Aegilops speltoides is an important genetic resource for wheat improvement and has high levels of heat tolerance. A heat-tolerant accession of Ae. speltoides pau3809 was crossed with Triticum durum cv. PDW274, and BC2F4-6 backcross introgression lines (BILs) were developed, phenotyped for important physiological traits, genotyped using SSR markers and used for mapping the QTL governing heat tolerance component traits. A set of 90 BILs was selected from preliminary evaluation of a broader set of 262 BILs under heat stress. Phenotyping was conducted for physiological traits such as cell membrane thermostability, chlorophyll content, acquired thermotolerance, canopy temperature and stay green. Much variation for these traits was observed in random as well as selected sets of BILs, and comparison of the BILs with the recurrent parent showed improvement for these traits under normal as well as heat stress conditions, indicating that introgressions from Ae. speltoides might have led to the improvement in the heat tolerance potential of the BILs. Introgression profiling of the 90 BILs using SSR markers identified Ae. speltoides introgression on all the 14 chromosomes with introgressions observed on A as well as B genome chromosomes. QTL mapping identified loci for various heat tolerance component traits on chromosomes 2B, 3A, 3B, 5A, 5B and 7A at significant LOD scores and with phenotypic contributions varying from 11.1 to 28.7 % for different traits. The heat-tolerant BILs and QTL reported in the present study form a potential resource that can be used for wheat germplasm enhancement for heat stress tolerance.  相似文献

Carbon balancing within the plant species is an important feature for climatic adaptability. Photosynthesis and respiration traits are directly linked with carbon balance. These features were studied in 20 wild rice accessions Oryza spp., and cultivars. Wide variation was observed within the wild rice accessions for photosynthetic oxygen evolution or photosynthetic rate (A), dark (R d), and light induced respiration (LIR) rates, as well as stomatal density and number. The mean rate of A varied from 10.49 μmol O2 m?2 s?1 in cultivated species and 13.09 μmol O2 m?2 s?1 in wild spp., The mean R d is 2.09 μmol O2 m?2 s?1 and 2.31 μmol O2 m?2 s?1 in cultivated and wild spp., respectively. Light induced Respiration (LIR) was found to be almost twice in wild rice spp., (16.75 μmol O2 m?2 s?1) compared to cultivated Oryza spp., Among the various parameters, this study reveals LIR and A as the key factors for positive carbon balance. Stomatal contribution towards carbon balance appears to be more dependent on abaxial surface where several number of stomata are situated. Correlation analysis indicates that R d and LIR increase with the increase in A. In this study, O. nivara (CR 100100, CR 100097), O. rufipogon (IR 103404) and O. glumaepatula (IR104387) were identified as potential donors which could be used in rice breeding program. Co-ordination between gas exchange and patchiness in stomatal behaviour appears to be important for carbon balance and environmental adaptation of wild rice accessions, therefore, survival under harsh environment.  相似文献

In this study, we have compared the photosynthetic characteristics of two contrasting species of Tradescantia plants, T. fluminensis (shade-tolerant species), and T. sillamontana (light-resistant species), grown under the low light (LL, 50–125 µmol photons m?2 s?1) or high light (HL, 875–1000 µmol photons m?2 s?1) conditions during their entire growth period. For monitoring the functional state of photosynthetic apparatus (PSA), we measured chlorophyll (Chl) a emission fluorescence spectra and kinetics of light-induced changes in the heights of fluorescence peaks at 685 and 740 nm (F 685 and F 740). We also compared the light-induced oxidation of P700 and assayed the composition of carotenoids in Tradescantia leaves grown under the LL and HL conditions. The analyses of slow induction of Chl a fluorescence (SIF) uncovered different traits in the LL- and HL-grown plants of ecologically contrasting Tradescantia species, which may have potential ecophysiological significance with respect to their tolerance to HL stress. The fluorometry and EPR studies of induction events in chloroplasts in situ demonstrated that acclimation of both Tradescantia species to HL conditions promoted faster responses of their PSA as compared to LL-grown plants. Acclimation of both species to HL also caused marked changes in the leaf anatomy and carotenoid composition (an increase in Violaxanthin?+?Antheraxantin?+?Zeaxanthin and Lutein pools), suggesting enhanced photoprotective capacity of the carotenoids in the plants grown in nature under high irradiance. Collectively, the results of the present work suggest that the mechanisms of long-term PSA photoprotection in Tradescantia are based predominantly on the light-induced remodeling of pigment-protein complexes in chloroplasts.  相似文献

