby dye maximally emits at 610 nm (red). As little as 300 pg of alpha 1-acid glycoprotein (40% carbohydrate) and 1 ng of glucose oxidase (12% carbohydrate) or avidin (7% carbohydrate) are detectable in gels after staining with Pro-Q Emerald 300 dye. Besides glycoproteins, as little as 2-4 ng of lipopolysaccharide is detectable in gels using Pro-Q Emerald 300 dye while 250-1000 ng is required for detection with conventional silver staining. Detection of glycoproteins may be achieved in sodium dodecyl sulfate-polyacrylamide gels, two-dimensional gels and on polyvinylidene difluoride membranes.  相似文献

Peronospora cristata, causing downy mildew, has become an economically important pathogen of oilseed poppy (Papaver somniferum) since its first record in Tasmania in 1996. DOWNCAST, originally developed for downy mildew of onion, was evaluated for its ability to predict sporulation and infection of poppy by P. cristata. To assess the influence of the parameters within DOWNCAST, two modifications (POPCAST1 and POPCAST2) of the standard DOWNCAST model were also developed and tested. Initial accuracy of determination of sporulation events by DOWNCAST was 75%, while POPCAST1 (but not POPCAST2) was a significantly more accurate predictor of sporulation events (86%) than DOWNCAST (χ2 = 5.74, P = 0.025). This increase in accuracy was attributed to increasing both the rainfall threshold inhibitory to sporulation from 0.2 to 3.0 mm, and the relative humidity threshold for induction and development of sporulation from 95% to 96%. While DOWNCAST, POPCAST1 and POPCAST2 had accuracies of infection determination of 65%, 80% and 75%, respectively, chi‐squared analysis indicated no significant difference between models. Preliminary field evaluation indicated that POPCAST1 and POPCAST2 were more sensitive in detecting early infection events than DOWNCAST. Field observations indicated that conditions are frequently conducive for downy mildew spread, therefore the ability to predict early season infection events would allow growers to reduce initial infection rates, and thus inoculum loads later in the season. POPCAST1 and POPCAST2 represent a significant step towards the development of a predictive model for downy mildew in poppy crops.  相似文献

Reduced plasma retinol concentrations occur in human malaria but the benefits of supplementation remain uncertain. We assessed the in vivo efficacy of retinol administration, and its effect on lipid peroxidation, in a Plasmodium berghei murine model. Animals received vehicle (n=17) or retinol (i) before P. berghei inoculation (four doses), (ii) at parasitaemia 10-15% (three to four doses) or (iii) before and after inoculation (six to seven doses; n=15 in each group), with euthanasia on day 8 post-inoculation or when the parasitaemia exceeded 50%. Multiple-dose pre-inoculation retinol reduced endpoint parasitaemia by 24% (P=0.001 versus controls). A reduction of 18% (P=0.042) was observed when retinol was given to parasitaemic animals. Retinol was ineffective when given both before and after infection (11% reduction; P=0.47). Although retinol supplementation did not change plasma retinol concentrations, liver retinol content increased and correlated inversely with endpoint parasitaemia (r=-0.45, P=0.001). Malaria infection augmented concentrations of the free radical lipid peroxidation end-product F(2)-isoprostanes in plasma, erythrocytes and liver by 1.8-, 2.8- and 4.9-fold, respectively, but retinol supplementation had no effect on these increases. Consistent with some human malaria studies, prophylactic retinol reduces P. berghei parasitaemia. This effect relates to augmentation of tissue retinol stores rather than to retinol-associated changes in oxidant status.  相似文献

