Our study describes the basic ecological characteristics of the entomopathogenic nematode Steinernema anatoliense including its response to temperature, moisture, and host range. The effect of temperature and soil moisture on the infection of Galleria mellonella larvae by S. anatoliense was determined. The temperature range for infectivity was greater than that for development. The optimal temperature for infection and development was 25 degrees C. Although S. anatoliense infected the hosts at 10 degrees C, no reproduction occurred at this temperature. This nematode species that was isolated from a cold region of Turkey exhibited warm-adapted temperature characteristics. Optimum water content of the soil for S. anatoliense to infect the host was 10%. 相似文献

The infectivity of entomopathogenic nematode (EPN), Steinernema mushtaqi was tested against legume pod borer, Maruca vitrata, tobacco caterpillar, Spodoptera litura, blue butterfly, Lampides boeticus, red hairy caterpillar, Amsacta moorei, brown bug, Clavigralla gibbosa, mealy bug, Centrococcus somatics, fruit borer, Earias vittella and green bug, Nezara viridula larvae and in vivo mass production of the above-tested species of EPN have been carried out during 2008. S. mushtaqi was found to be more pathogenic to A. moorei, as it brought about 100% mortality within 48 h, than to S. litura, L. boeticus, N. viriduala and E. vittella, as mortality occurred within 72 h; whereas this level of mortality was recorded in C. somatis, C. gibbosa and M. vitrata within 144 h. No mortality was observed in the control treatment. Multiplication of S. mushtaqi and the yield of infective juveniles (IJs) on these insects was the highest (0.94 × 105 IJs/cadaver) from N. viriduala, followed by S. litura (0.76 × 105 IJs/cadaver), L. boeticus as also C. gibbosa (0.31 × 105 IJs/cadaver) and M. vitrata (0.20 × 105 IJs/cadaver). Very poor populations of IJs were found from A. moorei (0.15 × 105 IJs/cadaver) and C. somatics (0.01 × 105 IJs/cadaver). No multiplication of IJs was found from the cadaver of E. vittella. Steinernema scapterisci n. sp., isolated in Uruguay from the mole cricket Scapteriscus vicinus, can be distinguished from other members in the genus by the presence of prominent cheilorhabdions, an elliptically shaped structure associated with the excretory duct, and a double-flapped epitygma in the first-generation female. The spicules of the male are pointed, tapering smoothly to a small terminus, and the shaft (calomus) is long, bearing a sheath. The gubernaculum has a long, upward-bent anterior part. The ratio of head to excretory pore divided by tail length of the third-stage juvenile is greater for S. scapterisci n. sp. than for S. carpocapsae. Steinernema scapterisci n. sp. did not hybridize with S. carpocapsae strain Breton. In laboratory tests, S. scapterisci n. sp. killed 10% or less of non-orthopteran insects, including the wax moth larva, a universal host for other species of Steinernema. 相似文献

