Human serum albumin (HSA) displays esterase activity reflecting multiple irreversible chemical modifications rather than turnover. Here, kinetics of the pseudo-enzymatic hydrolysis of 4-nitrophenyl acetate (NphOAc) are reported. Under conditions where [HSA] ? 5×[NphOAc] and [NphOAc] ? 5×[HSA], the HSA-catalyzed hydrolysis of NphOAc is a first-order process for more than 95% of its course. From the dependence of the apparent rate constants kapp and kobs on [HSA] and [NphOAc], respectively, values of Ks, k+2, and k+2/Ks were determined. Values of Ks, k+2, and k+2/Ks obtained at [HSA] ? 5×[NphOAc] and [NphOAc] ? 5×[HSA] are in good agreement, the deacylation step being rate limiting in catalysis. The pH-dependence of k+2/Ks, k+2, and Ks reflects the acidic pKa shift of the Tyr411 catalytic residue from 9.0 ± 0.1 in the substrate-free HSA to 8.1 ± 0.1 in the HSA:NphOAc complex. Accordingly, diazepam inhibits competitively the HSA-catalyzed hydrolysis of NphOAc by binding to Tyr411.  相似文献

Abstract

Endogenous S-nitrosated human serum albumin (E-Mono-SNO-HSA) is a large molecular weight nitric oxide (NO) carrier in human plasma, which has shown many beneficial effects in different animal models. To construct more efficient SNO-HSA preparations, SNO-HSA with many conjugated SNO groups has been prepared using chemical modification (CM-Poly-SNO-HSA). We have compared the properties of such a preparation to those of E-Mono-SNO-HSA. Cellular uptake of NO from E-Mono-SNO-HSA partly takes place via low molecular weight thiol, and it results in cytoprotective effects by induction of heme oxygenase-1. By contrast, transfer of NO from CM-Poly-SNO-HSA into the cells is faster and more pronounced. The influx mainly takes place by cell-surface protein disulfide isomerase. The considerable NO inflow results in apoptotic cell death by ROS induction and caspase-3 activation. Thus, increasing the number of SNO groups on HSA does not simply intensify the cellular responses to the product but can also result in very different effects.  相似文献

Acid-catalyzed nitrosation and production of potentially carcinogenic nitrosative species is focused at the gastroesophageal junction, where salivary nitrite, derived from dietary nitrate, encounters the gastric juice. Ascorbic acid provides protection by converting nitrosative species to nitric oxide (NO). However, NO may diffuse into adjacent lipid, where it reacts with O2 to re-form nitrosative species and N-nitrosocompounds (NOC). In this way, ascorbic acid promotes acid nitrosation. Using a novel benchtop model representing the gastroesophageal junction, this study aimed to clarify the action of a range of water-soluble antioxidants on the nitrosative mechanisms in the presence or absence of lipids. Caffeic, ferulic, gallic, or chlorogenic and ascorbic acids were added individually to simulated gastric juice containing secondary amines, with or without lipid. NO and O2 levels were monitored by electrochemical detection. NOC were measured in both aqueous and lipid phases by gas chromatography–tandem mass spectrometry. In the absence of lipids, all antioxidants tested inhibited nitrosation, ranging from 35.9 ± 7.4% with gallic acid to 93 ± 0.6% with ferulic acid. In the presence of lipids, the impact of each antioxidant on nitrosation was inversely correlated with the levels of NO they generated (R2 = 0.95, p < 0.01): gallic, chlorogenic, and ascorbic acid promoted nitrosation, whereas ferulic and caffeic acids markedly inhibited nitrosation.  相似文献

A series of fluorinated-phenylsulfamates have been prepared by sulfamoylation of the corresponding phenols and the inhibition of four physiologically relevant carbonic anhydrase (CA, EC 4.2.1.1) isozymes, the cytosolic CA I and II (off-targets), and the transmembrane, tumor-associated CA IX and XII is investigated. Unlike the lead molecule (phenylsulfamate), a very potent CA I and II inhibitor and a modest CA IX/XII inhibitor, the fluorinated sulfamates were stronger inhibitors of CA IX (KIs of 2.8–47 nM) and CA XII (KIs of 1.9–35 nM) than of CA I (KIs of 53–415 nM) and CA II (KIs of 20–113 nM). Some of these compounds were selective CA IX over CA II inhibitors, with selectivity ratios in the range of 11.4–12.1, making them interesting candidates for targeting hypoxic tumors overexpressing CA IX and/or XII.  相似文献

