The transport of human-mouse hybrid class I histocompatibility antigens has been studied in a mutant human cell line, 174 × CEM.T2 (T2). T2, a somatic cell hybrid of human B- and T-lymphoblastoid cell lines (B-LCL and T-LCL, respectively), synthesizes HLA-A2 and HLA-B5 glycoproteins, but expresses only low levels of A2 and undetectable levels of B5 at the cell surface. We have previously shown that the products of human class I genes introduced into T2 by transfection behave like the endogenous HLA-B5 glycoproteins, while the products of mouse class I alleles similarly introduced are transported normally to the cell surface. We have now determined that the surface expression of class I glycoproteins in T2 depends on the origin of the 1 and 2 domains. Human (HLA-B7) and mouse (H-2D p ) hybrid class I genes, encoding the leader, 1, and 2 sequences of one species fused to the 3, transmembrane, and cytoplasmic domains of the other, were transfected into T2. Normal surface expression of the hybrid class I molecule was observed in T2 only when the leader, 1, and 2-encoding exons were derived from the mouse gene. The reciprocal construct, encoding human leader, 1, and 2 domains fused to the mouse 3, transmembrane, and cytoplasmic regions, resulted in biosynthesis of a hybrid glycoprotein which was not transported to the cell surface. The products of both constructs were expressed normally in control cells. The effects of glycosylation on class I antigen transport were also studied using mutant class I constructs with altered glycosylation sites. Two mutant B7 genes encoding either an extra glycosylation site at position 176 or no glycosylation sites were transfected into T2. These mutant products were expressed at the cell surface in control cells, but were synthesized and not surface-expressed in T2. These data demonstrate that the HLA/H-2 transport dichotomy in T2 is a function of the origin of the 1 and/or 2 domains of the class I glycoprotein, and is not a reflection of glycosylation differences between the human and mouse molecules. Alterations inN- andO-linked glycosylation affect cell surface expression and antigenicity of recombinant glycophorin A expressed in transfected Chinese hamster ovary (CHO) cells. To understand these effects further, glycophorin A was purified by immunoaffinity chromatography from transfected wild type and glycosylation deficient CHO cells. TheO-glycans were characterized both biochemically, using gel filtration and high performance anion exchange chromatography, and immunologically, using carbohydrate specific monoclonal antibodies to probe Western blots. TheO-glycans of human erythrocyte glycophorin A consist mainly of short oligosaccharides with one, two, or three sialic acid residues linked to a common disaccharide core, Gal1-3GalNAc1-Ser/Thr, with the disialylated structure being the most abundant. With the exception of the trisialylated derivative, the same structures were found on recombinant glycophorin A expressed by wild type CHO cells. However, in contrast to human crythrocyte glycophorin A, the monosialylated oligosaccharide was the most abundant structure on the recombinant protein. Furthermore, recombinant glycophorin A was shown to express a small amount of the Tn antigen (GalNAc1-Ser/Thr). Recombinant glycophorin A had the sameO-glycan composition, whether purified from clones expressing high or moderate levels of the recombinant glycoprotein. This indicates that the level of expression of the transfected glycoprotein did not affect itsO-glycan composition. Deletion of theN-linked glycosylation site at Asn26, by introducing the Mi.I mutation (Thr28 Met) by site-directed mutagenesis, did not markedly affect theO-glycan composition of the resulting recombinant glycoprotein expressed in wild type CHO cells. This demonstrates that the presence or absence of theN-glycan did not influenceO-linked glycosylation of the recombinant glycoprotein. Finally, theO-glycans on recombinant glycophorin A expressed in the Lec 2 and Lec 8 glycosylation deficient CHO cells were characterized. TheO-glycans on Lec 2 cell glycophorin A were predominantly Gal1-3GalNAc1-Ser/Thr (T antigen), while those on Lec 8 glycophorin A were exclusively GalNAc1-Ser/Thr (Tn antigen). These results will lead to a better understanding of the cell biology and immunology of this important human erythrocyte glycoprotein.  相似文献

In this report, we describe the efficient expression and glycosylation, in insect cells, of -phaseolin polypeptides (M r 45 and 48 kDa) from Phaseolus vulgaris, by means of a baculovirus expression vector. N-terminal sequence analysis demonstrated that the signal peptide was efficiently processed. Tunicamycin treatment suppressed both phaseolin bands seen in untreated or control cells, and resulted in a single species (M r 43 kDa). We provide evidence that the observed size heterogeneity arises by asymmetric glycosylation of a single, high-molecular weight precursor. These results also indicate that differential glycosylation of phaseolin polypeptides can occur on the product of a single gene, and, in that sense, is not dependent on amino acid sequence variations. Phaseolin accumulates to a very high level (90 µg/106 cells), 90% of it being secreted into the culture medium. Immuno-gold staining and electron microscopy demonstrated phaseolin polypeptides in electron-dense, membrane-bound vesicles seen at the periphery of the cytoplasm of infect cells and in cytoplasmic multivesicular bodies. The effect on protein accumulation of a single-basepair transversion (G»C) at position +6 is also described. This study constitutes, to our knowledge, one of the first instances of a plant protein being expressed in insect cells and suggests possible differences in the sorting mechanisms of glycoproteins from legume seeds and those from Spodoptera frugiperda cell line Sf9.  相似文献