Abelmoschus manihot, an ornamental plant, was examined for phytoremediation purposes in accordance with the ability to accumulate cadmium and physiological mechanisms of cadmium tolerance. A net photosynthetic rate (A N) glasshouse experiment for 60 days was conducted to investigate the influence of different cadmium amounts (0–100 mg kg?1) on the growth, biomass, photosynthetic performance, reactive oxygen species (ROS) production, antioxidative enzyme activities, Cd uptake and accumulation of A. manihot. Exposure to cadmium enhanced plant growth even at 100 mg kg?1, without showing symptoms of visible damage. The cadmium concentration of shoots (stems or leaves) and roots was more than the critical value of 100 mg kg?1 and reached 126.17, 185.26 and 210.24 mg kg?1, respectively. BCF values of A. manihot plants exceeded the reference value 1.0 for all the Cd treatments, and TF values were greater than 1 at 15–60 mg kg?1 Cd treatment. The results also showed that cadmium concentrations of 60 mg kg?1 or less induced a significant enhancement in plant net photosynthetic rate (A N), stomatal conductance (G s), transpiration rate (T r), photosynthetic pigments and F v/F m. These parameters were slightly decreased at the higher concentration (100 mg kg?1). The ROS production (O2 ?, H2O2) and antioxidative response including SOD, CAT and POD were significantly enhanced by increasing cadmium. These results suggest that A. manihot can be considered as a Cd-hyperaccumulator and the hormetic effects may be taken into consideration in remediation of Cd contamination soil.  相似文献

Impact of different levels of elevated CO 2 on the activity of Frankia (Nitrogen-fixing actinomycete) in Casuarina equisetifolia rooted stem cuttings has been studied to understand the relationship between C. equisetifolia, Frankia and CO2. The stem cuttings of C. equietifolia were collected and treated with 2000 ppm of Indole Butyric Acid (IBA) for rooting. Thus vegetative propagated rooted stem cuttings of C. equisetifolia were inoculated with Frankia and placed in the Open top chambers (OTC) with elevated CO2 facilities. These planting stocks were maintained in the OTC for 12 months under different levels of elevated CO2 (ambient control, 600 ppm, 900 ppm). After 12 months, the nodule numbers, bio mass, growth, and photosynthesis of C. equisetifolia rooted stem cuttings inoculated with Frankia were improved under 600 ppm of CO2. The rooted stem cuttings of C. equisetifolia inoculated with Frankia showed a higher number of nodules under 900 ppm of CO2 and cuttings without Frankia inoculation exhibited poor growth. Tissue Nitrogen (N) content was also higher under 900 ppm of CO2 than ambient control and 600 ppm levels. The photosynthetic rate was higher (17.8 μ mol CO2 m?2 s?1) in 900 ppm of CO2 than in 600 ppm (13.2 μ mol CO2 m?2 s?1) and ambient control (8.3 μ mol CO2 m?2 s?1). This study showed that Frankia can improve growth, N fixation and photosynthesis of C. equietifolia rooted stem cuttings under extreme elevated CO2 level conditions (900 ppm).  相似文献