The effect of three neurotoxic insecticides, three photosynthetic inhibitor herbicides and three enzymatic inhibitor herbicides on infective juveniles (IJs) of Steinernema feltiae Rioja (native) and ENTONEM® (commercial) strains were evaluated after a 48-h exposure at field tank concentrations and overnight treatment in mQ-water, using Spodoptera littoralis as target. Nematode survival was not affected by acetyl-cholinesterase inhibitors chlorpyrifos and pirimicarb, although chlorpyrifos seriously reduced their virulence. Both nematode strains showed differential sensitivity to cypermethrin, which affects the sodium channels of the nerve membrane, with the ENTONEM® strain being more tolerant than Rioja strain. However, these chemicals showed a strong sublethal effect on the nematode reproductive potential, limiting seriously their possible recycling in the field. Herbicides showed differential toxic effects on nematode survival. The commercial strain was tolerant to enzymatic inhibitor herbicides, whereas tribenuron and chlorsulfuron reduced Rioja strain survival. However, photosynthetic inhibitor herbicides severely affected survival of both nematode strains, with the Rioja strain being more sensitive. Sublethal effects on both nematode strains were observed only after exposition to terbutryn+chlortoluron+triasulfuron, increasing the time to kill insect larvae. The Diaprepes root weevil, Diaprepes abbreviatus, is a pest of vegetables, ornamental plants, sugarcane, and citrus in Florida and the Caribbean. The entomopathogenic nematode, Steinernema riobrave, can reduce larval populations of D. abbreviatus substantially. Efficacy of entomopathogenic nematodes, however, may be affected by culture method and formulation. Using D. abbreviatus as the host, we compared the efficacy of two commercial S. riobrave formulations, a liquid and a waterdispersible granule (WDG), with each other and with in vivo produced S. riobrave. Nematodes in the commercial formulations were produced in vitro through liquid fermentation; the in vivo nematodes were cultured in Galleria mellonella and applied in aqueous suspension. Laboratory experiments measured nematode virulence in plastic cups containing soil and seventh-eighth instar D. abbreviatus. One laboratory experiment was conducted using only fresh nematodes (less than 5 days old); another experiment included WDG nematodes that were stored for 25 days at 10 °C. Two field experiments were conducted in which nematodes were applied either to potted citrus (containing D. abbreviatus larvae) placed beneath mature citrus trees or to soil directly beneath the tree. In the latter experiment, efficacy was determined by measuring mortality of caged D. abbreviatus larvae that were buried beneath the soil surface prior to application. Mortality of D. abbreviatus treated with nematodes ranged from 80-98% and 50-75% in laboratory and field experiments, respectively. Three species of Steinernema, S. carpocapsae, S. feltiae, and S. scapterisci consisting of 12 different strains, were tested for their infectivity towards adults of the litter beetle Alphitobius diaperinus. Of the five most promising nematode strains, LC50 values ranged from 1.5 to 77.0 nematodes per host in the filter paper assays. Assays in poultry litter material revealed LC50 values to be 5.8 and 14.6 nematodes per host for the Mexican S. carpocapsae strain and Pye S. feltiae strain. 相似文献

During a survey of entomopathogenic nematodes (EPNs) in the eastern Black Sea region of Turkey in 2009–2012, a steinernematid species was recorded and isolated using the Galleria-baiting method. The isolate was identified as Steinernema kraussei based on its morphological and molecular properties. The analysis of the ITS rDNA sequence placed the Turkish population of S. kraussei in the “feltiae-kraussei” group in the clade that contains different isolates of the species. This is the first record of S. kraussei from Turkey. The efficacy of S. kraussei was tested on Agrotis segetum (Lepidoptera: Noctuidea) larvae at different densities (100, 300, and 500 infective juveniles (IJs) g−1 dry sand ) in laboratory conditions at 25 °C. The highest mortality (98%) was obtained with 500 IJs g−1 dry sand within 7 d after inoculation. Our results indicate that the new isolate is a highly promising biological control agent against A. segetum, one of the most serious soil pests of agricultural area and fruits worldwide. 相似文献

The application of entomopathogenic nematodes (EPN) is generally done using standard spray application techniques. However, in contrast to chemical pesticides, these biological antagonists must remain viable during and after the application process. For the application of EPN, a good agitation system is indispensable as the nematodes tend to sediment fast in a spray tank without agitation. Three agitation systems, viz. mechanical, pneumatic and hydraulic agitation were tested for their ability to keep Steinernema carpocapsae (Rhabditida: Steinernematidae) suspended in an undamaged way. Hydraulic agitation was tested using a centrifugal and a diaphragm pump. Nematode damage was quantified based on viability and infectivity of the EPN. The ability of the agitation system to keep the nematodes in suspension was examined by comparing the nematode concentration observed in the samples taken at different agitation times. Only the hydraulic agitation using the centrifugal pump damaged the nematodes. After 120 min of recirculation, only 19.3% of the nematodes survived. Infectivity was even reduced to 0%. An additional experiment revealed that the temperature rise, from 21.7 to 45.4°C, was responsible for the observed nematode damage. The concentration measurements showed that the pneumatic agitation was unstable. Agitation during 120 min using the other agitation systems resulted in a significant loss of nematodes at 15 cm above the spray tank bottom. In conclusion, mechanical and hydraulic agitation using a diaphragm pump can be recommended when S. carpocapsae is applied, although attention should be paid to possible nematode loss during application. 相似文献

