To investigate the functional involvement of carotenoid in the insect circadian rhythm, we observed the effect of carotenoid depletion on the hatching patterns of the silkworm under several light conditions. When eggs were transferred from continuous light (LL) to continuous darkness (DD), an overt hatching rhythm was initiated. The first hatching peak, which was observed at 13.2 h after the transfer in the carotenoid-depleted silkworm, was reduced remarkably in both control groups (carotenoid-rich and carotenoid-depleted but with lutein supplementation), though subsequent peaks occurred with similar timing. Under LD 4 : 20, LD 12 : 12 and LD 20 :4, hatching patterns depended on the dietary carotenoid and the light intensity of the photophase. At a low light intensity, carotenoid depletion suppressed hatching as a masking effect just after light-on. Under LD 4 : 20 at a low light illumination, hatchings in the carotenoid-depleted silkworm were observed during scotophase, but few larvae hatched during scotophase in the control groups. Our findings suggest that carotenoid is not indispensable for the photoreception, formation and entrainment of the circadian hatching rhythm, but that a carotenoid-dependent process that is induced by light-off and damps out in an hour-glass mode, suppresses the hatching during darkness without affecting the circadian oscillation. The possibility that this carotenoid-dependent process is involved in the photoperiodic induction in the silkworm was discussed.  相似文献

Giant freshwater prawns, Macrobrachium rosenbergii de Man, were reared under three different lighting conditions: continuous darkness (DD), 12 hr of light and 12 hr of darkness (LD 12:12) and continuous light (LL). After one month, the prawns were sacrificed and optic lobes isolated from the eyestalks were determined for N-acetyltransferase (NAT) activities and melatonin concentrations. Gonads were weighed and examined under light microscopy. The optic lobes from LL prawns contained significantly higher activities of NAT than those from LD 12:12 prawns. The melatonin concentrations and size and histological features of the gonads from the three groups of prawns did not differ. The results indicate that continuous light increases NAT activities in the optic lobe of M. rosenbergii but has no drastic effect on gonadal growth.  相似文献

Galleria mellonella larvae reared in a light:dark (LD) 12:12 regime terminate feeding and pupate 1 day earlier than insects kept in constant darkness. Rearing conditions have no influence on the body weight attained at pupation. In both rearing conditions body weights attained by females were greater on the average by 50% than the body weights of males.In LD 12:12 all distinctive changes in the juvenile hormone titre and juvenile hormone esterase activity occurred 1 day earlier than in constant darkness. The peak value of juvenile hormone esterase activity was by 22% higher in insects reared under light conditions as compared with animals kept in constant darkness.Last-instar G. mellonella larvae reared in constant darkness were more sensitive to cooling and juvenile hormone analogue application. The chilling-induced elevation of juvenile hormone titre was also higher in constant darkness. The chilling-induced elevation of the brain allatotropic activity was not influenced by rearing conditions (brain activity measured as the number of extra-larval moults produced by hosts implanted with chilled brains). However, rearing in an LD 12:12 regime decreases the host sensitivity to implanted brains.  相似文献

The wrasse,Suezichthys gracilis, is a diurnal fish which buries itself in sand during the night-time. The present paper deals with the locomotor activity rhythms ofS. gracilis, examined by using an actograph with infra-red photo-electric switches in a dark room. The fish were kept in eight experimental tanks (each 30l in capacity), with three different bottom conditions: sand (grain size about 1 mm in diameter and 5 cm deep); 1 or 2 stones (about 10cm in diameter) without sand; and transparent acrylic pellets (2 × 2 × 3 mm in size, 5 cm deep). The light intensities were 550–700 lux just above the water surface, decreasing to 21.3% under the acrylic pellets at a water depth of 20cm. The water temperatures were kept at 22.0–25.0°C during the experiments for 7 to 14 days. In the aquarium with bottom sand, diel activity rhythms ofS. gracilis were mostly synchronized to LD (LD12:12; 06:00–18:00 light, 18:00–06:00 dark), free-running activity rhythms continued distinctly under LL (constant illumination), and locomotor activity was greatly suppressed, with disappearance of the activity rhythm, under DD (constant darkness). In the aquarium without sand, locomotor activity ofS. gracilis could be summarized as follows. The fish moved throughout almost the entire period under LD, though more frequent movements were observed in light conditions than in dark ones. Under LL they showed continuous locomotor activity during the experiment, with no obvious periodicity. Under DD the activity of the species was somewhat suppressed, but irregular movement or indistinct periodicity was observed. In the aquarium with transparent acrylic pellets, locomotor activity under LD and DD, respectively, bore a close resemblance to activity patterns under the same light conditions with sand, whilst activity under LL was identical to that under LL without sand. Accordingly, it seems that maintenance of normal activity rhythms in the wrasse was due not only to the darkness, but also to the presence of bottom sand. It therefore seems that the biological clock inS. gracilis is not related to locomotor activity, but to burying behavior.  相似文献

