Transgenic modification of plants is a key enabling technology for developing sustainable biofeedstocks for biofuels production. Regulatory decisions and the wider acceptance and development of transgenic biofeedstock crops are considered from the context of science-based risk assessment. The risk assessment paradigm for transgenic biofeedstock crops is fundamentally no different from that of current generation transgenic crops, except that the focus of the assessment must consider the unique attributes of a given biofeedstock crop and its environmental release. For currently envisioned biofeedstock crops, particular emphasis in risk assessment will be given to characterization of altered metabolic profiles and their implications relative to non-target environmental effects and food safety; weediness and invasiveness when plants are modified for abiotic stress tolerance or are domesticated; and aggregate risk when plants are platforms for multi-product production. Robust risk assessments for transgenic biofeedstock crops are case-specific, initiated through problem formulation, and use tiered approaches for risk characterization.  相似文献

Horizontal gene transfer (HGT) is part of prokaryotic life style and a major factor in evolution. In principle, any combinations of genetic information can be explored via HGT for effects on prokaryotic fitness. HGT mechanisms including transformation, conjugation, transduction, and variations of these plus the role of mobile genetic elements are summarized with emphasis on their potential to translocate foreign DNA. Complementarily, we discuss how foreign DNA can be integrated in recipient cells through homologous recombination (HR), illegitimate recombination (IR), and combinations of both, site-specific recombination, and the reconstitution of plasmids. Integration of foreign DNA by IR is very low, and combinations of IR with HR provide intermediate levels compared to the high frequency of homologous integration. A survey of studies on potential HGT from various transgenic plants indicates very rare transfer of foreign DNA. At the same time, in prokaryotic habitats, genes introduced into transgenic plants are abundant, and natural HGT frequencies are relatively high providing a greater chance for direct transfer instead of via transgenic plants. It is concluded that potential HGT from transgenic plants to prokaryotes is not expected to influence prokaryotic evolution and to have negative effects on human or animal health and the environment.  相似文献

Monitoring and modeling horizontal gene transfer   总被引:1,自引:0,他引:1  
Monitoring efforts have failed to identify horizontal gene transfer (HGT) events occurring from transgenic plants into bacterial communities in soil or intestinal environments. The lack of such observations is frequently cited in biosafety literature and by regulatory risk assessment. Our analysis of the sensitivity of current monitoring efforts shows that studies to date have examined potential HGT events occurring in less than 2 g of sample material, when combined. Moreover, a population genetic model predicts that rare bacterial transformants acquiring transgenes require years of growth to out-compete wild-type bacteria. Time of sampling is there-fore crucial to the useful implementation of monitoring. A population genetic approach is advocated for elucidating the necessary sample sizes and times of sampling for monitoring HGT into large bacterial populations. Major changes in current monitoring approaches are needed, including explicit consideration of the population size of exposed bacteria, the bacterial generation time, the strength of selection acting on the transgene-carrying bacteria, and the sample size necessary to verify or falsify the HGT hypotheses tested.  相似文献

Environmental risk assessment of transgenic crops is implemented under the Cartagena Protocol domestic law in accordance with guidelines for implementing the assessment established by the Ministry of Agriculture, Forestry and Fisheries (MAFF) and the Ministry of Environment (MOE) in Japan. Environmental risk assessments of transgenic crops are implemented based on the concept of ‘substantial equivalence’ to conventional crops. A unique requirement in Japan to monitor the production of harmful substances, or allelochemicals, is unparalleled in other countries. The potential for allelochemicals to be secreted from the roots of transgenic crops to affect other plants or soil microflora or for substances in the plant body to affect other plants after dying out must be evaluated. We evaluated the allelopathic potential of seven transgenic oilseed rape (Brassica napus L.) lines that express glufosinate tolerance in terms of substantial equivalence to conventional oilseed rape lines, and established evaluation methods. Our results indicate no potential production of allelochemicals for any of the seven transgenic oilseed rape lines compared with conventional oilseed rape lines.  相似文献