Abstract The survival of Steinernema masoodi and S. carpocapsae (Rhabditida: Steinernematidae) was investigated after foliar application on pigeonpea and chickpea twigs, respectively, at flowering and fruiting stage. The concentration used was 2500 infective juveniles (IJs)/ml water for both the species. On pigeonpea, the mean number of IJs of S. masoodi found alive were 303.4, 158.4 and 51.6 after 0, 30 and 60 minutes of spray in evening hours whereas 236.1, 44.4 and 6.8 IJs were found alive when sprayed in morning hours, respectively. S. masoodi survival at 30 minutes post-spray in the morning was on par with 60 minutes post-spray in the evening hours. On chickpea, the mean numbers of IJs of S. carpocapsae were 165.4, 65.8, 4 and 0 at 0, 1, 2 and 3 h post-spray in the morning hours whereas in the evening spray, 159.4, 111.8, 83.8 and 11.4 IJs found alive at 0, 1, 2 and 3 h post-spray, respectively. Overall, nematode survival in the evening hours was higher compared to morning spray at a given time. Addition of glycerine and UV retardant improved the survival of nematode. Results indicated that survival rate of IJs decreased fast and viability remained up to 3 h and in evening hours very few nematodes remained alive. Serious attempts are needed to improve the survival of nematodes after foliar spray by adding efficient adjuvant, humectant, antidesiccant and/or UV retardant for the management of aerial insect pests. 相似文献

Subterranean termites are major global pests of wood structures and wood products. Among the most economically important subterranean termite species in the US are Heterotermes aureus, Reticulitermes flavipes, and Coptotermes formosanus. In prior studies, the entomopathogenic nematode, Steinernema riobrave strain 355, exhibited a high level of virulence to H. aureus compared with other nematode species. However, S. riobrave 355 was reported to be poorly or only moderately virulent to R. flavipes and C. formosanus, respectively. We hypothesized that other strains of S. riobrave may possess a high level of virulence to all three termite species. Under laboratory conditions we compared three novel strains of S. riobrave (3-8b, 7-12, and TP) with the 355 strain for virulence to H. aureus, R. flavipes, and C. formosanus workers. H. aureus was very susceptible to all the S. riobrave strains, and termites in all nematode treatments were dead after 4 d. The TP strain of S. riobrave caused greater mortality in R. flavipes and C. formosanus compared to the other nematode strains. Specifically, the TP strain caused 75% and 91% mortality in R. flavipes and C. formosanus, respectively, which was more than 300% and 70% higher than the mortality caused by other strains. Additional studies are warranted to determine the ability of S. riobrave (TP) to control the targeted termite species under field conditions. 相似文献

A survey was done in the summer months along the Alaska Highway, in other parts of British Columbia, in northern Alberta, and in the Yukon Territory for steinernematid and heterorhabditid nematodes occurring in the top 10 cm of soil. Steinernema feltiae and Steinernema spp. were found at 18 and Heterorhabditis megidis at 7 sites of 125 sampled. Most nematodes were found where visible insect infestation occurred and where human influence on the habitat was substantial (e.g., agricultural, forested and bush-hedgerow habitats); none was found in grassland or virgin forests. Heterorhabditis megidis occurred in only the southern, warmer, drier region of British Columbia. In the laboratory some steinernematid isolates and H. megidis killed Galleria mellonella larvae at 13 and 22 C, whereas some isolates of Steinernema killed the larvae at only 13 C. Steinernema spp. from three high altitude sites with low, average July temperatures (13-14 C) are cold-active in that they produced infective juveniles at 13 C and killed G. mellonella at 6 C. 相似文献