Abstract. The effects of thermoperiods on diapause induction in continuous darkness or under a 12 : 12 h LD photoperiod were investigated in the cabbage beetle, Colaphellus bowringi Baly, a typical short‐day species. The diapause response curves both at different constant temperatures and at the thermocycle of format CT x: (24 ? x) h (16 : 28 °C) under continuously dark rearing conditions showed that the incidence of diapause depended mainly on whether or not the mean temperature was ≤20 °C or >20 °C. If the mean temperature was ≤20 °C, all individuals entered diapause; if >20 °C, the incidence of diapause declined gradually with increasing mean temperatures. The thermocycle (CT 12 : 12 h) with a series of different cryophases (8–22 °C) and thermophases (24–32 °C) under continuous darkness demonstrated a cryophase response threshold temperature of approximately 19 °C and a thermophase response threshold temperature of approximately 31 °C. Thermoperiodic amplitude (temperature difference between cryophase and thermophase) was shown to have a significant influence on diapause induction at the mean temperatures of 22, 23 and 24 °C, but not at ≥25 °C. Thermoperiodic responses under LD 12 : 12 h clearly showed that the incidence of diapause was influenced strongly by the photophase temperature. The thermoperiod under LD 12 : 12 h induced a much lower incidence of diapause than the thermoperiod with the same temperature in continuous darkness. The ecological significance of thermoperiodic induction of diapause in this species is discussed.  相似文献

The optimal temperature and illumination photoperiod requirements for the phototrophic growth of a novel microplantlet suspension culture derived from the macrophytic marine red alga Agardhiella subulata were determined. The optimal growth temperature was 24 degrees C. The effects of illumination light-dark (LD) photoperiod (hour of light:hours of darkness within a 24 h cycle) on biomass production was studied within a bubble-column photobioreactor. The 4.5 cm diameter photobioreactor was maintained at near-saturation conditions with respect to light flux (38 mciromol photons m(-2) s(-1)), nutrient medium delivery (20% nutrient replacement per day), and CO(2) delivery (0.35 mmol CO(2) L(-1) h(-1)) so that the cumulative effects of photodamage on the cell density versus time curve at photoperiods approaching continuous light could be observed. Biomass production was maximized at 16:8 LD, where biomass densities exceeding 3.6 g dry cell mass L(-1) were achieved after 60 days in culture. Biomass production was proportional to photoperiod at low fractional photoperiods (< or =10:14 LD), but high fractional photoperiods approaching continuous light (> or = 20:4 LD) shut down biomass production. Biomass production versus time profiles under resource-saturated cultivation conditions were adequately described by a cumulative photodamage growth model, which coupled reversible photodamage processes to the specific growth rate. Under light-saturated growth conditions, the rate constant for photodamage was kd = 1.17 +/- 0.28 day(-1) (+/-1.0 SE), and the rate constant for photodamage repair was kr = 5.12 +/- 0.95 day(-1) (+/-1.0 SE) at 24 degrees C.  相似文献

