A range of feed supplements, including antibiotics, have been commonly used in poultry production to improve health and productivity. Alternative methods are needed to suppress pathogen loads and maintain productivity. As an alternative to antibiotics use, we investigated the ability of biochar, bentonite and zeolite as separate 4% feed additives, to selectively remove pathogens without reducing microbial richness and diversity in the gut. Neither biochar, bentonite nor zeolite made any significant alterations to the overall richness and diversity of intestinal bacterial community. However, reduction of some bacterial species, including some potential pathogens was detected. The microbiota of bentonite fed animals were lacking all members of the order Campylobacterales. Specifically, the following operational taxonomic units (OTUs) were absent: an OTU 100% identical to Campylobacter jejuni; an OTU 99% identical to Helicobacter pullorum; multiple Gallibacterium anatis (>97%) related OTUs; Bacteroides dorei (99%) and Clostridium aldenense (95%) related OTUs. Biochar and zeolite treatments had similar but milder effects compared to bentonite. Zeolite amended feed was also associated with significant reduction in the phylum Proteobacteria. All three additives showed potential for the control of major poultry zoonotic pathogens.  相似文献

Because there is considerable variation in gene expression even between closely related species, it is clear that gene regulatory mechanisms evolve relatively rapidly. Because primary sequence conservation is an unreliable proxy for functional conservation of cis-regulatory elements, their assessment must be carried out in vivo. We conducted a survey of cis-regulatory conservation between C. elegans and closely related species C. briggsae, C. remanei, C. brenneri, and C. japonica. We tested enhancers of eight genes from these species by introducing them into C. elegans and analyzing the expression patterns they drove. Our results support several notable conclusions. Most exogenous cis elements direct expression in the same cells as their C. elegans orthologs, confirming gross conservation of regulatory mechanisms. However, the majority of exogenous elements, when placed in C. elegans, also directed expression in cells outside endogenous patterns, suggesting functional divergence. Recurrent ectopic expression of different promoters in the same C. elegans cells may reflect biases in the directions in which expression patterns can evolve due to shared regulatory logic of coexpressed genes. The fact that, despite differences between individual genes, several patterns repeatedly emerged from our survey, encourages us to think that general rules governing regulatory evolution may exist and be discoverable.  相似文献

After many years of the excessive use of praziquantel against Schistosoma mansoni (S. mansoni), it has already led to the development of drug resistance. While schistosomiasis is still affecting millions of people every year, vaccination may be one realistic alternative way to control the disease. Currently, S. mansoni 14-kDa fatty acid-binding protein (Sm14) has shown promising results as a vaccine antigen. Yet, the use of an adjuvant may be necessary to further increase the effectiveness of the vaccine. Herein, we investigated the potential of using heat-killed Cutibacterium acnes (C. acnes) as an adjuvant for recombinant Sm14 (rSm14). Immunization of mice with C. acnes-adjuvanted rSm14 showed increased humoral immune responses, compared with mice immunized with rSm14 alone. Additionally, C. acnes-adjuvanted rSm14 vaccination provided higher protection to mice against S. mansoni infection and liver injuries. These results suggest that C. acnes increases the immunogenicity of rSm14, which leads to better protection against S. mansoni infection. Therefore, heat-killed C. acnes may be a promising adjuvant to use with rSm14.  相似文献

A collection of over 200 petrified Middle Devonian plants was made from a single locality near Cairo, New York. This paper represents the second of a series enumerating the plants of the flora. Reimannia aldenense with a single lateral appendage containing a terete xylem strand that divides is present. Reimannia is thought to represent young branches within the Aneurophytales. Many of the sections made for the study of the flora contain small, terete axes. Some have a very distinctive epidermis. Some dichotomize several times. Cairoa lamanekii gen. et sp. n. shows closest affinity to the Aneurophytales yet differs in that the shape of the primary xylem is not consistently repeated from one order of axis to the next. It is suggested that Cairoa and Proteokalon represent a distinctive subgroup in the Aneurophytales.  相似文献

Background

Ten secreted aspartyl proteinase (Sap) genes were identified in Candida albicans. The products of SAP genes are considered to be virulent factors of C. albicans that participated in causing mucocutaneous and systemic candidiasis in humans. Depending on environmental conditions, C. albicans may stay in yeast-form or convert into invasive hypha-form, and these issues may affect the expression of SAP genes. In this study we explored the component(s) of culture media that may affect the expression of hypha-associated SAP genes.

Results

We demonstrate that glucose levels modulate both the hyphae development and the expression strength of hypha-associated SAP genes (SAP4-6). In contrast to high glucose concentration (2%), lower glucose level (0.1%) is more potent to promote hyphae development and to promptly elicit the expression of hypha-associated Sap proteins during yeast-to-hypha transition of C. albicans. Both Cph1-mediated MAP kinase cascade and Efg1-mediated cAMP/PKA pathway, although the latter seemed dominant, participate in convey the glucose signaling to regulate the expression of hypha-associated SAP genes and this glucose level effect may perform at very early stage of yeast-to-hypha transition. In addition, when C. albicans was co-cultured with THP-1 human monocytes, the engulfed C. albicans was developing hypha efficiently within 1 hr and the expression of hypha-associated Sap proteins could be detected on the distal surface of hyphae.