A nematode collected from Diaprepes abbreviatus is identified and described as a new species, Steinernema diaprepesi n. sp. The new species is closely related to S. feltiae, S. glaseri, and S. oregonense and can be distinguished from these species by the following characteristics: Males: Spicule averaging 79 (71-90) µm and spicule shape; D% (distance from anterior end to excretory pore/ esophagus length × 100) about 80; the ratio SW (spicule length/anal body width) about 1.8. Females: Vulva with short, double- flapped epiptygma; tail terminus usually with 5 papillae-like structures. Infective juveniles: Body averaging 1,002 (880-1,133) µm, EP (distance from anterior end to excretory pore) = 74 (66-83) µm; tail length = 83 (65-91) µm, and E% (EP/tail length × 100) = 89.6 (78-114). Lateral field pattern variable, the formula for the arrangement of ridges from head to tail is: 2, 6, 7, 8, 4, 2. The portion with eight ridges is the longest. This new species can be differentiated further from three closest species (S. feltiae, S. glaseri, and S. oregonense) by characteristic sequences of their ITS regions, including sequence lengths, ratios of similarity, composition, and differences in base characters in sequence alignment. 相似文献

DNA sequence analysis was used to characterize the nuclear ribosomal DNA ITS1 region and a portion of the COII and 16S rDNA genes of the mitochondrial genome from Steinernema entomopathogenic nematodes. Nuclear ITS1 nucleotide divergence among seven Steinernema spp. ranged from 6 to 22%, and mtDNA divergence among five species ranged from 12 to 20%. No intraspecific variation was observed among three S. feltiae strains. Phylogenetic analysis of both nuclear and mitochondrial DNA sequences confirms the existing morphological relationships of several Steinernema species. Both the rDNA ITS1 and mtDNA sequences were useful for resolving relationships among Steinernema taxa. 相似文献

Entomopathogenic nematode species differ in their optimum storage temperature; therefore, we conducted a study on the survival and infectivity of the recently described Steinernema innovationi from South Africa at five storage temperatures (5°C, 10°C, 15°C, 20°C, and 25°C) over 84 d using 20,000 infective juveniles (IJ) in 25 ml aqueous suspension containing 0.1% formalin. Our results showed that survival was highest and most stable at 15°C, ranging from 84% to 88% after 84 d. Infectivity of IJ against Galleria mellonella larvae was >90% for all temperatures except for 5°C at which survival decreased to 10% after 84 d. In addition, we stored 2.5 million IJ on a sponge formulation in 15 ml of 0.1% formalin solution for 84 d at the optimum 15°C followed by 2 wk storage at 25°C. Storage of the IJ on a sponge formulation for 14 d at 25°C post 15°C storage for 84 d did not have a detrimental effect on IJ survival (87%) or infectivity to G. mellonella (95%). 相似文献

We compared the pathogenicity of the entomopathogenic nematodes Heterorhabditis bacteriophora, Steinernema glaseri, and S. scarabaei against third instars of 12 white grub species. The Japanese beetle (Popillia japonica) was highly susceptible to all nematode species. Oriental beetle [Exomala (=Anomala) orientalis], European chafer (Rhizotrogus majalis), Asiatic garden beetle (Maladera castanea), and the May/June beetles Phyllophaga crinita, Ph. congrua, and Ph. (Subgenus Phytalus) georgiana were highly susceptible to S. scarabaei but had mediocre to low susceptibility to H. bacteriophora and S. glaseri. The black turfgrass ataenius (Ataenius spretulus) was very susceptible to H. bacteriophora but had mediocre susceptibility to S. glaseri and S. scarabaei. Northern (Cyclocephala borealis) and southern masked chafer (C. lurida) had mediocre and southwestern masked chafer (C. pasadenae) and green June beetle (Cotinis nitida) had low susceptibility to all nematode species. 相似文献