Although the anterior chamber of the eye expresses immune privilege, some ocular tumors succumb to immune rejection. Previous studies demonstrated that adenovirus-induced tumors, adenovirus type 5 early region 1 (Ad5E1), underwent immune rejection following transplantation into the anterior chamber of syngeneic mice. Intraocular tumor rejection required CD4(+) T cells, but did not require the following: 1) CD8(+) T cells, 2) B cells, 3) TNF, 4) perforin, 5) Fas ligand, or 6) NK cells. This study demonstrates that CD4(+) T cell-dependent tumor rejection does not occur in IFN-gamma-deficient mice. Ad5E1 tumor cells expressed DR5 receptor for TRAIL and were susceptible to TRAIL-induced apoptosis. Although IFN-gamma did not directly induce apoptosis of the tumor cells, it rendered them 3-fold more susceptible to TRAIL-induced apoptosis. Both CD4(+) T cells and corneal endothelial cells expressed TRAIL and induced apoptosis of Ad5E1 tumor cells. The results suggest that Ad5E1 tumor rejection occurs via TRAIL-induced apoptosis as follows: 1) tumor cells express TRAIL-R2 and are susceptible to TRAIL-induced apoptosis, 2) IFN-gamma enhances TRAIL expression on CD4(+) T cells and ocular cells, 3) IFN-gamma enhances tumor cell susceptibility to TRAIL-induced apoptosis, 4) apoptotic tumor cells are found in the eyes of rejector mice, but not in the eyes of IFN-gamma knockout mice that fail to reject intraocular tumors, 5) CD4(+) T cells and corneal endothelial cells express TRAIL and induce apoptosis of tumor cells, and 6) apoptosis induced by either CD4(+) T cells or corneal cells can be blocked with anti-TRAIL Ab.  相似文献

Light plays a key role in the development of biological rhythms in fish. Previous research on Senegal sole has revealed that both spawning rhythms and larval development are strongly influenced by lighting conditions. However, hatching rhythms and the effect of light during incubation are as yet unexplored. Therefore, the aim of this study was to investigate the impact of the light spectrum and photoperiod on Solea senegalensis eggs and larvae until day 7 post hatching (dph). To this end, eggs were collected immediately after spawning during the night and exposed to continuous light (LL), continuous darkness (DD), or light-dark (LD) 12L:12D cycles of white light (LD(W)), blue light (LD(B); λ(peak)?=?463?nm), or red light (LD(R); λ(peak)?=?685?nm). Eggs exposed to LD(B) had the highest hatching rate (94.5%?±?1.9%), whereas LD(R) and DD showed the lowest hatching rate (54.4%?±?3.9% and 48.4%?±?4.2%, respectively). Under LD conditions, the hatching rhythm peaked by the end of the dark phase, but was advanced in LD(B) (zeitgeber time 8 [ZT8]; ZT0 representing the onset of darkness) in relation to LD(W) and LD(R) (ZT11). Under DD conditions, the same rhythm persisted, although with lower amplitude, whereas under LL the hatching rhythm split into two peaks (ZT8 and ZT13). From dph 4 onwards, larvae under LD(B) showed the best growth and quickest development (advanced eye pigmentation, mouth opening, and pectoral fins), whereas larvae under LD(R) and DD had the poorest performance. These results reveal that developmental rhythms at the egg stage are tightly controlled by light characteristics, underlining the importance of reproducing their natural underwater photoenvironment (LD cycles of blue wavelengths) during incubation and early larvae development of fish.  相似文献

ABSTRACT. Eclosion in Lucilia cuprina (Wiedemann) occurs near dawn. The rhythm of eclosion persists in both darkness and constant light of high intensity (490μW cm-2) with a period close to 24h. The sensitivity to light of the circadian clock controlling eclosion varies greatly according to the stage of the life cycle. During larval life the free running rhythm in darkness can be phase shifted by light pulses of 100μW cm-2 intensity, with the transition from a Type 1 phase response curve to a Type 0, occurring with pulses of between 1 and 8h. Extending the last light period of LD to 24 h followed by constant darkness resets the phase of the rhythm by 12h, a transition from constant light to constant darkness initiates rhythmicity in flies made arrhythmic by being reared from eggs collected from adults maintained in constant light. After pupariation, the rhythm is relatively insensitive to light. Rhythmicity is sometimes induced by a transition from constant light to constant darkness, but the phase of the rhythm is not shifted by extending the last light period of LD before entering constant darkness. Repeated LD cycles applied after pupariation initiate and entrain the rhythm.  相似文献