The asymmetric hydrolysis of dimethyl 3-phenylglutarate (1) by different immobilized preparations of a phospholipase A1 (Lecitase Ultra (LECI)) at pH 7 and 25 °C has been studied. Agarose beads coated with octyl, cyanogen bromide (CNBr), polyethylenimine (PEI) or glyoxyl groups were used as supports for the immobilization of LECI. The different derivatives behaved very differently in terms of activity, discrimination between 1 and methyl 3-phenylglutarate (2) resulting from the hydrolysis of 1, enantioselectivity (in the hydrolysis of 1 to produce R or S-2) and enantiospecificity in the hydrolysis of R-2 and S-2. Using 1 mM of 1, CNBr-LECI showed the highest activity (13 × 10−3 μmol/min mg protein) while octyl-LECI was about 20 times less active. All the enzyme preparations mainly produced (S)-2, but with different enantioselectivity. CNBr-Lecitase was the most enantioselective, producing the S-2 10 fold more rapidly than the R-2, while octyl-Lecitase gave only half of that difference.LECI adsorbed on octyl-agarose allowed to get a yield up to 99% of S-2 (ee was 66%). The reaction stopped in the monoester and no isomer of this compound was further hydrolyzed by the enzyme. However, when the reaction was catalyzed by the other immobilized LECI preparations, the enzyme was able to hydrolyze mainly the minority isomer, permitting to improve the ee of the remaining S-2. The best results were obtained using CNBr-LECI, which gave (S)-methyl-3-phenylglutarate with a yield of 80% and an ee exceeding 99%.  相似文献

A comparative study of hypotensive effects of binuclear forms of dinitrosyl iron complexes (DNICs) with glutathione, S-nitrosoglutathione (GS-NO) and sodium nitrite (NaNO2) on rats has been carried out. The latter appeared to be the least efficient, viz., mean arterial pressure (MAP) decreased by 10 and 30 mm Hg at 25 and 100 μmoles/kg of NaNO2. In contrast, DNIC and GS-NO produced an appreciable hypotensive effect when used at much lower concentrations. GS-NO reduced MAP to the same extent, viz., to 90 mm Hg, on a hundredfold dose scale (from 0.4 up to 50 μmoles/kg) with subsequent restoration of MAP within the next 6–15 min. A similar effect was observed for DNIC except that the amplitude of the MAP drop was lower and the duration of hypotension was essentially greater. DNIC with glutathione were selected as a basic material for pilot-scale production of a hypotensive drug (commercial name Oxacom®). Preliminary pharmacological testing of Oxacom did not establish any adverse or deleterious side effects.Clinical trials of Oxacom® were performed on 14 healthy male volunteers in whom single intravenous infusion of the drug (5 mg/kg or 0.2 μmoles/kg of DNIC, respectively) evoked a characteristic response manifested as a 3–4 min drop by 24–27 mm Hg of both diastolic and systolic AP with its subsequent slow restoration within the next 8–10 h. The heart rate was quickly normalized after an initial increase. Cardiac output was unchanged despite reduced cardiac filling. A comprehensive analysis of clinical and biochemical data failed to establish any significant pathological changes in these parameters. The data obtained suggest that Oxacom® can be recommended for the second phase of clinical trials.  相似文献