Inheritance pattern of wood traits viz. specific gravity, fibre dimensions and fibre-derived biometrical indices and their interactions among themselves and with that of growth are reported in Hevea brasiliensis. Girth (h2 =???0.02?±?0.44 to h2 =?0.35?±?0.24) showed moderate genetic control. Among wood traits, specific gravity (h2?=?0.15?±?0.31 to h2 =?0.33?±?0.28) was found to be under moderate genetic control. Fibre traits viz., fibre length (h2 =???0.26?±?0.30 to h2 =?0.50?±?0.34), fibre diameter (h2 =?0.19?±?0.49 to h2 =?0.70?±?0.11), fibre lumen diameter (h2 =???0.18?±?0.35 to h2 =?0.56?±?0.47) and fibre wall thickness (h2 =???5.17?±?5.26 to h2 =?0.50?±?0.50) were under moderate to strong genetic control. Among fibre-derived indices, flexibility coefficient (h2 =?0.48?±?0.21 to h2 =?0.89?±?0.29) showed moderate to very strong genetic control. The Runkel ratio (h2 =???0.40?±?0.27 to h2 =?0.42?±?0.29) and slenderness ratio (h2 =???0.36?±?0.29 to h2 =?0.43?±?0.28) showed moderate genetic control. Girth showed very strong positive genetic correlation with fibre wall thickness and strong positive correlation with fibre width indicating scope of indirect selection potential for these traits. Wood specific gravity was not correlated with either girth or fibre traits. Hence, it would be possible to concomitantly improve growth and fibre traits without adversely affecting wood specific gravity. Moderate to very high estimates of heritability for fibre traits, girth and specific gravity indicated that considerable genetic gain can be realised for these traits. Implications of the above findings in genetic improvement of wood in Hevea are discussed.  相似文献

Photosynthetic parameters including net photosynthetic rate (PN), transpiration rate (E), water-use efficiency (WUE), and stomatal conductance (gs) were studied in indoor C3 plants Philodendron domesticum (Pd), Dracaena fragans (Df), Peperomia obtussifolia (Po), Chlorophytum comosum (Cc), and in a CAM plant, Sansevieria trifasciata (St), exposed to various low temperatures (0, 5, 10, 15, 20, and 25°C). All studied plants survived up to 0°C, but only St and Cc endured, while other plants wilted, when the temperature increased back to room temperature (25°C). The PN declined rapidly with the decrease of temperature in all studied plants. St showed the maximum PN of 11.9 μmol m?2 s?1 at 25°C followed by Cc, Po, Pd, and Df. E also followed a trend almost similar to that of PN. St showed minimum E (0.1 mmol m?2 s?1) as compared to other studied C3 plants at 25°C. The E decreased up to ≈4-fold at 5 and 0°C. Furthermore, a considerable decline in WUE was observed under cold stress in all C3 plants, while St showed maximum WUE. Similarly, the gs also declined gradually with the decrease in the temperature in all plants. Among C3 plants, Pd and Po showed the maximum gs of 0.07 mol m?2 s?1 at 25°C followed by Df and Cc. However, St showed the minimum gs that further decreased up to ~4-fold at 0°C. In addition, the content of photosynthetic pigments [chlorophyll a, b, (a+b), and carotenoids] was varying in all studied plants at 0°C. Our findings clearly indicated the best photosynthetic potential of St compared to other studied plants. This species might be recommended for improving air quality in high-altitude closed environments.  相似文献

Terminalia arjuna (Ta) bark contains various natural antioxidants and has been used to protect animal cells against oxidative stress. In the present study, we have examined alleviating effects of Ta bark aqueous extract against Ni toxicity in rice (Oryza sativa L.). When rice seedlings were raised for 8 days in hydroponics in Yoshida nutrient medium containing 200 μM NiSO4, a decline in height, reduced biomass, increased Ni uptake, loss of root plasma membrane integrity, increase in the level of O2˙?, H2O2 and ˙OH, increased lipid peroxidation, decline in photosynthetic pigments, increase in the level of antioxidative enzymes superoxide dismutase, catalase and glutathione peroxidase and alterations in their isoenzyme profile patterns were observed. Transmission electron microscopy (TEM) showed damage to chloroplasts marked by disorganised enlarged starch granules and disrupted thylakoids under Ni toxicity. Exogenously adding Ta bark extract (3.2 mg ml?1) to the growth medium considerably alleviated Ni toxicity in the seedlings by reducing Ni uptake, suppressing generation of reactive oxygen species, reducing lipid peroxidation, restoring level of photosynthesis pigments and ultrastructure of chloroplasts, and restoring levels of antioxidative enzymes. Results suggest that Ta bark extract considerably alleviates Ni toxicity in rice seedlings by preventing Ni uptake and reducing oxidative stress in the seedlings.  相似文献