Transformation techniques are making it possible to produce novel and unusual plant phenotypes. When considering the environmental impact of these, it is important to do so in the context of what is known about conventional plant breeding and the thousands of varieties that have been produced during this century and earlier. There has now been over ten years of experience of environmental impact assessment with transgenic plants, and research has enabled that assessment process to be better informed scientifically. There are, however, important challenges for the future. Fundamental changes in plant biology, including enhanced tolerance to stressful environments, may create a class of plants that are different from those that have been produced so far, and there may be lessons to be learnt from the experience worldwide of the release of exotic species into different countries. Scale-dependent effects of transgenic plants in agriculture can only effectively be measured by large scale production and monitoring. The monitoring process presents a number of challenges to provide oversight that is meaningful and helpful in assessing environmental impact. The international transboundary movement of transgenic plants is already a reality, and it is important that our environmental impact assessments take this possibility into account. This includes both intentional transboundary movement, through trade of commodity crops, but also unintentional transboundary movement, including the possibility of seeds being moved by animals, by transportation and by humans across the world. There are some major challenges in devising agricultural strategies for the transgenic crops that will become available in the future. The responsibility for developing agricultural strategy rests at a number of levels. To achieve this, it will be necessary to have effective dialogue between the regulatory authorities, the plant breeding and agrochemical industries, and the farming industry. There are already encouraging moves in this direction and hopefully this will continue.  相似文献

Detection and monitoring of insect resistance to transgenic Bt crops   总被引:5,自引:0,他引:5  
Transgenic crops expressing Bacillus thuringiensis (Bt) endotoxins havebecome one of the most important tools for managing corn and cotton insect pests in the US and other countries. The widespread adoption of transgenic Bt crops could place a high degree of selection pressure on the target insect populations and accelerate development of resistance, raising concerns about the long-term durability of Bt plants as an effective pest management tool. Conservation of Bt susceptibility in insects has become one of the most active research areas in modern agriculture. One of the key factors for a successful Bt resistance management plan is to have a cost-effective monitoring system that can provide information on. (i) the initial Bt resistance allele frequencies at low levels in field insect populations; and (ii) early shifts in Bt resistance allele frequencies so that proactive measures for managing resistance can be deployed well before field control failures. Developing such a monitoring program has been difficult because: (i) resistance traits that occur at very low frequencies are hard to detect; (ii) many factors affect the sensitivity and accuracy of a Bt resistance monitoring program; and (iii) monitoring resistance is costly. Several novel methods for detecting Bt resistance alleles developed during the last decade have made a cost-effective monitoring system possible. Future studies should focus on how to improve and standardize the methodologies for insect sampling and Bt resistance detection.  相似文献

近20年来,转基因植物的商业化应用规模越来越大,而转基因生物安全问题依然是转基因植物产业进一步发展的最主要制约因素。转基因植物在商业化应用之前虽然预先进行了风险评估,但是,包括环境监测在内的风险管理措施是确保转基因植物安全应用的必要手段。在转基因作物大规模应用近20年之后,其在靶标生物抗性、对生物多样性的影响、基因漂移、在生态系统中的长期存留等方面产生的环境风险已经渐渐显现出来,表明风险评估无法为转基因植物应用提供足够的安全保障,还必须通过开展系统而长期的环境监测,明确转基因植物在生产应用后的实际环境影响。联合国环境规划署和欧盟等已经制定了转基因植物环境监测的法规和技术指南,一些国家实施了系统的转基因植物环境监测。对转基因植物所产生的环境风险以及环境监测应包括的内容进行了综述。  相似文献

Biotechnologies have been utilized "ante litteram" for thousands of years to produce food and drink and genetic engineering techniques have been widely applied to produce many compounds for human use, from insulin to other medicines. The debate on genetically modified (GM) organisms broke out all over the world only when GM crops were released into the field. Plant ecologists, microbiologists and population geneticists carried out experiments aimed at evaluating the environmental impact of GM crops. The most significant findings concern: the spread of transgenes through GM pollen diffusion and its environmental impact after hybridisation with closely related wild species or subspecies; horizontal gene transfer from transgenic plants to soil microbes; the impact of insecticide proteins released into the soil by transformed plants on non-target microbial soil communities. Recent developments in genetic engineering produced a technology, dubbed "Terminator", which protects patented genes introduced in transgenic plants by killing the seeds in the second generation. This genetic construct, which interferes so heavily with fundamental life processes, is considered dangerous and should be ex-ante evaluated taking into account the data on "unexpected events", as here discussed, instead of relying on the "safe until proven otherwise" claim. Awareness that scientists, biotechnologists and genetic engineers cannot answer the fundamental question "how likely is that transgenes will be transferred from cultivated plants into the natural environment?" should foster long-term studies on the ecological risks and benefits of transgenic crops.  相似文献

Advances in recombinant DNA technology have created advantages for the development of plants with high agro-economical values. Since the production of transgenic plants, some issues concerning the safe use of these plants and their products have been under debate throughout the world. In this respect, the potential risks and benefits of transgenic plants need to be evaluated objectively. Risk assessment of transgenic crops is a basic prerequisite for monitoring the possible risks that could arise upon the release and use of transgenic plants. To get a meaningful tool for decision making, risk assessment needs to be carried out in a scientific sound and transparent manner. There are specific governmental regulations in many countries for the safety assessment of genetically modified (GM) crops. Furthermore, there are some international agreements, which regulate the cultivation and commercialization of transgenic plants and their derivatives. Internationally accepted risk assessment strategies have been performed to evaluate the safe use of a large variety of GM crops. The main objectives of these regulations and risk assessment strategies are focused to protect human/animal health and the environment.  相似文献