We review an extensive body of single-crystal high-frequency electron paramagnetic resonance (HFEPR) data in order to determine the transverse spin Hamiltonian parameters that control the tunneling of the direction of magnetization in a variety of integer and half-integer-spin single-molecule magnets (SMMs). The SMMs studied are members of the following families: S = 9/2 [Mn4O3Cl]6+; S = 5 [Mn3NiO4]6+; S = 6 [Mn3ZnO4]6+; and S = 4 [Ni4(OR)4]4+. HFEPR spectra for the half-integer S = 9/2 Mn4 complexes that have C3 symmetry do not provide measurable evidence for transverse spin Hamiltonian terms. This finding is consistent with the relatively large coercive field seen in the magnetization hysteresis loops for these complexes. On the other hand, a low symmetry S = 9/2 complex exhibits a much faster rate of ground-state magnetization tunneling, in agreement with HFEPR spectra for a powder sample that gives a rhombic zero-field splitting (ZFS) parameter of E = 0.140 cm?1. The S = 5 Mn3Ni systems exhibit magnetization tunneling that is much faster than seen for the high-symmetry S = 9/2 Mn4 complexes. This can be attributed to their integer-spin ground states. Like the C3 symmetry Mn4 SMMs, the HFEPR spectra for high-symmetry Mn3Ni complexes do not provide measurable evidence for transverse ZFS terms. However, the spectra exhibit broad peaks, suggesting distributions in the local molecular environments brought about by disordered solvate molecules. This disorder likely explains the fast tunneling in the high-symmetry S = 5 Mn3Ni systems, though one cannot rule out fourth- (and higher-) order interactions that cannot be detected by HFEPR due to the broad resonances. The one S = 6 Mn3Zn complex shows an even faster rate of tunneling compared to the isostructural S = 5 Mn3Ni complex. Finally, the S = 4 [Ni(hmp)(dmb)Cl]4 complex provides unique insights into the origin of fourth- (and higher-) order interactions found for many SMMs on the basis of analysis using a giant spin Hamiltonian (GSH) approximation. We conclude that the fourth-order anisotropy found for the S = 4 ground state of [Ni(hmp)(dmb)Cl]4 originates from the second-order ZFS interactions associated with the individual NiII ions, but only as a result of higher-order processes that occur via S-mixing between the ground state and higher-lying (S < 4) spin-multiplets. The S-mixing is relatively strong in this system because of comparable exchange and anisotropy energy scales. The relatively fast tunneling is a direct consequence of this S-mixing, as opposed to any intrinsic fourth-order (spin–orbit) anisotropy associated with NiII.  相似文献

Cibacron Blue F3GA (CB) was covalently attached onto the bacterial cellulose (BC) nanofibers for human serum albumin (HSA) depletion from human serum. The BC nanofibers were produced by Acetobacter xylinum in the Hestrin–Schramm medium in a static condition for 14 days. The CB content of the BC nanofibers was 178 μmol/g. The specific surface area of the BC nanofibers was determined to be 914 m2/g. HSA adsorption experiments were performed by stirred-batch adsorption. The non-specific adsorption of HSA on the BC nanofibers was very low (1.4 mg/g polymer). CB attachment onto the BC nanofibers significantly increased the HSA adsorption (1800 mg/g). The maximum HSA adsorption was observed at pH 5.0. The HSA adsorption capacity decreased drastically with an increase of the aqueous phase concentration of sodium chloride. The elution studies were performed by adding 1 M NaCl to the HSA solutions in which adsorption equilibria had been reached. The elution results demonstrated that the binding of HSA to the adsorbent was reversible. The depletion efficiencies for HSA were above 96.5% for all studied concentrations. Proteins in the serum and eluted portion were analyzed by SDS-PAGE for testing the efficiency of HSA depletion from human serum. Eluted proteins include mainly HSA.  相似文献

The interaction between paracetamol and human serum albumin (HSA) under physiological conditions has been investigated by fluorescence, circular dichroism (CD) and docking. Fluorescence data revealed that the fluorescence quenching of HSA by paracetamol was the result of the formed complex of HSA–paracetamol, and the binding constant (Ka) and binding number obtained is 1.3 × 104 at 298 K and 2, respectively for the primary binding site. Circular dichorism spectra showed the induced conformational changes in HSA by the binding of paracetamol. Moreover, protein–ligand docking study indicated that paracetamols (two paracetamols bind to HSA) bind to residues located in the subdomain IIIA.  相似文献

AimsThe effects of human serum albumin (HSA) complexed with various free fatty acids (FFAs) on ß-cells have not been studied in detail. In this study, we examined the effects of HSA and its mutants on FFA-induced cell viability changes and insulin secretion from the hamster pancreatic insulinoma cell line, HIT-TI5.Main methodsCells were exposed to different FFAs in the presence of HSA or its mutants and/or bovine serum albumin (BSA) for 24 h. Cell viability, apoptosis, insulin secretion, and unbound FFA (FFAu) levels were determined.Key findingsIn the presence of 0.1 mM HSA, palmitate and stearate induced significant cell death at 0.1 mM or higher, whereas myristate, palmitoleate, oleate, elaidate, linoleate, linoelaidate, and conjugated linoleate showed minimal changes on cell viability. Furthermore, oleate and linoleate were clearly cytoprotective against palmitate-induced cell death. The apoptosis inhibitors, cyclosporin A (csA) and the caspase inhibitor ZVAD-FMK, did not completely prevent FFA-induced cell death, although ZVAD-FMK blocked apoptosis with no differences in the presence of either HSA or BSA. In addition, insulin secretion from the cells was significantly reduced in the presence of HSA/oleate complexes. We also found differential effects of HSA mutants complexed with FFAs on cell viability.SignificanceIn summary, our results showed that saturated FFAs induced more cell death than unsaturated FFAs. Furthermore, modified HSA/FFA interactions caused by mutations of key amino acids involved in the binding of FFA to HSA resulted in changes in cell viability, suggesting a possible role of HSA polymorphism on FFA-induced changes in cellular functions.  相似文献