Net photosynthetic rate (PN), transpiration rate (E), water use efficiency (WUE), stomatal conductance (gs), and stomatal limitation (Ls) were investigated in two Syringa species. The saturation irradiance (SI) was 400 µmol m-2s-1 for S. pinnatifolia and 1 700 µmol m-2s-1 for S. oblata. Compared with S. oblata, S. pinnatifolia had extremely low g s . Unlike S. oblata, the maximal photosynthetic rate (Pmax) in S. pinnatifoliaoccurred around 08:00 and then fell down, indicating this species was sensitive to higher temperature and high photosynthetic photon flux density. However, such phenomenon was interrupted by the leaf development rhythms before summer. A relatively lower PN together with a lower leaf area and shoot growth showed the capacity for carbon assimilation was poorer in S. pinnatifolia.  相似文献

A Gram-stain negative, aerobic, motile by flagella, rod-shaped strain (THG-T16T) was isolated from rhizosphere of Hibiscus syriacus. Growth occurred at 10–40 °C (optimum 28–30 °C), at pH 6.0–8.0 (optimum 7.0) and at 0–1.0% NaCl (optimum 0%). Based on 16S rRNA gene sequence analysis, the near phylogenetic neighbours of strain THG-T16T were identified as Nibribacter koreensis KACC 16450T (98.6%), Rufibacter roseus KCTC 42217T (94.7%), Rufibacter immobilis CCTCC AB 2013351T (94.5%) and Rufibacter tibetensis CCTCC AB 208084T (94.4%). The DNA G+C content of strain THG-T16T was determined to be 46.7 mol%. DNA–DNA hybridization values between strain THG-T16T and N. koreensis KACC 16450T, R. roseus KCTC 42217T, R. immobilis CCTCC AB 2013351T, R.tibetensis CCTCC AB 208084T were 33.5?±?0.5% (31.7?±?0.7% reciprocal analysis), 28.1?±?0.2% (25.2?±?0.2%), 17.1?±?0.9% (10.2?±?0.6%) and 8.1?±?0.3% (5.2?±?0.1%). The polar lipids were identified as phosphatidylethanolamine, two unidentified aminophospholipids, an unidentified aminolipid and three unidentified lipids. The quinone was identified as MK-7 and the polyamine as sym-homospermidine. The major fatty acids were identified as C16:1 ω5c, C17:1 ω6c, iso-C15:0, summed feature 3 (C16:1 ω7c and/or C16:1 ω6c) and summed feature 4 (iso-C17:1 I and/or anteiso-C17:1 B). On the basis of the phylogenetic analysis, chemotaxonomic data, physiological characteristics, and DNA–DNA hybridization data, strain THG-T16T represents a novel species of the genus Nibribacter, for which the name Nibribacter flagellatus sp. nov. is proposed. Horse heart carboxymethylated cytc (CM-cytc) displays myoglobin-like properties. Here, the effect of cardiolipin (CL) liposomes on the nitrite reductase activity of ferrous CM-cytc [CM-cytc-Fe(II)], in the presence of sodium dithionite, is reported between pH 5.5 and 7.6, at 20.0 °C. Cytc-Fe(II) displays a very low value of the apparent second-order rate constant for the NO2 ?-mediated conversion of cytc-Fe(II) to cytc-Fe(II)-NO [k on = (7.3 ± 0.7) × 10?2 M?1 s?1; at pH 7.4], whereas the value of k on for NO2 ? reduction by CM-cytc-Fe(II) is 1.1 ± 0.2 M?1 s?1 (at pH 7.4). CL facilitates the NO2 ?-mediated nitrosylation of CM-cytc-Fe(II) in a dose-dependent manner, the value of k on for the NO2 ?-mediated conversion of CL–CM-cytc-Fe(II) to CL–CM-cytc-Fe(II)-NO (5.6 ± 0.6 M?1 s?1; at pH 7.4) being slightly higher than that for the NO2 ?-mediated conversion of CL–cytc-Fe(II) to CL–cytc-Fe(II)-NO (2.6 ± 0.3 M?1 s?1; at pH 7.4). The apparent affinity of CL for CM-cytc-Fe(II) is essentially pH independent, the average value of B being (1.3 ± 0.3) × 10?6 M. In the absence and presence of CL liposomes, the nitrite reductase activity of CM-cytc-Fe(II) increases linearly on lowering pH and the values of the slope of the linear fittings of Log k on versus pH are ?1.05 ± 0.07 and ?1.03 ± 0.03, respectively, reflecting the involvement of one proton for the formation of the transient ferric form, NO, and OH?. These results indicate that Met80 carboxymethylation and CL binding cooperate in the stabilization of the highly reactive heme-Fe atom of CL–CM-cytc.  相似文献

Objectives

To enhance activity of cis-epoxysuccinate hydrolase from Klebsiella sp. BK-58 for converting cis-epoxysuccinate to tartrate.