Helicobacter pylori infects half the world's population, and carriage is lifelong without antibiotic therapy. Current regimens prescribed to prevent infection-associated diseases such as gastroduodenal ulcers and gastric cancer can be thwarted by antibiotic resistance. We reported that administration of 1% D,L-α-difluoromethylornithine (DFMO) to mice infected with H. pylori reduces gastritis and colonization, which we attributed to enhanced host immune response due to inhibition of macrophage ornithine decarboxylase (ODC), the rate-limiting enzyme in polyamine biosynthesis. Although no ODC has been identified in any H. pylori genome, we sought to determine if DFMO has direct effects on the bacterium. We found that DFMO significantly reduced the growth rate of H. pylori in a polyamine-independent manner. Two other gram-negative pathogens possessing ODC, Escherichia coli and Citrobacter rodentium, were resistant to the DFMO effect. The effect of DFMO on H. pylori required continuous exposure to the drug and was reversible when removed, with recovery of growth rate in vitro and the ability to colonize mice. H. pylori exposed to DFMO were significantly shorter in length than those untreated and they contained greater internal levels of ATP, suggesting severe effects on bacterial metabolism. DFMO inhibited expression of the H. pylori virulence factor cytotoxin associated gene A, and its translocation and phosphorylation in gastric epithelial cells, which was associated with a reduction in interleukin-8 expression. These findings suggest that DFMO has effects on H. pylori that may contribute to its effectiveness in reducing gastritis and colonization and may be a useful addition to anti-H. pylori therapies.  相似文献

The ability of difluoromethylornithine (DFMO), a specific polyamine synthesis inhibitor, to interact with various biological modifiers to inhibit the colony-forming growth of human melanoma cells was determined by using the median effect principle to computer model the strength of two agent interactions. Either alpha- or gamma-IFN (interferon) in combination with DFMO resulted in a synergistic inhibition on human melanoma colony formation. For human melanoma cells which were not resistant to 13-cis RA (retinoic acid), an additive suppression on colony formation was obtained with the retinoid-DFMO combination. Dexamethasone (DEX) interacted with DFMO to yield a synergistic reduction in melanoma colony number on glucocorticoid sensitive cells and no growth enhancement with DFMO on glucocorticoid resistant melanoma lines. Human melanoma cells displayed differential long-term growth sensitivity to DFMO treatment. C8146C human melanoma cells were terminally growth-inhibited by a 96 h exposure to DFMO, in a manner which was concentration and time dependent. The proliferation of C82-7A melanoma cells was inhibited by 95% in presence of DFMO, but upon removal of DFMO the cells regained their ability to proliferate and form colonies. The simultaneous addition of DEX plus alpha-IFN plus 13-cis-RA with DFMO caused most of the human melanoma cells in these lines to become permanently growth arrested. Pre-treatment with DEX plus alpha-IFN plus 13-cis RA, but without DFMO, did not have any long term effect on the ability of melanoma cells to recover and proliferate in soft agar.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Application of 3.6 mm silicon (Si+) to the rose (Rosa hybrida) cultivar Smart increased the concentration of antimicrobial phenolic acids and flavonoids in response to infection by rose powdery mildew (Podosphaera pannosa). Simultaneously, the expression of genes coding for key enzymes in the phenylpropanoid pathway (phenylalanine ammonia lyase, cinnamyl alcohol dehydrogenase, and chalcone synthase) was up-regulated. The increase in phenolic compounds correlated with a 46% reduction in disease severity compared with inoculated leaves without Si application (Si-). Furthermore, Si application without pathogen inoculation induced gene expression and primed the accumulation of several phenolics compared with the uninoculated Si- control. Chlorogenic acid was the phenolic acid detected in the highest concentration, with an increase of more than 80% in Si+ inoculated compared with Si- uninoculated plants. Among the quantified flavonoids, rutin and quercitrin were detected in the highest concentrations, and the rutin concentration increased more than 20-fold in Si+ inoculated compared with Si- uninoculated plants. Both rutin and chlorogenic acid had antimicrobial effects on P. pannosa, evidenced by reduced conidial germination and appressorium formation of the pathogen, both after spray application and infiltration into leaves. The application of rutin and chlorogenic acid reduced powdery mildew severity by 40% to 50%, and observation of an effect after leaf infiltration indicated that these two phenolics can be transported to the epidermal surface. In conclusion, we provide evidence that Si plays an active role in disease reduction in rose by inducing the production of antifungal phenolic metabolites as a response to powdery mildew infection.  相似文献