In the present study, interactions of Au(III) and Ga(III) ions on human serum albumin (HSA) were studied comparatively via spectroscopic and thermal analysis methods: UV–vis absorbance spectroscopy, fluorescence spectroscopy, Fourier transform infrared (FT-IR) spectroscopy and isothermal titration calorimetry (ITC). The potential antitumor effects of these ions were studied on MCF-7 cells via Alamar blue assay. It was found that both Au(III) and Ga(III) ions can interact with HSA, however; Au(III) ions interact with HSA more favorably and with a higher affinity. FT-IR second derivative analysis results demonstrated that, high concentrations of both metal ions led to a considerable decrease in the α-helix content of HSA; while Au(III) led to around 5% of decrease in the α-helix content at 200 μM, it was around 1% for Ga(III) at the same concentration. Calorimetric analysis gave the binding kinetics of metal–HSA interactions; while the binding affinity (Ka) of Au(III)–HSA binding was around 3.87 × 105 M−1, it was around 9.68 × 103 M−1 for Ga(III)–HSA binding. Spectroscopy studies overall suggest that both metal ions have significant effects on the chemical structure of HSA, including the secondary structure alterations. Antitumor activity studies on MCF7 tumor cell line with both metal ions revealed that, Au(III) ions have a higher antiproliferative activity compared to Ga(III) ions.  相似文献

Secoisolariciresinol diglucosides (SDGs) (S,S)-SDG-1 (major isomer in flaxseed) and (R,R)-SDG-2 (minor isomer in flaxseed) were synthesized from vanillin via secoisolariciresinol (6) and glucosyl donor 7 through a concise route that involved chromatographic separation of diastereomeric diglucoside derivatives (S,S)-8 and (R,R)-9. Synthetic (S,S)-SDG-1 and (R,R)-SDG-2 exhibited potent antioxidant properties (EC50 = 292.17 ± 27.71 μM and 331.94 ± 21.21 μM, respectively), which compared well with that of natural (S,S)-SDG-1 (EC50 = 275.24 ± 13.15 μM). These values are significantly lower than those of ascorbic acid (EC50 = 1129.32 ± 88.79 μM) and α-tocopherol (EC50 = 944.62 ± 148.00 μM). Compounds (S,S)-SDG-1 and (R,R)-SDG-2 also demonstrated powerful scavenging activities against hydroxyl [natural (S,S)-SDG-1: 3.68 ± 0.27; synthetic (S,S)-SDG-1: 2.09 ± 0.16; synthetic (R,R)-SDG-2: 1.96 ± 0.27], peroxyl [natural (S,S)-SDG-1: 2.55 ± 0.11; synthetic (S,S)-SDG-1: 2.20 ± 0.10; synthetic (R,R)-SDG-2: 3.03 ± 0.04] and DPPH [natural (S,S)-SDG-1: EC50 = 83.94 ± 2.80 μM; synthetic (S,S)-SDG-1: EC50 = 157.54 ± 21.30 μM; synthetic (R,R)-SDG-2: EC50 = 123.63 ± 8.67 μM] radicals. These results confirm previous studies with naturally occurring (S,S)-SDG-1 and establish both (S,S)-SDG-1 and (R,R)-SDG-2 as potent antioxidants and free radical scavengers for potential in vivo use.  相似文献

A series of novel sulfamides incorporating the dopamine scaffold were synthesized. Reaction of amines and tert-butyl-alcohol/benzyl alcohol in the presence of chlorosulfonyl isocyanate (CSI) afforded sulfamoyl carbamates, which were converted to the title compounds by treatment with trifluoroacetic acid or by palladium-catalyzed hydrogenolysis. Inhibition of six α-carbonic anhydrases (CAs, EC 4.2.1.1), that is, CA I, CA II, CA VA, CA IX, CA XII and CA XIV, and two β-CAs from Candida glabrata (CgCA) and Mycobacterium tuberculosis (Rv3588) with these sulfamides was investigated. All CA isozymes were inhibited in the low micromolar to nanomolar range by the dopamine sulfamide analogues. Kis were in the range of 0.061–1.822 μM for CA I, 1.47–2.94 nM for CA II, 2.25–3.34 μM for CA VA, 0.041–0.37 μM for CA IX, 0.021–1.52 μM for CA XII, 0.007–0.219 μM for CA XIV, 0.35–5.31 μM for CgCA and 0.465–4.29 μM for Rv3588. The synthesized sulfamides may lead to inhibitors targeting medicinally relevant CA isoforms with potential applications as antiepileptic, antiobesity antitumor agents or anti-infective.  相似文献