Results

By semi-saturation mutagenesis, all the mutants of the six important conserved residues almost completely lost activity. Then random mutation by error-prone PCR and high throughput screening were further performed to screen higher activity enzyme. We obtained a positive mutant F10D after screening 6000 mutations. Saturation mutagenesis on residues Phe10 showed that most of mutants exhibited higher activity than the wild-type, and the highest mutant was F10Q with activity of 812 U mg?1 (k cat /K m , 9.8 ± 0.1 mM?1 s?1), which was 230 % higher than that of wild-type enzyme 355 U mg?1 (k cat /K m , 5.3 ± 0.1 mM?1 s?1). However, the thermostability of the mutant F10Q slightly decreased.

Conclusions

The catalytic activity of a cis-epoxysuccinate hydrolase was efficient improved by a single mutation F10Q and Phe10 might play an important role in the catalysis.

Guadua amplexifolia and Guadua angustifolia are the most promising timber substitutes amongst American bamboos due to their outstanding dimensions and structural properties. Despite the commercial potential of these species, there are few studies on the survival and adaptability of juveniles in plantations. The present study dealt with survival, growth, and ecophysiological response of juvenile clonal plants of these species, cultivated in abandoned pastures in Mérida, Venezuela. Survivorship, growth (height and culm diameter), and ecophysiological parameters were monitored the first year during wet and dry seasons. Survival rates were high in both species (95% in G. amplexifolia and 89% in G. angustifolia). Midday leaf water potentials decreased in both species during dry months (–1.28 to–2.72 MPa in G. amplexifolia and–1.67 to–2.37 MPa in G. angustifolia, respectively). Net photosynthetic rates measured during wet [16.57 ± 1.40 and 13.68 ± 2.40 μmol(CO2) m–2 s–1, respectively] and dry seasons [12.19 ± 2.82 and 8.12 ± 1.81 μmol(CO2) m–2 s–1, respectively], demonstrated that G. amplexifolia maintained consistently higher photosynthetic rates compared to G. angustifolia, which could explain the higher growth rates of the former. Similar trends were observed for stomatal conductance, transpiration, water-use efficiency, electron transport rate, and photochemical quenching of PSII. G. angustifolia maintained higher nonphotochemical quenching as well as a higher consumption of electrons per molecule of CO2 fixed, indicating a lower photosynthetic efficiency. The maximal photochemical efficiency of PSII (0.73–0.76) suggested that neither of these species suffered from photoinhibition, despite persistently high radiation and air temperatures at the study site.  相似文献

Effects of oxygen transfer on recombinant protein production by Pichia pastoris under glyceraldehyde-3-phosphate dehydrogenase promoter were investigated. Recombinant glucose isomerase was chosen as the model protein. Two groups of oxygen transfer strategies were applied, one of which was based on constant oxygen transfer rate where aeration rate was Q O/V = 3 and 10 vvm, and agitation rate was N = 900 min?1; while the other one was based on constant dissolved oxygen concentrations, C DO = 5, 10, 15, 20 and 40 % in the fermentation broth, by using predetermined exponential glucose feeding with μ o = 0.15 h?1. The highest cell concentration was obtained as 44 g L?1 at t = 9 h of the glucose fed-batch phase at C DO = 20 % operation while the highest volumetric and specific enzyme activities were obtained as 4440 U L?1 and 126 U g?1 cell, respectively at C DO = 15 % operation. Investigation of specific enzyme activities revealed that keeping C DO at 15 % was more advantageous with an expense of relatively higher by-product formation and lower specific cell growth rate. For this strategy, the highest oxygen transfer coefficient and oxygen uptake rate were K L a = 0.045 s?1 and OUR = 8.91 mmol m?3 s?1, respectively.  相似文献