At least two types of quinone reductases are present in plants: (1) the ζ-crystallin-like quinone reductases (QR1, EC 1.6.5.5) that catalyze the univalent reduction of quinones to semiquinone radicals, and (2) the DT-diaphorase-like quinone reductases (QR2, EC 1.6.99.2) that catalyze the divalent reduction of quinones to hydroquinones. QR2s protect cells from oxidative stress by making the quinones available for conjugation, thereby releasing them from the superoxide-generating one electron redox cycling, catalyzed by QR1s. Two genes, putatively encoding a QR1 and a QR2, respectively, were isolated from an expressed sequence tag collection derived from the epidermis of a diploid wheat Triticum monococcum L. 24 h after inoculation with the powdery mildew fungus Blumeria graminis (DC) EO Speer f. sp. tritici Em. Marchal. Northern analysis and tissue-specific RT-PCR showed that TmQR1 was repressed while TmQR2 was induced in the epidermis during powdery mildew infection. Heterologous expression of TmQR2 in Escherichia coli confirmed that the gene encoded a functional, dicumarol-inhibitable QR2 that could use either NADH or NADPH as an electron donor. The localization of dicumarol-inhibitable QR2 activity around powdery mildew infection sites was accomplished using a histochemical technique, based on tetrazolium dye reduction.  相似文献

Soil bacteria such as pseudomonads may reduce pathogen pressure for plants, both by activating plant defence mechanisms and by inhibiting pathogens directly due to the production of antibiotics. These effects are hard to distinguish under field conditions, impairing estimations of their relative contributions to plant health. A split-root system was set up with barley to quantify systemic and local effects of pre-inoculation with Pseudomonas fluorescens on the subsequent infection process by the fungal pathogen Fusarium graminearum. One root half was inoculated with F. graminearum in combination with P. fluorescens strain CHA0 or its isogenic antibiotic-deficient mutant CHA19. Bacteria were inoculated either together with the fungal pathogen or in separate halves of the root system to separate local and systemic effects. The short-term plant response to fungal infection was followed by using the short-lived isotopic tracer (11)CO(2) to track the delivery of recent photoassimilates to each root half. In the absence of bacteria, fungal infection diverted carbon from the shoot to healthy roots, rather than to infected roots, although the overall partitioning from the shoot to the entire root system was not modified. Both local and systemic pre-inoculation with P. fluorescens CHA0 prevented the diversion of carbon as well as preventing a reduction in plant biomass in response to F. graminearum infection, whereas the non-antibiotic-producing mutant CHA19 lacked this ability. The results suggest that the activation of plant defences is a central feature of biocontrol bacteria which may even surpass the effects of direct pathogen inhibition.  相似文献

In two experiments, winter wheat (Triticum aestivum cv. Cerco) was grown in 350 (ambient) and 700 μmol mol-1 (elevated) atmospheric CO2 concentrations. In the first experiment, plants were grown at five levels of nitrogen fertilization, and in the second experiment, plants were grown at three levels of water supply. All plants were infected with powdery mildew, caused by the fungus Erysiphe graminis. Plants grown in elevated atmospheric CO2 concentrations had significantly reduced % shoot nitrogen contents and significantly increased % shoot water contents. At elevated atmospheric CO2 concentrations, where plant nitrogen content was significantly reduced, the severity of mildew infection was significantly reduced, and where host water content was significantly increased, the severity of mildew infection was significantly increased. In a moderate water supply treatment, the plants grown in elevated atmospheric CO2 concentrations had significantly reduced nitrogen contents (9·9%) and significantly increased water content (4%), the amount of mildew infection was unchanged. The severity of mildew infection appeared to be more sensitive to host water content than to host nitrogen content.  相似文献