The testing of transgenic crops in the field is a necessary part of the validation of genetically engineered cultivars, but in the early stages of testing, biosafety procedures must be carefully monitored to ensure that the modified plants do not have deleterious effects on the environment. This study was carried out over two seasons to determine the effectiveness of containment procedures under australian environmental conditions by measuring the dispersal of pollen amay from a test plot of transgenic cotton into a surrounding buffer field of non-transgenic cotton plants whose function was to act as a sink for pollen carried by nectar feeding and pollen-gathering insects. Dispersal was estimated by measuring the frequency of the dominant selectable marker transgene, neomycin phosphotransferase (NptII) in the progeny of the buffer plants. The presence of nptii was determined by a sensitive radioactive enzyme assay. Pollen dispersal was low in both years, but increased with an increase in the size of the source plot in the second year. In the first year outcrossing averaged from 0.15% of progeny at 1 m to below 0.08% at 4 m from the test plot. Outcrossing was highest within the central test plot where progeny from non-transgenic control plants, immediately adjacent to transgenic plants, had transgenic progeny at frequencies of up to 1.7%. In the second year, with a bigger source of transgenic plants, outcrossing declined on average from 0.4% at 1 m to below 0.03% at 16 m into the buffer zone. These results indicate that 20 m buffer zones would serve to limit dispersal of transgenic pollen from small-scale field tests.  相似文献

转基因树木与农作物相比, 人们更关注其长时间种植可能导致转基因扩散到周围野生近缘种。由于生长周期长, 转基因树木会增加转基因不稳定性, 对非靶标生物的影响, 靶标害虫对转基因植物产生抗性, 增加树木入侵性(杂草化), 以及由于基因漂移或基因逃逸对环境产生的负面影响或新的环境风险。过去十几年, 针对我国抗食叶害虫的两个商业化转Bt基因欧洲黑杨(Populus nigra)和转双抗虫基因741杨[P. alba× (P. davidiana + P. simonii) × P. tomentosa], 已开展了有关生态安全性方面的多项研究。本文围绕抗虫转基因树木生态安全性研究进展进行了综述。抗虫转基因杨树对节肢动物种群和群落结构产生了一定影响, 使昆虫的多样性提高, 但对土壤微生物区系未见明显影响。转基因欧洲黑杨通过花粉和种子发生的基因漂移几率很低。转基因杨树通过内生菌发生的水平转移可能会对环境造成的潜在危险也进行了评价。文章最后指出对抗虫转基因杨树农林复合生态系统开展生物安全研究的必要性。  相似文献

Transgenic plants that are being developed for commercial cultivation must be tested under field conditions to monitor their effects on surrounding wildlife and conventional crops. Developers also use this opportunity to evaluate the performance of transgenic crops in a typical environment, although this is a matter of commercial necessity rather than regulatory compliance. Most countries have adapted existing regulations or developed new ones to deal specifically with transgenic crops and their commodities. The European Union (EU) is renowned, or perhaps notorious, for having the broadest and most stringent regulations governing such field trials in the world. This reflects its nominal adherence to the precautionary approach, which assumes all transgenic crops carry an inherent risk. Therefore, field trials in the EU need to demonstrate that the risk associated with deploying a transgenic crop has been reduced to the level where it is regarded as acceptable within the narrowly defined limits of the regulations developed and enforced (albeit inconsistently) by national and regional governments, that is, that there is no greater risk than growing an equivalent conventional crop. The involvement of national and regional competent authorities in the decision-making process can add multiple layers of bureaucracy to an already-intricate process. In this review, we use country-based case studies to show how the EU, national and regional regulations are implemented, and we propose strategies that could increase the efficiency of regulation without burdening developers with further unnecessary bureaucracy.  相似文献