Sublethal concentrations of chemical insecticides may cause changes in some behavioral characteristics of natural enemies such as functional responses. The residual effect of three synthetic insecticides including deltamethrin, fenvalerate and azadirachtin were studied on functional response of Habrobracon hebetor Say to Ephestia kuehniella Zeller larvae. Seven host densities (2, 4, 8, 16, 32, 64 and 96) were used during a 24 h period. The resulting data were appropriately fit to Type II functional response models in all treatments: (1) control (0.0916 h?1; and T h  = 0.2011 h); (2) deltamethrin (a = 0.0839 h?1; and T h  = 0.3560 h); (3) fenvalerate (a = 0.0808 h?1 and T h  = 0.3623 h); and (4) azadirachtin (a = 0.0900 h?1 and T h  = 0.2042 h). Maximum theoretical parasitism rate (T/T h ) was 119.34 estimated for control wasps. There was no significant difference between the values of attack rates (a and a + D a ) in all treatments while the handling time was statistically affected in female wasps treated with fenvalerate. Our findings will be useful in safe application of these insecticides in pest management programmes.  相似文献

In this study, we presented cellular morphological changes, time-resolved biochemical composition, photosynthetic performance and proteomic profiling to capture the photosynthetic physiological response of Scenedesmus acuminatus under low nitrogen (3.6 mM NaNO3, N?) and high nitrogen supplies (18.0 mM NaNO3, N+). S. acuminatus cells showed extensive lipid accumulation (53.7% of dry weight) and were enriched in long-chain fatty acids (C16 & C18) under low nitrogen supply. The activity of PSII and photosynthetic rate decreases, whereas non-photochemical quenching and dark respiration rates were increased in the N? group. In addition, the results indicated a redistribution of light excitation energy between PSII and PSI in S. acuminatus exists before lipid accumulation. The iTRAQ results showed that, under high nitrogen supply, protein abundance of the chlorophyll biosynthesis, the Calvin cycle and ribosomal proteins decreased in S. acuminatus. In contrast, proteins associated with the photosynthetic machinery, except for F-type ATPase, were increased in the N+ group (N+, 3 vs. 9 days and 3 days, N+ vs. N?). Under low nitrogen supply, proteins involved in central carbon metabolism, fatty acid synthesis and branched-chain amino acid metabolism were increased, whereas the abundance of proteins of the photosynthetic machinery had decreased, with exception of PSI (N?, 3 vs. 9 days and 9 days, N+ vs. N?). Collectively, the current study has provided a basis for the metabolic engineering of S. acuminatus for biofuel production.  相似文献

Saltol, the major salinity tolerance quantitative trait loci (QTL) in rice, was introgressed from IR66946-3R-230-1-1 (FL530) into Khao Dawk Mali 105 (KDMl105) by two rounds of marker-assisted backcrossing (MAB). Twenty-eight BC2F2 introgression lines (BILs) with positive Saltol allele (BIL+Saltol) and 19 BILs with negative Saltol allele (BIL?Saltol) were validated for the effect of Saltol as key salinity tolerant trait at seedling stage. A hydrophonic system with salt stress of 12 dS m?1 (130 mM Na+) was conducted, and significant differences between BILs+Saltol and BILs?Saltol were observed for the period of plant survival (PPS), total K+ (T-K+) and Na+ (T-Na+) concentration, whole plant Na+-K+ ratio (T-Na+/K+), shoot Na+ (S-Na+) and K+ (S-K+) concentration, and shoot Na+-K+ ratio (S-Na+/K+). BILs+Saltol displayed higher PPS, uptake less Na+ (T-Na+; 43.4 ppm), and more K+ (T-K+; 30.9 ppm), while the BILs?Saltol uptake more Na+ (T-Na+; 45.7 ppm) and less K+ (T-K+; 28.2 ppm). Direct effects on PPS and salt injury score (SIS) were observed, indicating Na+/K+ homeostasis mechanism by the Saltol under hydrophonic salt stress. All BILs+Saltol recovered KDML105 cooking quality profile such as low apparent amylose content (AAC), high score of alkaline spreading value (ASV), intermediate gel consistency (GC), and strong fragrance. Botryococcus braunii is a green microalga capable of producing large amounts of external long-chain hydrocarbons suitable as a source of biofuel. There have been several studies indicating that cultures of B. braunii can reduce the energy and water requirement for mass biofuel production, especially if non-destructive extraction methods for milking hydrocarbons are used. Growing microalgae as a raw material for biofuel using conventional liquid-based cultivation (i.e., raceway ponds) has yet to be shown to be economically successful. An alternative solid growth (biofilm) cultivation method can markedly reduce the energy requirements and costs associated with the harvesting and dewatering processes. We evaluated the growth of biofilms of several strains of B. braunii (from races A, B, L and S) and found that three of the four tested races successfully grew to stationary phase in 10 weeks with no contamination. Among all races, B. braunii BOT22 (race B) reached the highest biomass and lipid yields (3.80 mg dry weight cm?2 day?1 and 1.11 mg dry weight cm?2). Irrespective of the race, almost all photosynthetic parameters (F V /F 0 , PIABS and the OJIP curve) showed that the biofilm cultures were more stressed during lag and stationary phases than in logarithmic phase. We also studied the Botryococcus biofilm profiles using confocal microscopy and found that this method is suitable for estimating the overall biomass yield when compared with gravimetric measurement. In conclusion, the growth characteristics (biomass and lipid) and photosynthetic performance of all races indicated that B. braunii BOT22 is the most promising strain for biofilm cultivation.  相似文献