Appearance of myeloid bodies (MB) in the retinal pigment epithelium (RPE) precedes photoreceptor outer segment development in Poecilia reticulata embryos reared under a 12 hrs LD cycle, in constant darkness (DD) and constant light (LL). When first formed, MB are predominantly continuous with rough endoplasmic reticulum (RER). The same is observed in the peripheral growth zone of the developed eye, whereas in differentiated parts, MB are continuous with the smooth endoplasmic reticulum (SER). At onset of photomechanical movements, wavy MB predominate in light-adapted LD embryos, are exclusively present in LL and are located in the RPE processes. SER abounds. Straight MB predominate in dark-adapted LD embryos, are exclusively present in DD and contain electrondense material between lamellae. Diurnal appearance of electrondense material may be coupled with transfer of retinol, mediated by various transport proteins.  相似文献

ABSTRACT. In constant light or constant darkness, Glossina morsitans morsitans Westwood were found to exhibit no obvious rhythms of parturition, but in LD 12:12 most larvae were deposited during the photophase. When a divided photoperiod chamber was used to expose the female's head to LD 12:12 and the abdomen (containing the larvae in utero ) to constant light, parturition coincided with the photophase of the female's head. But, if the abdomen was exposed to LD 12:12 and the female's head received constant light or was painted with black enamel, parturition coincided with the photophase of the larva. When the female's head and abdomen both received LD 12:12 but 11 h out of phase with each other, parturition was equally frequent in each 12h fraction. The results thus suggest that both the mother and larva are capable of entraining the rhythm, and that in a normal pregnancy both participate in establishing the rhythm.  相似文献

1. The effects of raising cockroaches, Leucophaea maderae, in non-24 h light cycles on circadian rhythms in adults were examined. The average period (tau) of freerunning rhythms of locomotor activity of animals exposed to LD 11:11 (T22) during post-embryonic development was significantly shorter (tau = 22.8 +/- 0.47 SD, n = 85) than that of animals raised in LD 12:12 (T24) (tau = 23.7 +/- 0.20 h, n = 142), while animals raised in LD 13:13 (T26) had significantly longer periods (tau = 24.3 +/- 0.21 h, n = 65). Animals raised in constant darkness (DD) had a significantly shorter period (tau = 23.5 +/- 0.21 h, n = 13) than siblings raised in constant light (LL) (tau = 24.0 +/- 0.15 h, n = 10). 2. The differences in tau between animals raised in T22 and T24 were found to be stable in DD for at least 7 months and could not be reversed by exposing animals to LD 12:12 or LD 6:18. 3. Animals raised in either T24 or DD and then exposed as adults to T22 exhibited average freerunning periods that were not different from animals not exposed to T22. 4. Measurement of freerunning periods at different temperatures of animals raised in T22, T24, or T26 showed that the temperature compensation of tau was not affected by the developmental light cycle. These results indicate that the lighting conditions during post-embryonic development can permanently alter the freerunning period of the circadian system in the cockroach, but do not affect its temperature compensation.  相似文献

Larval Typhlotriton spelaeus collected from five caves in Pulaski Co., Missouri, were kept as larvae or induced to transform in darkness or continuous fluorescent illumination. Larvae maintained in darkness for 215 and 279 days had smaller eyes, smaller rod inner and outer segments, and fewer metaphase figures in the genninative zone of the neural retina than comparable larvae maintained in light (258 lux). Except for visual cell size, differences were small and for each characteristic exceptions were observed. One larva kept in light showed early retinal degeneration comparable to that in transformed adults of T. spelaeus. All larvae exhibited optomotor behavior both before and after the experiment. Among animals induced to transform by L-thyroxin and maintained in darkness 111 to 366 days, visual cell and pigment epithelium degeneration was more extensive and more frequent than in animals kept for the same length of time in light (237-298 lux). In darkness the frequency of animals with retinal degeneration increased between 111 and 366 days. In light some animals exhibited pigment epithelium reduction with normal visual cells, and others had free, pigmented cells in the subretinal space. These effects were not comparable to degeneration in darkness. Eyelids covered the eyes of only a few animals in both light and dark treatments. The extent of eyelid encroachment over the eye was greater in darkness than in light. Most animals exhibited optomotor responses after experiments, but responses of animals kept in darkness were impaired in comparison to those of animals kept in light.  相似文献

