Abstract A new isocratic ion chromatographic technique is described for the sensitive measurement of tetrathionate, trithionate and thiosulfate. The sulfur oxyanions were separated on a polymer-coated, silica-based anion exchange column and directly detected by UV absorption at 216 nm. Aqueous saline acetonitrile/methanol mixtures were used as eluent. The three anions could be quantified in less than 10 min with detection limits of about 0.6 pmol for tetrathionate and of 40 and 10 pmol for trithionate and thiosulfate, respectively. The retention times of tetrathionate responded to changes of the solvent concentration, whereas the elution of thiosulfate depended predominatly on the ionic strenght of the eluent. Starting at the lowest detectable concentrations, calibration curves for all three compounds were linear over a concentration range of three orders of magnitude. The analysis of freshwater and saline samples worked equally well. Since contact of the eluent with metallic components caused shifts in retention times during operation, the solvent delivery system had to consist of plastic material. Examples of applicaton are given for determination of tetrathionate and thiosulfate in natural samples and for the turnover of these two compounds in sediment slurries and anaerobic enrichment cultures.  相似文献

Rhodopseudomonas globiformis is able to assimilate both sulfur moieties of thiosulfate. During growth on 35S-labelled thiosulfate the amino acids cysteine, homocysteine and methionine were labelled. The bulk of thiosulfate, however, was oxidized to tetrathionate and accumulated in the medium. A thiosulfate: acceptor oxidoreductase was partially purified and characterized. The enzyme oxidized thiosulfate to tetrathionate in the presence of ferricyanide. A c-type cytochrome isolated from this organism was reduced by this enzyme.  相似文献

The oxidation of thiosulfate to tetrathionate exerts a definite growth rate stimulation in glucose, acetate, and yeast extract cultures of some marine pseudomonads. The failure to find this effect in earlier studies with terrestrial isolates may lie in the particular conditions used in the present experiments (constant pH, high ratio of thiosulfate to organic substrate) or in the different metabolic characteristics of the marine isolates.Contribution No. 3220 from the Woods Hole Oceanographic Institution.  相似文献

A periplasmic thiosulfate dehydrogenase (EC 1.8.2.2) was purified to homogeneity from the neutrophilic, obligately chemolithoautotrophicThiobacillus sp. W5. A five-step procedure resulted in an approximately 2,300-fold purification. The purified protein had a molecular mass of 120±3 kDa, as determined by gel filtration. It is probably a tetramer containing two different subunits with molecular masses of 33±1 kDa and 27±0.5 kDa, as determined by SDS-PAGE. UV/visible spectroscopy revealed that the enzyme contained haemc; haem staining showed that both subunits contained haemc. A haemc content of 4 mol per mol of enzyme was calculated using the pyridine haemochrome test. The pH optimum of the enzyme was 5.5 At pH 7.5, the Km and Vmax were 120±10 M and 1,160±30 U mg-1, respectively. The absence of 2-heptyl-4-hydroquinoline-N-oxide (HQNO) inhibition for the oxidation of thiosulfate by whole cells suggested that the electrons enter the respiratory chain at the level of cytochromec. The unicellular cyanobacterial strain 16Som2, isolated from a Somaliland saltpan and identified asCyanothece sp., is characterized by cells surrounded by a thick polysaccharidic capsule, the external part of which dissolves into the medium during growth, causing a progressive increase in culture viscosity. In spite of this, the thickness of the capsule remained almost constant under all the culture conditions tested, demonstrating that the processes of its synthesis and solubilization occurred at a similar rate. The synthesis of carbohydrates was neither enhanced by increasing salinity (sea-water enriched with NaCl in the range 0 to 2.0 M) nor by Mg2+, K+ or Ca2+ deficiencies. In contrast, N-limitation and, to a lesser extent, P-limitation induced a significant enhancement of carbohydrate synthesis; in particular, N-deficiency stimulated the synthesis of all the carbohydrate fractions (intracellular, capsular and soluble). The soluble polysaccharide, separated from the culture medium and hydrolyzed with 2N trifluoroacetic acid, showed a sugar composition consisting of glucuronic acid: galacturonic acid: galactose: glucose: mannose: xylose: fucose in a molar ratio of 1: 2: 2.4: 6.8: 4.8: 2.9: 1.6.Cyanothece sp. culture subjected to nitrogen starvation synthesized polysaccharide with a mean productivity of 115 mg (EPS) l–1d–1, for the polymer solubilized into the medium, and of 15 mg (CPS) l–1d–1 for the capsular polysaccharide.  相似文献