Phycobiliproteins, light-harvesting pigments found in cyanobacteria and in some eukaryotic algae, have numerous commercial applications in food, cosmetic, and pharmaceutical industries. Colorant production from cyanobacteria offers advantages over their production from higher plants, as cyanobacteria have fast growth rate and high photosynthetic efficiency and require less space. In this study, three cyanobacteria strains were studied for phycobiliprotein production and the influence of sodium nitrate, potassium nitrate and ammonium chloride on the growth and phycobiliprotein composition of the strains were evaluated. In the batch culture period of 12 days, Phormidium sp. and Pseudoscillatoria sp. were able to utilize all tested nitrogen sources; however, ammonium chloride was the best nitrogen source for both strains to achieve maximum growth rate μ?=?0.284?±?0.03 and μ?=?0.274?±?0.13 day?1, chlorophyll a 16.2?±? 0.5 and 12.2?±? 0.2 mg L?1, and phycobiliprotein contents 19.38?±?0.09 and 19.99?±?0.14% of dry weight, whereas, for Arthrospira platensis, the highest growth rate of μ?=?0.304?±?0.0 day?1, chlorophyll a 19.1?±?0.5 mg L?1, and phycobiliprotein content of 22.27?±?0.21% of dry weight were achieved with sodium nitrate. The phycocyanin from the lyophilized cyanobacterial biomass was extracted using calcium chloride and food grade purity (A620/A280 ratio >?0.7) was achieved. Furthermore, phycocyanin was purified using two-step chromatographic method and the analytical grade purity (A620/A280 ratio >?4) was attained. SDS-PAGE demonstrated the purity and presence of two bands corresponding to α- and β-subunits of the C-phycocyanin. The results showed that Phormidium sp. and Pseudoscillatoria sp. could be good candidates for phycocyanin production.  相似文献

A recombinant alcohol dehydrogenase (ADH) from Kangiella koreensis was purified as a 40 kDa dimer with a specific activity of 21.3 nmol min?1 mg?1, a K m of 1.8 μM, and a k cat of 1.7 min?1 for all-trans-retinal using NADH as cofactor. The enzyme showed activity for all-trans-retinol using NAD + as a cofactor. The reaction conditions for all-trans-retinol production were optimal at pH 6.5 and 60 °C, 2 g enzyme l?1, and 2,200 mg all-trans-retinal l?1 in the presence of 5 % (v/v) methanol, 1 % (w/v) hydroquinone, and 10 mM NADH. Under optimized conditions, the ADH produced 600 mg all-trans-retinol l?1 after 3 h, with a conversion yield of 27.3 % (w/w) and a productivity of 200 mg l?1 h?1. This is the first report of the characterization of a bacterial ADH for all-trans-retinal and the biotechnological production of all-trans-retinol using ADH.  相似文献