Conclusion

We propose that the glucose level of bloodstream (approximately 0.1%) may be facilitated for stimulation of C. albicans to develop invasive hypha-form and to elicit promptly production of high-level hypha-associated Sap proteins.  相似文献

We determined partial sequences of the gyrA and parC genes of Citrobacter freundii type strain, and then examined 38 C. freundii clinical strains isolated from patients with urinary tract infections for the association of alterations in GyrA and ParC with susceptibility to fluoroquinolones. Our results suggest that in C. freundii DNA gyrase may be a primary target of quinolones, that an amino acid change at Thr-83 or Asp-87 in GyrA is sufficient to decrease susceptibility to fluoroquinolones, and that accumulation of changes in GyrA with the simultaneous presence of an alteration at Ser-80 or Glu-84 in ParC may be associated with the development of high-level fluoroquinolone resistance in C. freundii clinical isolates.  相似文献

The use of granulocyte-rich concentrates from leukapheresis purified by counterflow centrifugation—elutriation to obtain pure granulocytes for transfusion studies in cyclo-phosphamide-induced neutropenic animal models is reported. Our data for granulocyterich leukapheresis concentrates indicate that room temperature (20 °C) appears to be preferred to 6 °C for short-term granulocyte storage. The data also indicate that although the granulocytes isolated by counterflow centrifugation—elutration may retain in vitro functions of chemotaxis, phagocytosis, and bactericidal activity, the in vivo function of migration into skin chambers for isolated granulocytes is seriously impaired after storage for 18 to 24 hr at both 6 and 20 °C. This loss of in vivo function of stored granulocytes occurs in isolated granulocytes obtained by both counterflow centrifugation-elutriation and dextran sedimentation, and it is not observed in the leukocyte concentrates held at 20 °C. The results of these studies are fourfold. First, freshly isolated granulocytes display no apparent loss of either in vivo or in vitro function. Second, granulocytes isolated by counterflow centrifugation-elutriation or dextran sedimentation and stored at 6 or 20 °C are severely impaired in terms of their in vivo chemotactic function but display no loss of in vitro efficacy. Third, 20 °C storage of granulocyte-rich leukapheresis concentrates for 18 to 24 hr is superior to 6 °C storage. Fourth, in vitro analysis may be limited in its ability to indicate in vivo function as a measure of success in granulocyte preservation studies.  相似文献

Corynosoma strumosum (Acanthocephala), a widespread parasite of pinnipeds, is reported in marine foraging North American mink (Neogale vison) and river otter (Lontra canadensis) on Vancouver Island, British Columbia. This is the first confirmed case of infection by C. strumosum in river otters on the west coast of North America and may be the first confirmed case of infection in wild North American mink; C. strumosum has previously been reported in river otters in Europe (Lutra lutra) and in farmed mink fed with marine fish. We also detected a case of acanthocephalan associated peritonitis in a juvenile mink. Furthermore, though infections with Corynosoma spp. are often assumed to be accidental in mustelids, some C. strumosum individuals found in mink showed signs of reproductive activity. These findings indicate that mink may be a competent definitive host and represent a reservoir in coastal habitats although further research is needed to confirm this. Investigating whether river otters may be competent hosts and determine the prevalence of infection in coastal populations would determine the potential implications of C. strumosum for coastal otters and minks. Our report indicates that mink and possibly river otter living in coastal areas are vulnerable to this previously unreported parasitic infection with mortality risk, at least in juvenile individuals.  相似文献

Background

Chlamydia pneumoniae is an obligate intracellular respiratory pathogen for humans. Infection by C. pneumoniae may be linked etiologically to extra-respiratory diseases of aging, especially atherosclerosis. We have previously shown that age promotes C. pneumoniae respiratory infection and extra-respiratory spread in BALB/c mice.

Findings

Aged C57BL/6 mice had a greater propensity to develop chronic and/or progressive respiratory infections following experimental intranasal infection by Chlamydia pneumoniae when compared to young counterparts. A heptavalent CTL epitope minigene (CpnCTL7) vaccine conferred equal protection in the lungs of both aged and young mice. This vaccine was partially effective in protecting against C. pneumoniae spread to the cardiovascular system of young mice, but failed to provide cardiovascular protection in aged animals.