We present a field-based approach to detect and monitor insects with resistance to insecticidal toxins produced by transgenic plants. Our objective is to estimate the phenotypic frequency of resistance in a population by relating the densities of insects on genetically transformed plants to densities on nontransformed plants. We focus on European corn borer, Ostrinia nubilalis (Hübner), in sweet corn, Zea mays L., expressing Cry1Ab from Bacillus thuringiensis subsp. kurstaki Berliner to illustrate principles underlying the method. The probability of detecting one or more rare, resistant larvae depends on sample size, the density of larvae on nontransformed plants, and an assumed frequency of resistant phenotypes in a given population. Probability of detection increases with increases in sample size, background density, or the frequency of resistant individuals. Following binomial probability theory, if a frequency of 10(-4) is expected, 10(3)-10(4) samples must be collected from a B. thuringiensis (Bt) crop to have at least a 95% probability of locating one or more resistant larvae. In-field screens using transgenic crops have several advantages over traditional laboratory-based methods, including exposure to a large number of feral insects, discrimination of resistant individuals based on Bt dosages expressed in the field, incorporation of natural and Bt-induced mortality factors, simultaneous monitoring for more than one insect species, and ease of use. The approach is amenable to field survey crews working in research, extension, and within the seed corn industry. Estimates of the phenotypic frequency of resistance from the in-field screen can be useful for estimating initial frequency of resistant alleles. Bayesian statistical methods are outlined to estimate phenotype frequencies, allele frequencies, and associated confidence intervals from field data. Results of the approach are discussed relative to existing complementary methods currently available for O. nubilalis and corn earworm, Helicoverpa zea (Boddie).  相似文献

By the end of the 1980s, a broad consensus had developed that there were potential environmental risks of transgenic plants requiring assessment and that this assessment must be done on a case-by-case basis, taking into account the transgene, recipient organism, intended environment of release, and the frequency and scale of the intended introduction. Since 1990, there have been gradual but substantial changes in the environmental risk assessment process. In this review, we focus on changes in the assessment of risks associated with non-target species and biodiversity, gene flow, and the evolution of resistance. Non-target risk assessment now focuses on risks of transgenic plants to the intended local environment of release. Measurements of gene flow indicate that it occurs at higher rates than believed in the early 1990s, mathematical theory is beginning to clarify expectations of risks associated with gene flow, and management methods are being developed to reduce gene flow and possibly mitigate its effects. Insect pest resistance risks are now managed using a high-dose/refuge or a refuge-only strategy, and the present research focuses on monitoring for resistance and encouraging compliance to requirements. We synthesize previous models for tiering risk assessment and propose a general model for tiering. Future transgenic crops are likely to pose greater challenges for risk assessment, and meeting these challenges will be crucial in developing a scientifically coherent risk assessment framework. Scientific understanding of the factors affecting environmental risk is still nascent, and environmental scientists need to help improve environmental risk assessment.  相似文献

The persistence and movement of strain JS414 of Xanthomonas campestris pv. campestris, which was genetically engineered to bioluminesce, were monitored during a limited field introduction. Bioluminescence and traditional dilution plate counts were determined. Strain JS414 was applied to cabbage plants and surrounding soil by mist inoculation, by wound inoculation, by scattering infested debris among plants, and by incorporating bacteria into the soil. Bioluminescent X. campestris pv. campestris was detected in plant samples and in the rhizosphere up to 6 weeks after inoculation. Movement to uninoculated plants was detected on one occasion, but movement from the immediate release area was not detected. Strain JS414 was detected in soil samples beneath mist- and wound-inoculated plants only at intentionally infested locations and in aerial samples only on the day of inoculation. Our bioluminescence methods proved to be as sensitive as plating methods for detecting the genetically engineered microorganisms in environmental samples. Our results demonstrate that transgenic incorporation of the luxCDABE operon provides a non-labor-intensive, sensitive detection method for monitoring genetically engineered microorganisms in nature.  相似文献

The transgenic traits associated with the majority of commercial genetically modified crops are focused on improving herbicide and insecticide management practices. The use of the transgenic technology in these crops and the associated chemistry has been the basis of studies that provide evidence for occasional improvement in environmental benefits due to the use of less residual herbicides, more targeted pesticides, and reduced field traffic. This is nicely exemplified through studies using Environmental Impact Quotient (EIQ) assessments. Whilst EIQ evaluations may sometimes illustrate environmental benefits they have their limitations. EIQ evaluations are not a surrogate for Environmental Risk Assessments and may not reflect real environmental interactions between crops and the environment. Addressing the impact cultivated plants have on the environment generally attracts little public attention and research funding, but the introduction of GM has facilitated an expansion of research to address potential environmental concerns from government, NGOs, industry, consumers, and growers. In this commentary, some evidence from our own research and several key papers that highlight EIQ assessments of the impact crops are having on the environment are presented. This information may be useful as an education tool on the potential benefits of GM and conventional farming. In addition, other deliberate, accidental, and GM-driven benefits derived from the examination of GM cropping systems is briefly discussed.  相似文献