We investigated circadian ocular rhythms in the Japanese quail, Coturnix coturnix japonica. The birds were placed under light-dark cycles (LD 1212), constant light (LL) and constant darkness (DD), and the retinas were dissected out at four-hour intervals throughout 24 h. Following measurements were performed. (1) Melatonin content in the retina was measured by radioimmunoassay. It was low in light and several folds higher in darkness under LD 1212. The rhythm continued in DD, but disappeared in LL. (2) Mitotic figures in the corneal epithelium were counted. Similar rhythms to the melatonin content were observed in the corneal mitotic rate with a slight phase delay. (3) The retinas were fixed at 4-h intervals and immunostained with anti-bovine rhodopsin serum and anti-chicken iodopsin monoclonal antibodies. The outer segments of photoreceptor cells were stained intensively throughout 24 h in LD 1212, LL and DD. In contrast, the stainability of the locus close to the outer limiting membrane where the Golgi apparatus exists changed diurnally. Scores showing the ratio of cells with positive staining indicated high values from 4 h after the onset of light to the beginning of dark phase under LD 1212. The values were high throughout 24 h in LL and intermediate or low in DD. (4) To investigate the effect of melatonin on the corneal mitotic rate and visual pigments at the Golgi region, melatonin was injected into one eye and saline into the contralateral eye. Melatonin induced a phase advance in the corneal mitotic rate under LD 1212, but did not induce a rhythm under LL. Egg to eclosion development time and survivorship were assayed on four laboratory populations of Drosophila melanogaster that had been reared for over 600 generations in continuous light (LL) and constant temperature. The assays were performed in three environments: continuous light (LL), periodically varying light/dark cycles (LD 12:12 hr), and continuous darkness (DD). Development time in LL was significantly less than that in LD, which, in turn, was significantly less than that in DD, whereas survivorship did not differ significantly among the three treatments. The results indicate that individuals from Drosophila populations routinely maintained in LL do not suffer any deleterious effects of LL treatment on pre-adult fitness. Other studies on these populations have shown that free-running period (t) of the eclosion rhythm in DD is greater than that in LD. Our results are, thus, also consistent with the notion that development time may be a function of the free-running period.  相似文献

The development of the chamber angle was studied in the eyes of heterozygous Pax6(lacZ/+) mutant mice (Nature 387 (1997) 406). Mutations in PAX6 cause aniridia, a condition that is frequently associated with glaucoma, a blinding disease that may be associated with chamber angle defects. Mesenchymal cells were seen in the chamber angle at P1-P5. In wild-type mice, these cells differentiated into typical trabecular meshwork (TM) cells next to Schlemm's canal. In Pax6(lacZ/+) mice, TM cells remained undifferentiated and Schlemm's canal was absent. From P1 to P4, staining for beta-galactosidase and immunoreactivity for Pax6 were observed in chamber angle mesenchyme, but were absent later. Cultured murine TM cells expressed Pax6. The defects in chamber angle and TM differentiation were associated with a wide spectrum of other anterior eye defects, which included various degrees of iris hypoplasia and corneal haze, isolated iridocorneal adhesions and atypical coloboma, and a vascularized cornea in all adult animals. A third of the animals showed Peters' anomaly including corneal opacity and iridocorneal adhesions. The separation of the lens from the cornea was incomplete, and epithelial layers of lens and cornea were continuous. Pax6 activity is directly required for differentiation of the chamber angle. Variations in phenotype of Pax6(lacZ/+) mice appear not to involve direct dominant-negative or dose-dependent effects.  相似文献