The effects of soil depth, soil type and temperature on the activity of the nematode Steinernema carpocapsae (Filipjev) were examined using larvae of the West Indian fruit fly, Anastrepha obliqua (Macquart). Bioassays involved applying infective juveniles (IJs) to the surface of sterilized sand in PVC tubes previously inoculated with fly larvae of two ages. The 50% lethal concentration (LC50) values estimated for 6-day-old larvae were 9, 20 and 102 IJs/cm2 in tubes containing 2, 5 and 8 cm depth of sand, respectively, whereas for 8-day-old larvae, LC50 values were 16, 40 and 157 IJs/cm2, respectively. The effect of soil texture on the activity of S. carpocapsae was tested by applying the corresponding LC50 concentrations of nematodes to sand, sand–clay and loamy–sand soils. For 6-day-old larvae, soil type had a highly significant effect on infection with the highest percentages of infection observed in the sand–clay mixture (60–82% depending on depth) compared to 45–64% infection in sand and 23–30% infection in loamy–sand soil. A very similar pattern was observed in 8-day-old larvae except that infection rates were significantly lower than in younger larvae. There was a significant interaction between soil type and soil depth. The effect of three temperatures (19, 25 and 30°C) on infection was examined in sand–clay soil. The infectivity of S. carpocapsae was affected by temperature and soil depth and by the interaction of these two factors. Response surface analysis applied to second order multiple linear regression models indicated that the optimal temperature for infection of larvae of both ages was ~26°C, at a depth of 7.9 cm for 6-day-old larvae and <2 cm for 8-day-old larvae, resulting in a predicted 91.4% infection of 6-day-old larvae and 61.2% infection of 8-day-old larvae. These results suggest that S. carpocapsae may have the potential to control fruit fly pests in tropical ecosystems with warm temperatures and high soil moisture levels, although this assertion requires field testing. 相似文献

A recently introduced fungus gnat, Bradysia agrestis, has caused serious problems in Korean propagation houses where vegetable seedlings are produced for transplant into the fields. Although chemical insecticides are available against this pest, alternate control measures are needed. A Korean isolate of the entomopathogenic nematode, Steinernema carpocapsae Pocheon strain, was tested against this insect in the laboratory and propagation house. In the laboratory, S. carpocapsae affected oviposition, with the untreated females laying an average of 121±25 eggs, whereas the treated females averaged 7±2 eggs. The infectivity of S. carpocapsae to the fungus gnat was affected by the developmental stage and temperature, with highest mortality observed with the third and fourth instars and pupal stage. Nematode mortality in the second instar fungus gnat ranged between 23 and 35%, but showed no significant differences among the temperatures tested. The egg and first instar were not infected by the nematode. In nematode dispersal studies, adult female fungus gnats alone dispersed S. carpocapsae from the nematode-treated area to the control area at a higher rate than male and female gnats or male gnats alone. In the propagation house experiments with watermelon seedlings, no significant difference was observed in fungus gnat larval reduction at S. carpocapsae concentrations of 5, 10, or 20 infective juveniles (IJs)/g of soil at 7, 14, and 21 days after treatment. In comparison with the control, the S. carpocapsae treatments significantly reduced B. agrestis larval numbers. When the watermelon seed was treated with S. carpocapsae at sowing, the larval density of B. agrestis was significantly reduced, that is, the number of B. agrestis larvae ranged from four to eight and from five to eight in the nematode-treated plots compared with 26 and 30 in the control plots on the 17th and 34th day post-treatment, respectively. In the chemical insecticide treatments, diflubenzuron and chlorpyrifos were significantly more effective than S. carpocapsae and diazinon in reducing larval populations of the fungus gnat. Our data show that, although some of the chemical insecticides were more effective than S. carpocapsae Pocheon strain, the nematode was still an effective tool for management of the fungus gnat larvae and in protecting seedlings from damage in propagation houses. 相似文献