In vitro studies suggested that nitrite may play a cytoprotective role in inflammation. The aim of the present clinical study was to investigate the relationship between the NO metabolites nitrite and nitrate and the biomarkers of oxidative stress 3-nitrotyrosine (3-NT) and 15(S)-iso-PGF in patients suffering from chronic inflammatory rheumatic diseases. In morning urine from 28 patients with different chronic inflammatory rheumatic diseases (23–82 years of age) and from 41 healthy persons of both genders, nitrite and nitrate were quantitated by GC-MS, and 3-NT and 15(S)-iso-PGF by GC-MS/MS. Mean creatinine-corrected urinary excretion rates of nitrite (1.1 versus 0.19 μmol/mmol, P = 0.00012) and 3-NT (1.2 versus 0.39 nmol/mmol, P = 0.01629), but not of nitrate (105 versus 106 μmol/mmol), were significantly elevated in rheumatism as compared to health. Urinary excretion rate of 15(S)-iso-PGF did not differ between patients and healthy subjects (65 versus 69 pmol/mmol creatinine, P = 0.48). In rheumatism, urinary 3-NT correlated closely with nitrite (R = 0.788, P < 0.0001) and moderately with nitrate (R = 0.45, P < 0.016), but did not correlate with 15(S)-iso-PGF (R = ?0.083, P = 0.68). In healthy persons there was no correlation between urinary 3-NT and nitrite or nitrate. Our study suggests that urinary nitrite may represent a novel specific biomarker of nitrative stress in chronic inflammatory rheumatic disease. In another eight patients with chronic inflammatory rheumatic diseases we found higher nitrite concentrations in synovial fluid as compared to serum (1.30 versus 0.35 μM). We hypothesize that in chronic inflammatory rheumatic diseases nitrite concentration is elevated in the inflamed joint and contributes to the inactivation of myeloperoxidase-catalyzed production of hypochloric acid by forming nitryl chloride which eventually nitrates tyrosine to form 3-NT.  相似文献

A novel method for real-time investigating the binding interaction between human serum albumin (HSA) and salicylic acid with capacitive sensing technique was successfully proposed. HSA was immobilized on the surface of a gold electrode modified with an insulating poly (o-phenylenediamine) (o-PD) film and colloid Au nanoparticles layers. The bioactivity of HSA was remained and major binding sites were available because of the excellent biocompatibility of gold nanoparticles. The capacitance and interfacial electron resistance of the sensor were altered, owing to the binding of HSA to salicylic acid. The time courses of the capacitance change were acquired with capacitive sensing technique during the binding process. Based on the capacitance response curves with time, the response model for the binding was derived in theory and the corresponding regression parameters were determined by fitting the real-time experimental data to the model. The binding and the dissociation rate constants (k1 and k 1) were estimated to be 54.8 (mol l 1) 1 s 1 and 2.9 × 10 3 s 1, respectively. And the binding equilibrium constant (Ka) was calculated to be 1.89 × 104 (mol l 1) 1.  相似文献

Megastigmane glycosides (15) together with seven (612) related known compounds were isolated from the whole plants of Gynostemma pentaphyllum. The structures were elucidated by means of spectroscopic methods, including 2D NMR, HR-ESIMS, and circular dichroism (CD), as well as chemical transformations to be (3R, 4R, 5S, 6S, 7E)-3,4,6-trihydroxymegastigmane-7-en-9-one-3-O-β-d-glucopyranoside (gynostemoside A, 1), (3S, 4S, 5R, 6R, 7E, 9R)-3,4,6,9-tetrahydroxymegastigmane-7-en-3-O-β-d-glucopyranoside (gynostemoside B, 2), (3S, 4S, 5S, 6S, 7E, 9R)-3,4,9-trihydroxymegastigmane-7-en-9-O-β-d-glucopyranoside (gynostemoside C, 3), (3S, 4S, 5S, 6S, 7E, 9R)-3,4,9-trihydroxymegastigmane-7-en-3-O-β-d-glucopyranoside (gynostemoside D, 4), and (3S, 4S, 5S, 6S, 7E, 9R)-3,4,9-trihydroxymegastigmane-7-en-4-O-β-d-glucopyranoside (gynostemoside E, 5), respectively.  相似文献