We have studied the exopolysaccharides (EPS) from a new group of moderately halophilic bacteria belonging to the genus Halomonas. The quantity of EPS produced, its chemical composition and physical properties depend greatly upon the bacterial strain. The majority of the polymers produced viscous solutions and/or emulsified different hydrocarbon compounds. The most interesting strain, S-30, produced EPS at 2.8 g/l with a maximum viscosity of 23.5 Pa·5 and exhibited pseudoplastic behavior. This EPS emulsified five hydrocarbons more efficiently than did four control surfactants tested. Its monosaccharide composition was glucose:galactose:manose:glucuronic acid in equimolar ratio. Some two-thirds of the strains tested emulsified crude oil better than control surfactants did. There are many potential industrial applications for polysaccharides with these qualities. Two heterotrophic bacteria that oxidized thiosulfate to tetrathionate were isolated from soil. The enzyme system in one of the isolates (C-3) was constitutive, but in the other isolate (A-50) it was induced by thiosulfate or tetrathionate. The apparent K(m) for oxygen for thiosulfate oxidation by A-50 was about 223 mum, but, for lactate oxidation by A-50 or thiosulfate oxidation by C-3, the apparent K(m) for oxygen was below 2 mm. The oxidation of thiosulfate by A-50 was first order with respect to oxygen from 230 mum. The rate of oxidation was greatest at pH 6.3 to 6.8 and at about 10 mm thiosulfate, and it was strongly inhibited by several metal-binding reagents. Extracts of induced A-50 reduced ferricyanide, endogenous cytochrome c, and mammalian cytochrome c in the presence of thiosulfate. A-50, once induced to oxidize thiosulfate, also reduced tetrathionate to thiosulfate in the presence of an electron donor such as lactate. The optimal pH for this reaction was at 8.5 to 9.5, and the reaction was first order with respect to tetrathionate. There was no correlation between the formation of the thiosulfate-oxidizing enzyme of A-50 and the incorporation of thiosulfate-sulfur into cell sulfur. Thiosulfate did not affect the growth rate or yield of A-50.  相似文献

Abstract A thermophilic rod (strain SEBR 5268), isolated from an oil-producing well, was identified as a Thermoanaerobacter strain that was phenotypically related to T. finnii . Both SEBR 5268 and T. finnii oxidized H2 by reducing thiosulfate to sulfide using yeast extract as growth substrate. H2 oxidation in the presence of thiosulfate was significant at the end of the exponential growth of SEBR 5268 and was maintained during the lysis phase. In the absence of thiosulfate, H2 was inhibitory for both strains. The role of H2 consumption by these bacteria is discussed with regard to their metabolism on organic compounds.  相似文献

The “Old Sulphur Well” has a subterranean input of water containing 5.5 mM total sulfide, which would be inhibitory to the growth of most bacteria. The obligately chemolithoautotrophic Halothiobacillus neapolitanus is a sulfur bacterium known to tolerate and metabolize high sulfide concentrations, and we report the isolation of H. neapolitanus strain OSWA from this source. Strain OSWA grows well on thiosulfate and tetrathionate as energy sources, and tolerates at least 5 mM sulfide. Its specific growth rates and yields in batch culture were 0.22 h−1 and 5.3 g mol−1 (thiosulfate), and 0.23 h−1 and 9.5 g mol−1 (tetrathionate). Its 16S rRNA gene sequence shows >99% identity to reference sequences of H. neapolitanus, and it shares morphological and physiological characteristics typical of the species. It is one of a very small number of strains of H. neapolitanus described to date, and the first to be isolated from an ancient sulfide-rich natural spa.  相似文献

A pure culture of alkaliphilic haloarchaeon strain AArc-ST capable of anaerobic growth by carbohydrate-dependent sulfur respiration was obtained from hypersaline lakes in southwestern Siberia. According to phylogenetic analysis, AArc-ST formed a new genus level branch most related to the genus Natronoarchaeum in the order Halobacteriales. The strain is facultatively anaerobic with strictly respiratory metabolism growing either by anaerobic respiration with elemental sulfur and thiosulfate as the electron acceptors or by aerobic respiration at microoxic conditions. Thiosulfate is reduced partially to sulfide and sulfite. It is a first sulfur-reducing alkaliphilic haloarchaeon utilizing sugars, starch and glycerol as substrates for anaerobic growth. It is extremely halophilic (optimum at 3.5 M total Na+) and obligately alkaliphilic (optimum at pH 9.5). The dominant polar lipids include PG and PGP-Me with the archaeol (C20-C20) or extended archaeol (C20-C25) cores. The dominant respiratory lipoquinone is MK-8:8. On the basis of unique physiological properties and results of phylogenetic analysis, the soda lake isolate is suggested to be classified into a novel genus and species Natranaeroarchaeum sulfidigenes gen. nov., sp. nov. (=JCM 34033T = UNIQEM U1000T). Furthermore, on the bases of phylogenomic reconstruction, a new family Natronoarchaeaceae fam. nov. is proposed within the order Halobacteriales incorporating Natranaeroarchaeum and three related genera: Natronoarchaeum, Salinarchaeum and Halostella.  相似文献