We previously demonstrated that repeated instillation of alpha-difluoromethylornithine (DFMO), an irreversible inhibitor of ornithine decarboxylase (ODC), inhibits (or retards) urinary bladder carcinogenesis in rats. Since ODC catalyzes the first step in polyamine synthesis, the inhibition of polyamine formation may be responsible for tumor inhibition by DFMO. The present experiment was conducted using male Fischer rats with a heterotopically transplanted urinary bladder (HTB) to determine whether the effects of DFMO are prevented by exogeneous Pu. HTBs were treated with 0.25 mg of N-methyl-N-nitrosourea (MNU) once a week for 3 weeks and the animals were then arbitrarily divided into 6 groups. Beginning one week following the last MNU treatment, all rats received twice a week instillation (0.5 ml each) as follows: Group 1 rats, normal rat urine; Group 2, 2% DFMO in urine; Group 3, 250 microM Pu and 2% DFMO in urine; Group 4, 250 microM Pu in urine; Group 5, urine for 10 weeks followed by 2% DFMO in urine; and Group 6, urine for 10 weeks followed by 250 microM Pu and 2% DFMO in urine. At 10 weeks following the last MNU instillation 5 rats from each of Groups 1 through 4 were killed for determination of urothelial polyamine levels. An additional 4 rats of Group 1 were killed at 10 weeks for histological examination. All remaining rats were killed 20 weeks after the last MNU instillation. Polyamine levels showed no significant difference among the 4 groups. The incidence of carcinoma was significantly lower in the group treated with DFMO (p less than 0.001, Group 1 vs Group 2), confirming our previous observation.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

ABSTRACT: BACKGROUND: Mononuclear platinum anticancer agents hold a pivotal place in the treatment of many forms of cancers, however, there is a potential to improve response to evade resistance development and toxic side effects. BBR3464 is a promising trinuclear platinum anticancer agent, which is a polyamine mimic. The aim was to investigate the influence of polyamine pool reduction on the cytotoxic effects of the trinuclear platinum complex BBR3464 and cisplatin. Polyamine pool reduction was achieved by treating cells with either the polyamine biosynthesis inhibitor alpha-difluoromethylornithine (DFMO) or the polyamine analogue N1,N11-diethylnorspermine (DENSPM). METHODS: A human squamous cell carcinoma cell line, LU-HNSCC-4, established from a primary head and neck tumour was used to evaluate cellular effects of each drug alone or combinations thereof. High-performance liquid-chromatography was used to quantify intracellular polyamine contents. Inductively coupled mass spectroscopy was used to quantify intracellular platinum uptake. Cells were exposed to DFMO or DENSPM during 48 h at concentrations ranging from 0 to 5 mM or 0 to 10 muM, respectively. Thereafter, non-treated and treated cells were exposed to cisplatin or BBR3464 during 1 h at concentrations ranging from 0 to 100 muM. A 96-well assay was used to determine cytotoxicity after five days after treatment. RESULTS: The cytotoxic effect of BBR3464 on LU-HNSCC-4 cells was increased after cells were pre-treated with DENSPM or DFMO, and the interaction was found to be synergistic. In contrast, the interaction between cisplatin and DFMO or DENSPM was near-additive to antagonistic. The intracellular levels of the polyamines putrescine and spermidine were decreased after treatment with DFMO, and treatment with DENSPM resulted in an increase in putrescine level and concomitant decrease in spermidine and spermine levels. The uptake of BBR3464 was significantly increased after pre-treatment of the cells with DFMO, and varied dependent on the concentration of DENSPM. The uptake of cisplatin was unchanged. Conclusions: Taken together, these results demonstrate that combinations of polyamine synthesis inhibitors with BBR3464 appear to be a promising approach to enhance the anticancer activity against HSCC.  相似文献