A new strain of Steinernema feltiae (Rhabditida: Steinernematidae) was isolated in La Rioja (Spain) from larvae of Bibio hortulanus (Diptera: Bibionidae). A comparative morphometric analysis of this new strain and four additional S. feltiae isolates was performed. Although significant differences in morphometric measurements were observed, PCR-RFLP profiles and sequence analysis of the ITS region of rDNA confirmed the identity of the new strain as A2 RFLP type of S. feltiae. A comparative morphometric study among nematodes from three hosts, Galleria mellonella (Lepidoptera: Pyralidae), Spodoptera littoralis (Lepidoptera: Noctuidae) and B. hortulanus, was conducted. Ecological characterization of the Rioja isolate was performed in G. mellonella larvae. Larval mortality was 75.3 and 78.12% in penetration and sand column assays, respectively, and the percentage of penetrating infective juveniles was 12.0 and 2.8% in these assays. Larval mortality in the one-on-one bioassay was 4.2%, and in exposure-time bioassays, it was 50% at 11.25 hours. Relationships between morphometric characteristics and host mortality are discussed for this new strain of entomopathogenic nematode. 相似文献

We determined the mortality of adult tick Dermacentor silvarum Olenev after it was exposed to five entomopathogenic nematode species: Steinernema carpocapsae BJ (Sc BJ), S. glaseri NC32 (Sg NC32), S. ceratophorum D43 (Sc D43), S. feltiae Otio (Sf Otio) and Heterorhabditis bacteriophora E-6-7 (Hb E-6-7). All five species had lethal effects on adult ticks, and caused significant decreases in the amount of eggs laid by engorged female ticks (P < 0.05). Among them, Hb E-6-7 and Sc BJ were the most pathogenic to female adult ticks, with mortalities ranging from 70 to 90% (P < 0.05). Engorged females were more susceptible than unfed and partially fed females. The cumulative mortalities of engorged females exposed to Hb E-6-7 and Sc BJ for 14 days were 90%, with LT50 at 6.3 and 7.0 days, respectively. Dose-dependent effect was observed in the oviposition of the ticks after they were exposed to nematodes at concentrations of 1000–5000 IJs/dish. When the concentration was higher than 5000 IJs/dish, dose dependent effects disappeared (P > 0.05). All five nematode species were lethal to unfed male ticks, and there was significant difference in mortality when compared to the control group (P < 0.05); however, the pathogenicity varied among them. 相似文献

A method of selecting a Steinernema feltiae strain that is effective against a mushroom fly, Lycoriella solani, is described in detail. The pest control efficacy of the selected nematode strain was evaluated and compared with the efficacy of two unselected strains. The selection procedure was designed to give preference to nematode individuals with the greatest ability (1) to search effectively for the target insect larvae in their natural habitat, (2) to infect them shortly after application and (3) to reproduce in their haemocoel. Thirty‐four rounds of selection achieved a 4‐fold improvement in nematode ability to find and parasitize third‐ and fourth‐instar larvae of the pest in the mushroom substrate. In 24‐h laboratory experiments, mortality of the insect caused by nematode juveniles rose from 22.5%, recorded for the original unselected isolate, to 92.5% for the selected strain. In a 51‐day experiment conducted on a mixed age mushroom house population of L. solani, the enhanced pest control ability of the selected strain was detected shortly after nematode application and remained high throughout the experimental period. During the first 4 weeks of the trial the selected nematode strain was significantly better than both unselected strains and caused 91.1–92.7% reduction of the fly emergence from the mushroom substrate. No difference was observed between the efficacy of the selected nematodes applied at 1 × 106 and 3 ×106 infective juveniles per m2, while the unselected strains performed significantly better at the higher concentration. All the nematodes examined showed good persistence in the mushroom casing apparently due to recycling in the insect host. 相似文献