Tetrathionate hydrolase (4THase), a key enzyme of the S4-intermediate (S4I) pathway, was partially purified from marine acidophilic bacterium, Acidithiobacillus thiooxidans strain SH, and the gene encoding this enzyme (SH-tth) was identified. SH-Tth is a homodimer with a molecular mass of 97 ± 3 kDa, and contains a subunit 52 kDa in size. Enzyme activity was stimulated in the presence of 1 M NaCl, and showed the maximum at pH 3.0. Although 4THases from A. thiooxidans and the closely related Acidithiobacillus caldus strain have been reported to be periplasmic enzymes, SH-Tth seems to be localized on the outer membrane of the cell, and acts as a peripheral protein. Furthermore, both 4THase activity and SH-Tth proteins were detected in sulfur-grown cells of strain SH. These results suggested that SH-Tth is involved in elemental sulfur-oxidation, which is distinct from sulfur-oxidation in other sulfur-oxidizing strains such as A. thiooxidans and A. caldus.  相似文献

Two extremely halophilic archaea strains, CBA1112T and CBA1113, were isolated from solar salt in Korea. The genome sizes and G + C content of CBA1112T and CBA1113 were 3.77 and 3.53 Mb, and 66.0 and 66.5 mol%, respectively. Phylogenetic analysis based on closely related taxa and environmental Haloplanus sequences indicated that both CBA1112T and CBA1113 strains are grouped within the genus Haloplanus. OrthoANI and in silico DNA–DNA hybridization values were below the species delineation threshold. Pan-genomic analysis showed that the two novel strains and four reference strains had 6203 pan-orthologous groups in total. Six Haloplanus strains shared 1728 core pan-genome orthologous groups, which were mainly associated with amino acid transport and metabolism and translation, ribosomal structure and biogenesis categories, and amino acid metabolism and carbohydrate metabolism related categories. The novel strain-specific pan-genome orthologous groups were mainly involved with replication, recombination and repair category and replication and repair pathway or amino acid metabolism pathway. Cells of both strains were Gram-negative and pleomorphic, and colonies were red-pigmented. The major polar lipids of both strains were phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester, phosphatidylglycerol sulfate, and one glycolipid, sulfated mannosyl glucosyl diether. Based on genomic, phylogenetic, phenotypic, and chemotaxonomic features, strains CBA1112T and CBA1113 are described as novel species of the genus Haloplanus. Thus, we propose the name Haloplanus rubicundus sp. nov. The type strain is CBA1112T (=KCCM 43224T = JCM 30475T).  相似文献

An enzymatic complex from Rhodotorula was characterized and it was indicated that it possessed thiosulfate-oxidizing activity, forming tetrathionate as well as sulfite oxidase activity. Both activities coupled with ferricyanide and native cytochrome c but no with mammalian cytochrome c. Activities of these enzymes were inhibited by thiol inhibitors. Chelating agents did not affect thiosulfate oxidizing activity and only moderately inhibited sulfite oxidase. Both activities disappeared after treatment with proteolytic enzymes or sodium deoxycholate which indicates an essential role played not only by protein but also by phospholipids in the enzymatic activity of the complex. Thiosulfate oxidizing enzyme had a K m for thiosulfate of 0.16 mM with ferricyanide as electron acceptor and of 14 M with native cytochrome c and of 0.34 mM for ferricyanide. Optimum pH for this activity was 7.8. Other properties of this enzyme were similar to those of thiobacilli and heterotrophic bacteria. The activity of sulfite oxidase was inhibited by 50% with 10 M AMP. The K m values of this enzyme were 1 mM with ferricyanide as electron acceptor and 60 M with native cytochrome c for sulfite and 0.42 mM for ferricyanide. The enzyme did not show a specific optimum pH value with ferricyanide as electron acceptor. However, with native cytochrome c optimum pH was 7.8 for its activity. Sulfide oxidation and product formation was studied in the cyanobacterium Microcoleus chtonoplastes. Anoxygenic photosynthesis was induced in the presence of sulfide and light. It was demonstrated that thiosulfate was the only product of the 3(3,4-dichlorophenyl)1,1-dimethylurea (DCMU)-insensitive photo-oxidation of sulfide. The affinity of this system for sulfide was shown to be very low.Oxygenic photosynthesis continued in the presence of sulfide. After an induction period of 3 h, oxygenic and anoxygenic photosynthesis were shown to be operating simultaneously.The ecological importance of sulfide oxidation and thiosulfate production by M. chtonoplastes is discussed in the context of laminated microbial ecosystems, where cyanobacteria and purple sulfur bacteria coexist.  相似文献