SYNOPSIS. The coccidian parasite Eimeria gadi was found in the haddock, Melanogrammus aeglefinus , taken from the Nova Scotian fishing banks. The haddock infection rates ranged from a high of 58% on Emerald Bank to a low of 4% on Georges Bank, the average being 32%. There was no relationship between sex and degree or prevalence of infection. Although the probability of an occurrence of infection increased with size, small fish with heavy infections were observed. The degree of infection had no apparent effect on the condition factor (length/weight) of the fish. The infection rate reached a maximum in the fall of the year while the heaviest infections were observed in the spring. It is evident from the data that the infection is fatal.
The parasite mass, appearing as a creamy viscous to a yellow semisolid material in the swimbladder, consisted of various parasite stages, fibrous and cellular debris, and lipid material. Some aspects of the sporocyst stage are described.
No other gadoids from the Nova Scotian banks were found to be infected; however, a single specimen of the fourbeard rockling, Enchelyopus cimbrius , from St. John's, Newfoundland, was found to be heavily infected with E. gadi.  相似文献

Field experiments showed that the treatment of winter barley with microbial metabolites produced by a Bacillus subtilis strain led to increased yields in spite of a remaining mildew infection. Disease severity/yield relations obtained on a single tiller basis for either mildew infection at EC 75 or area under disease progress curve (AUDPC) were negatively correlated for untreated plants (R2= 89%, 94%) while this relation did not exist for inducer-treated ones (R2= 10%, 13%). Despite an increasing infection density. yields of main tillers of inducertreated plants were not decreased. On the other hand area under green leaf area curve (AUGLAC) showed a higher correlation with grain yield (R2= 89%) of inducer-treated plants. Possible explanations for the mitigated damaging effect of powdery mildew were expected in carbohydrate metabolism. especially carbohydrate formation and translocation. Assimilation rates of flag leaves of inducer-treated barley with similar infection densities to those of untreated plants were increased over a prolonged period and even exceeded those of non infected ones. In inducer-treated plants the export of 14CO2 from flag leaves into ears remained unimpaired by mildew infection and the allocation of assimilates to grains was highest at late stages of grain filling. Obviously plants were stimulated by inducer treatments to compensate for the damaging effect of powdery mildew and to tolerate mildew infection without yield loss.  相似文献

The effect of the presence of Erysiphe graminis tritici on the reactions of each of five wheats, Malakoff, Democrat, Mediterranean, Hussar and Webster, to Puccinia triticina was studied. Under normal environmental conditions, mildew increased the susceptibility to rust of Malakoff and Democrat, though not, except in isolated instances, that of Mediterranean or Hussar. High light intensities enhanced the effect of mildew on the reaction of Democrat to P. triticina. The susceptibility of Webster to rust was unaffected or decreased by the presence of mildew.
In all varieties there was a decided tendency for rust and mildew to develop on different parts of the leaf, and when a leaf was heavily mildewed the development of P. triticina was sometimes almost entirely inhibited. The reason for this is obscure, since spores of P. triticina and E. g. tritici germinate well in each others presence, and cytological studies revealed several cases of the presence of both mildew and rust haustoria in a single living host cell. Anatomical investigations also showed that the effect of mildew on rust infection was confined to rust pustules within 1 mm. of mildew colonies.
The results obtained are in accordance with Gassner's theory that susceptibility to rust is due to the presence of certain specific proteins in the host leaf.  相似文献

Local infections of either TMV or TNV in tobacco plants cv. Havana 425 (hypersensitive to TMV) proved effective in inducing systemic resistance to subsequent inoculation with the powdery mildew fungus Erysiphe cichoracearum DC. The proportion of leaf surface invaded by this pathogen and the amount of conidia it produced were both significantly lower in virus inoculated plants than in non-inoculated controls. However, the decrease in sporulation rate was less regularly observed than the reduction in leaf area infected. TMV was more effective than TNV in protecting tobacco plants from powdery mildew. E. cichoracearum is thus added to the list of challenge pathogens to which TMV or TNV are known to induce resistance in the host plants. Necrotic lesions caused to the leaves by local treatment with Ethephon (an ethylene-releasing compound) also conferred to tobacco some degree of systemic resistance to the same fungal pathogen, more frequently visible as a reduction of leaf area invaded. The protection due to the Ethephon lesions was in present experiments less marked than that of TMV. No effects against subsequent powdery mildew infection were obtained when point freeze necrotic lesions were provoked on the plants.  相似文献