Conclusions

Our findings suggest that vaccine strategies that target the generation of a C. pneumoniae-specific CTL response can protect the respiratory system of both young and aged animals, but may not be adequate to prevent dissemination of C. pneumoniae to the cardiovascular system or control replication in those tissues in aged animals.  相似文献

A numerical taxonomic study was conducted on the common narrow-leaved members of the genusChenopodium that occur in the western United States. As many as six and as few as two species have been recognized. Thirty-five individual popu lations were used as OTU’s, and 35 characters were considered. The data were subjected to cluster (both the weighted-pair group method and the within-group dispersion method) and principal component analysis. The results of both analyses are in general agreement and suggest phenetic relationships that differ from current interpretations in the literature. It is concluded thatC. desiccatum var.leptophylloides is much more closely related phenetically toC. atrovirens (and may not be distinct from it) than it is toC. desiccatum var.desiccatum. Chenopodium hians andC. leptophyllum appear to be phenetically distinct despite the fact that they have sometimes been viewed as conspecific.Cheno podium subglabrum is not very similar toC. leptophyllum, and should probably be treated as a separate species rather than as a variety of the latter. Popu lations referable toC. incognitum form a rather loose cluster that appears some what intermediate between populations ofC. desiccatum var.leptophylloides,C. atrovirens, C. hians, andC. leptophyllum.  相似文献

Candida albicans and Candida glabrata are predominant fungi associated with oral candidiasis. Histatin 5 (Hst 5) is a small cationic human salivary peptide with high fungicidal activity against C. albicans, however many strains of C. glabrata are resistant. Since Hst 5 requires fungal binding to cell wall components prior to intracellular translocation, reduced Hst 5 binding to C. glabrata may be the reason for its insensitivity. C. glabrata has higher surface levels of β-1,3-glucans as compared with C. albicans; however these differences did not account for reduced Hst 5 uptake and killing in C. glabrata. Similarly, the biofilm matrix of C. glabrata contained significantly higher levels of β-1,3-glucans compared with C. albicans, but it did not reduce the percentage of Hst 5 positive fungal cells in the biofilm. Hst 5 enters C. albicans cell through polyamine transporters Dur3p and Dur31p that are uncharacterized in C. glabrata. C. glabrata strains expressing CaDur3 and CaDur31 had two-fold higher killing and uptake of Hst 5. Thus, neither C. glabrata cell surface or biofilm matrix β-1,3-glucan levels affected Hst 5 toxicity; rather the crucial rate limiting step is reduced uptake that can be overcome by expression of C. albicans Dur proteins in C. glabrata.  相似文献

Odonate nymphs are voracious predators and may be useful natural control agents. However, odonates are normally found in complex guilds with a high degree of conspecific and inter specific predation that may reduce their overall predation efficiency. The present study investigates whether indirect interactions, mediated via chemical predation signals, inhibit the consumption of mosquito larvae by two common odonates: Crocothemis erythraea and Ischnura evansi. Our results show that the predation rate of C. erythraea is reduced by chemical cues from Anax imperator, but the response of I. evansi to C. erythraea was statistically non-significant. This study shows that intra-guild interactions may limit the effectiveness of odonates as predators of mosquitoes.  相似文献

Plastoquinones A and C have been found in all classes of algae, including representatives of greens, yellow-greens, blue-greens, reds, browns and the flagellate, Euglena. Plastoquinone C from red and brown algae can be separated into 6 different types. An additional plastoquinone C has been found in Gigartina and Rhydomela. From chromatographic evidence this may be equivalent to plastoquinone Co, a C type with a hydroxyl group on the first isoprene unit of the terpenoid sidechain of this substituted benzo-quinone. The ubiquinone, vitamin K and α-tocopherylquinone content of several algae is also reported. The presence of plastoquinone A in all green plants and many algae indicates that it may be a functional element in photosynthesis. Our study shows that plastoquinone C is more regularly present in algae than has been previously shown.  相似文献

The genera Dissodactylus and Clypeasterophilus are commensal or parasitic crabs that live in association with irregular sea urchins. Only a single pinnotherid species is expected to occur on a sand dollar; however, during field observations, D. crinitichelis and Clypeasterophilus stebbingi were found co-occurring on a single host (Clypeaster subdepressus). Laboratory experiments and in situ observations were conducted to explore mechanisms of host selection and use by C. stebbingi, as well as the possibility of competition with D. crinitichelis. Analysis of 213 specimens of Encope emarginata and 33 C. subdepressus revealed a high frequency of infestation (90 %) by brachyurans. The number of D. crinitichelis was positively correlated with the size of E. emarginata. During field observations and experiments, D. crinitichelis was able to colonize both echinoids, while C. stebbingi was found mainly on C. subdepressus. No exclusion was reported between individuals of the same sex for both crab species. However, D. crinitichelis may be territorial, since a single male occurred when adult females were added to the host. D. crinitichelis and C. stebbingi co-occurred on the same individual of C. subdepressus on rare occasions during field observations. C. stebbingi may exclude D. crinitichelis from the echinoid to avoid competition, as observed during the experiments. We suggest a species-specific association between C. stebbingi and C. subdepressus, while D. crinitichelis associated mainly with E. emarginata, but also with C. subdepressus. Both crabs may harm the echinoids by consuming their spines, but additional research is necessary to determine whether this is a parasitic association.  相似文献