Recently, heterotrophic nanoflagellates (HNF) have been reported to actively ingest prokaryotes in high salinity waters. We report the isolation and culture of an HNF from a Korean saltern pond of 300‰ salinity. The organism is biflagellated with an acronematic anterior flagellum and never glides on surfaces. The mitochondria have tubular cristae. Neither transitional helix nor spiral fiber were observed in the transition zones of the flagella. The cell has a cytostome supported by an arc of eight microtubules, suggesting that our isolate is a bicosoecid. Our isolate had neither mastigonemes, lorica, body scales, nor cytopharynx and thus could not be placed in any of the presently described bicosoecid genera. Phylogenetic analysis of 18S rRNA gene sequences from stramenopiles confirmed the bicosoecid affinities of our isolate, but did not place it within any established genus or family. Its closest relatives include Caecitellus and Cafeteria. The optimal range of growth temperature was 30–35°C. The isolated HNF grew optimally at 150‰ salinity and tolerated up to 363‰ salinity, but it failed to grow below 75‰ salinity, indicating that it could be a borderline extreme halophile. On the basis of its morphological features and position in 18S rRNA trees we propose a novel genus for our isolate; Halocafeteria, n. gen. The species name Halocafeteria seosinensis sp. nov. is proposed.  相似文献

Aims:  Optimization of medium components for extracellular protease production by Halobacterium sp. SP1(1) using statistical approach.
Methods and Results:  The significant factors influencing the protease production as screened by Plackett–Burman method were identified as soybean flour and FeCl3. Response surface methodology such as central composite design was applied for further optimization studies. The concentrations of medium components for higher protease production as optimized using this approach were (g l−1): NaCl, 250; KCl, 2; MgSO4, 10; tri-Na-citrate, 1·5; soybean flour, 10 and FeCl3, 0·16. This statistical optimization approach led to production of 69·44 ± 0·811 U ml−1 of protease.
Conclusions:  Soybean flour and FeCl3 were identified as important factors controlling the production of extracellular protease by Halobacterium sp. SP1(1). The statistical approach was found to be very effective in optimizing the medium components in manageable number of experimental runs with overall 3·9-fold increase in extracellular protease production.
Significance and Impact of the Study:  The present study is the first report on statistical optimization of medium components for production of haloarchaeal protease. The study also explored the possibility of using extracellular protease produced by Halobacterium sp. SP1(1) for various applications like antifouling coatings and fish sauce preparation using cheaper raw material.  相似文献

Thiosulfate-reductase activity (TSR) measured as sulfide release from thiosulfate was detected in crude extracts of Chlorella using dithioerythritol (DTE) as electron donor. Purification of this activity by ammonium-sulfate precipitation between 35% and 80% followed by Sephadex G-50 gel filtration, diethylaminoethyl-cellulose chromatography, and gel filtration on Biogel A 1.5 M led to four distinct proteins having molecular weights of: TSR I, 28000; TSR II, 26500; TSR IIIa, 55000; TSR IIIb, 24000 daltons. These thiosulfate reductases were most active with DTE; the monothiols glutathione, l-cysteine, and -mercaptoethanol had little activity towards this system. The following pH optima were obtained: for TSR I and TSR II, 9.0; for TSR IIIa, 8.5; and for TSR IIIb, 9.5. The apparent-Km data for DTE and thiosulfate were determined to: TSR I, 0.164 mmol·l-1 and TSR II, 0.156 mmol·l-1; KmDTE TSR I, 1.54 mmol·l-1 and TSR II 1.54 mmol·l-1. The thiosulfate reductases IIIa and IIIb were further stimulated by addition of thioredoxin.