With Candida antarctica lipase B (CALB)-catalyzed alcoholysis of (R,S)-naproxenyl 1,2,4-triazolide at the optimal conditions (i.e. anhydrous MTBE as the solvent, and methanol as the acyl acceptor at 45 °C) as the model system, the enzyme enantioselectivity in terms of VR/VS = 105.8 and specific activity for the fast-reacting (R)-azolide VR/(Et) = 0.979 mmol/(h g) were greatly improved in comparison with VR/VS = 8.0 and VR/(Et) = 0.113 mmol/(h g) of using (R,S)-naproxenyl 2,2,2-trifluoroethyl ester as the substrate. The resolution strategy was successfully extended to other (R,S)-profenyl 1,2,4-triazolides and lipases from Candida rugosa (Lipase MY) and Carica papaya (CPL) having opposite enantioselectivity to CALB. Moreover, the kinetic constants were estimated, compared with those obtained via hydrolysis, and employed for modeling time-course conversions of (R,S)-naproxenyl 1,2,4-triazolide in anhydrous MTBE. The advantages of easy substrate preparation, high enzyme reactivity and enantioselectivity, as well as easy product separation from the remaining substrate via reactive extraction demonstrate merits of using (R,S)-azolides but not the corresponding esters for the alcoholytic resolution.  相似文献

Fluorescent sulfonamide carbonic anhydrase (CA, EC 4.2.1.1) inhibitors (CAIs) were essential for demonstrating the role played by the tumor-associated isoform CA IX in acidification of tumors, cancer progression towards metastasis and for the development of imaging and therapeutic strategies for the management of hypoxic tumors which overexpress CA IX. However, the presently available such compounds are poorly water soluble which limits their use. Here we report new fluorescent sulfonamides 7, 8 and 10 with increased water solubility. The new derivatives showed poor hCA I inhibitory properties, but were effective inhibitors against the hCA II (KIs of 366–127 nM), CA IX (KIs of 8.1–36.9 nM), CA XII (KIs of 4.1–20.5 nM) and CA XIV (KIs of 12.8–53.6 nM). A high resolution X-ray crystal structure of one of these compounds bound to hCA II revealed the factors associated with the good inhibitory properties. Furthermore, this compound showed a three-fold increase of water solubility compared to a similar derivative devoid of the triazole moiety, making it an interesting candidate for ex vivo/in vivo studies.  相似文献

Despite their large secretome and wide applications in bioprocesses, fungi remain underexplored in metal nanoparticles (MNP) biosynthesis. Previous studies have shown that cell surface proteins of Rhizopus oryzae play a crucial role in biomineralization of Au(III) to produce gold nanoparticles (AuNPs). Therefore, it is hypothesized that purified cell surface protein may produce in vitro AuNPs with narrow size distribution for biomedical and biocatalytic applications. However, different protein extraction methods might affect protein stability and the AuNP biosynthesis process. Herein, we have explored the extraction of cell surface proteins from R. oryzae using common detergents and reducing agent (sodium dodecyl sulfate (SDS) Triton X-100, and 1,4-dithiothreitol (DTT)) and their effect on the size and shape of the biosynthetic AuNPs. The surface proteins extracted with reducing agent (DTT) and non-ionic detergent (Triton X-100) produce spherical AuNPs with a mean particle size of 16 ± 7 nm, and 19 ± 4 nm, respectively, while the AuNPs produced by the surface protein extracted by ionic detergent (SDS) are flower-like AuNPs with broader size distribution of 43 ± 19 nm. This synthetic approach does not require use of any harsh chemicals, multistep preparation and separation process, favouring environmental sustainability. The biosynthetic AuNPs thus formed, are stable in different physiological buffers and hemocompatible, making them suitable for biomedical applications.  相似文献

In this communication we report a serendipitously discovered hybrid molecule 1, combining fragment of 3 (an in vivo active antileishmanial molecule) with H2S donor moiety (known for bimodal behavior of cytoprotection and apoptosis), as antileishmanial agent. Compound 1 suppresses 99.82% parasitemia of L. donovani infected macrophages at 12.5 μg/ml without even deforming them (CC50 > 100 μg/ml). This compound appears cytotoxic for intracellular amastigotes while cytoprotective to host macrophages. The concept can be utilized to develop high therapeutic index NCE (New Chemical Entities) for other macrophage mediated diseases like tuberculosis and cancer.  相似文献