The term masking refers to immediate responses to stimuli that override the influence of the circadian timekeeping system on behavior and physiology. Masking by light and darkness plays an important role in shaping an organism's daily pattern of activity. Nocturnal animals generally become more active in response to darkness (positive masking) and less active in response to light (negative masking), and diurnal animals generally have opposite patterns of response. These responses can vary as a function of light intensity as well as time of day. Few studies have directly compared masking in diurnal and nocturnal species, and none have compared rhythms in masking behavior of diurnal and nocturnal species. Here, we assessed masking in nocturnal mice (Mus musculus) and diurnal grass rats (Arvicanthis niloticus). In the first experiment, animals were housed in a 12:12 light-dark (LD) cycle, with dark or light pulses presented at 6 Zeitgeber times (ZTs; with ZT0 = lights on). Light pulses during the dark phase produced negative masking in nocturnal mice but only at ZT14, whereas light pulses resulted in positive masking in diurnal grass rats across the dark phase. In both species, dark pulses had no effect on behavior. In the 2nd experiment, animals were kept in constant darkness or constant light and were presented with light or dark pulses, respectively, at 6 circadian times (CTs). CT0 corresponded to ZT0 of the preceding LD cycle. Rhythms in masking responses to light differed between species; responses were evident at all CTs in grass rats but only at CT14 in mice. Responses to darkness were observed only in mice, in which there was a significant increase in activity at CT 22. In the 3rd experiment, animals were kept on a 3.5:3.5-h LD cycle. Surprisingly, masking was evident only in grass rats. In mice, levels of activity during the light and dark phases of the 7-h cycle did not differ, even though the same animals had responded to discrete photic stimuli in the first 2 experiments. The results of the 3 experiments are discussed in terms of their methodological implications and for the insight they offer into the mechanisms and evolution of diurnality.  相似文献

The effects of raising cockroaches, Leucophaea maderae, in non-24-h light cycles on the response of the circadian system to light was examined. 1. Phase response curves (PRC) were measured for 6-h light pulses for animals raised in LD 11:11 (T22), LD 12:12 (T24), and LD 13:13 (T26). The delay portion of the PRC was found to be significantly reduced in T22 animals (compared to T24 animals) while the advance portion of the PRC was reduced in T26 animals. Compared to T26 animals, phase shifts were more positive at every phase for animals raised in T22. 2. When transferred from constant darkness (DD) to constant light (LL) the freerunning period lengthened significantly less for T22 animals than T24 animals, and in some cases tau in LL was actually shorter than tau in DD in T22 animals. Animals raised in LL were inactive when exposed to LL as adults, and unlike T24 animals, were consistently reset to the beginning of the subjective night (near CT 12) when transferred to DD. 3. Roaches raised in T22 would entrain to LD 6:18, but a few animals exhibited periods of relative coordination indicating that the 24-h light cycle was near the limits of entrainment. These results indicate that the circadian system's responsiveness to light, as well as its freerunning period (Barrett and Page 1989), is dependent on the lighting conditions to which the animals are exposed during development.  相似文献

Carbon dioxide emission (VCO2) has been continuously recorded in three laboratory animal species (Sprague-Dawley rats, Japanese quail, Hartley guinea-pigs) which differ by their nocturnal and diurnal activities. A 100 lux stimulus has been delivered at various time intervals. 2. A regular alternation of 12, 3 or 1.5 hr light (L) and darkness (D) gives VCO2 circadian and ultradian rhythms of 24, 6 or 3 hr periods, respectively, in quail and rats. 3. Such circadian and ultradian LD rhythms are not induced in all guinea-pigs. 4. The amplitudes of the VCO2 responses are greatest at D----L when the animals have a maximum diurnal activity and at L----D when their maximum activity is nocturnal. 5. Interactions between circadian and ultradian rhythms are seen in all LD experiments, as well as in continuous light (LL) or continuous dark (DD). 6. No more well-marked or even inverted VCO2 responses to the light stimuli may occur after several days of exposure to these LD alternations.  相似文献