Transgenic insect-resistant cotton has been released into the environment for more than a decade in China to effectively control the cotton bollworm (Helicoverpa armigera) and other Lepidoptera. Because of concerns about undesirable ecological side-effects of transgenic crops, it is important to monitor the potential environmental impact of transgenic insect-resistant cotton after commercial release. Our 2-year study included 1 cotton field where non-transgenic cotton had been planted continuously and 2 other cotton fields where transgenic insect-resistant cotton had been planted for different lengths of time since 1997 and since 2002. In 2 consecutive years (2009 and 2010), we took soil samples from 3 cotton fields at 4 different growth stages (seedling, budding, boll-forming and boll-opening stages), collected soil nematodes from soil with the sugar flotation and centrifugation method and identified the soil nematodes to the genus level. The generic composition, individual densities and diversity indices of the soil nematodes did not differ significantly between the 2 transgenic cotton fields and the non-transgenic cotton field, but significant seasonal variation was found in the individual densities of the principal trophic groups and in the diversity indices of the nematodes in all 3 cotton fields. The study used a comparative perspective to monitor the impact of transgenic insect-resistant cotton grown in typical ‘real world’ conditions. The results of the study suggested that more than 10 years of cultivation of transgenic insect-resistant cotton had no significant effects–adverse or otherwise–on soil nematodes. This study provides a theoretical basis for ongoing environmental impact monitoring of transgenic plants.  相似文献

【背景】转基因作物可能通过根系分泌物和植株残体组成的改变对土壤酶活性和养分转化产生影响,转基因作物对土壤质量的影响是其环境安全性评价的重要方面。【方法】本研究通过田间定位试验分析了连续3a种植2种转crylAc/cpti双价抗虫基因水稻后,土壤酶活性和养分有效性等土壤质量性状的变化。【结果】在水稻各生育期内,除齐穗期转基因稻科丰8号(GM1)田的土壤酸性磷酸酶活性显著(P〈0.05)高于其受体非转基因稻明恢86(CK1)外,转基因稻GM1、GM2(Ⅱ优科丰8号)的土壤酸性磷酸酶、碱性磷酸酶和脲酶活性与对应非转基因稻CK1、CK2(Ⅱ优明恢86)间均无显著差异。同时,在水稻生长过程中,土壤pH、有机质、有效氮、有效磷和速效钾等土壤理化指标在GM1和CK1或GM2和CK2间也均无显著差异。【结论与意义】连续38种植转crylAc/cpti双价抗虫基因水稻并未改变稻田土壤中主要营养元素的有效性及其相关土壤酶活性,即短期内种植转crylAc/cpti双价抗虫基因水稻不会影响土壤酶活性和养分状况。该结果为进一步评价转基因水稻的生态风险提供了一定的依据。  相似文献

Combinations of dissimilar insecticidal proteins (“pyramids”) within transgenic plants are predicted to delay the evolution of pest resistance for significantly longer than crops expressing a single transgene. Field-evolved resistance to Bacillus thuringiensis (Bt) transgenic crops has been reported for first generation, single-toxin varieties and the Cry1 class of proteins. Our five year data set shows a significant exponential increase in the frequency of alleles conferring Cry2Ab resistance in Australian field populations of Helicoverpa punctigera since the adoption of a second generation, two-toxin Bt cotton expressing this insecticidal protein. Furthermore, the frequency of cry2Ab resistance alleles in populations from cropping areas is 8-fold higher than that found for populations from non-cropping regions. This report of field evolved resistance to a protein in a dual-toxin Bt-crop has precisely fulfilled the intended function of monitoring for resistance; namely, to provide an early warning of increases in frequencies that may lead to potential failures of the transgenic technology. Furthermore, it demonstrates that pyramids are not ‘bullet proof’ and that rapid evolution to Bt toxins in the Cry2 class is possible.  相似文献

全球范围内转基因农作物的大量种植不仅带来巨大的经济利益,同时也引发了人们关于转基因作物对包含土壤微生物在内的土壤生态系统的潜在风险的忧虑.转基因作物对土壤微生物的影响包括外源基因表达蛋白对非靶标土壤微生物的直接影响,也包括因外源基因导入而植物根系分泌物组分变化引起的间接影响.目前,对转基因作物的大多数研究表明,转基因作物能引起土壤微生物种群数量和结构的变化.但是,转基因作物对土壤微生物的影响力度有大有小,持续时间有长有短,评价不一.本文综述了不同种类转基因作物对土壤微生物的影响,对转基因作物种类、试验技术和原则等影响评价结果准确性的因素进行了讨论,提出了进一步研究需要注意的问题.  相似文献