Many indirect serum studies show changes in protein glycosylation in disease, but further progress will require direct investigation of oligosaccharide composition. Current methods of deglycosylation using PNGase-F often result in incomplete removal of oligosaccharides. This is an unsatisfactory situation because only small quantities of material are often available in clinical studies, glycosylation changes may occur in only a small proportion of the molecules and quantification of the released oligosaccharides may be unreliable. The ability of PNGase-F to deglycosylate haptoglobin (Hp) under different conditions has been investigated. Oligosaccharides were completely removed from 50g of Hp by treatment for 24 h with PNGase-F in 50 mmol/l ammonium formate buffer, pH 8.6, in combination with sodium dodecyl sulphate, mercaptoethanol and Nonidet P40. This modified procedure was equally effective at removing oligosaccharides from other serum glycoproteins (1 glycoprotein, 1, transferrin) and fetuin, and its efficiency was independent of the polymeric structure of the molecule or the amount of glycosylation. The method has the additional advantage of using 20% less enzyme than previous methods, which substantially reduces costs.  相似文献

In this work, metabolite and antibody production kinetics of hybridoma cultures were investigated as a function of cell density and growth rate in a homogeneous perfusion reactor. Hydrophilized hollow fiber polypropylene membranes with a pore size of 0.2 m were used for medium perfusion. Oxygen was supplied to the cells through thin walled silicone tubing. The mouse-mouse hybridoma cells were grown in three identical bioreactors at perfusion rates of 1.1, 2.0, and 3.2/day for a period of eight days during which the viable cell concentrations reached stable values of 2.6×106, 3.5×106, and 5.2×106 cells/ml, respectively. Total cell densities reached values ranging from 8×106 to 1×106 cells/ml. Specific substrate consumption and product formation rates responded differently to changes in cell density and apparent specific growth rate, which were not varied independently. Using multiple regression analysis, the specific glucose consumption rate was found to vary with viable cell density while the specific glutamine uptake and lactate production rates varied with both viable cell density and apparent specific growth rate. These results suggest that cell density dictates the rate of glucose consumption while the cell growth rate influences how glucose is metabolized, i.e., through glycolysis or the TCA cycle. Blood specimens from 69 gibbons (63Hylobates lar, 4Hylobates concolor, and 2Hylobates pileatus) were tested for human-type ABO, MN, and Rh blood groups. AmongH. lar, three phenotypes were noted in the ABO and MN blood groups respectively, but all fourH. concolor were grouped as AM. All group A gibbons were of subgroup A1; subgroups A2B and A12B were observed at a low frequency in group AB gibbons. Leb antigen was detected in about 30% of the red cell samples fromH. lar, but all the samples were negative for Lea. All the gibbons tested had c(hr) antigen but no other Rh antigens (D, C, E, and e) in their red cells. Some selected blood samples fromH. lar were also tested for some other blood group antigens and for the Gm and Inv factors. The Jka antigen was detected in all the red cell samples tested, but the S, s, U, K, k, and Fya antigens were not. In the tests of plasma with anti-Gm (1),H. lar could be divided into two groups, i.e., Gm(1)Gi and Gm(–1)Gi; Gm(2), Gm(4), and Inv(1) were absent in the species.  相似文献

Niche width and inter‐individual diet variation in predator populations are known to be affected by intrinsic factors such as body size, age, cognitive constraints, and by intra‐ and interspecific competition. By contrast, how variation in prey biological traits may affect niche width and partitioning is still a poorly explored topic. One of these candidate traits is prey mobility, which can affect the predators’ niche because acting on the rate of encounter and, assuming mobility as a proxy for escape capability, on the success of predator attacks. Here we analysed 20 wasp populations and their prey as individual‐based food‐webs to test if prey mobility may explain niche width (here defined by the Shannon entropy‐based index ) and patterns of inter‐individual diet variation (here defined by the interaction‐exclusiveness index H2 and the interaction evenness index E2). Niche width was very variable among populations and overall network specialization (H2) was always higher than the expected by null models. In case of high‐speed flying prey (e.g. flies, bees), wasps showed wider niches () and lower specialization (H2) than in case of non‐flying or slow‐flying prey (e.g. spiders, beetles). Evenness (E2), on the other side, did not vary with prey mobility. Altogether, these results suggest that highly elusive prey may lead to wider predators’ niche but somehow limit their individual niche specialization.  相似文献

The absorption, circular dichroism, and resonance Raman spectra of horse heart ferricytochromec in the presence of 0.2 M KCl, 0.1 M NaClO4, and 0.2 M KNO3, in thepH region 7 to 0.5, have been investigated to determine the nature and the course of the processes involved. As in the absence of salts (Myer, Y., and Saturno, A. F. (1990)J. Protein Chem.,9, 379–387), the change from neutral to low acidicpH's in the presence of salts is a three-step process: state III s state III s,a state II s state I s , withpK a 's of 3.5±0.2, 2.2±0.2, and 1.1±0.2, and with two, one, and one number of protons, respectively. The addition of salts at neutralpH's has little or no effect on the protein conformation and the heme-iron configuration (i.e., they remain the same, low-spin hexacoordinated heme iron with a Met-80-Fe-His-18 axial coordination), but such addition does cause a slight tightening of the heme crevice and the enlargement of the porphyrin core. State III s,a is a folded state with about the same degree of folding and with a similar spin state and coordination configuration of iron, but the heme crevice is loosened and the porphyrin core is smaller. Both states II s and I s are also essentially folded forms, but with a smaller degree of protein secondary structure. State II s has a high-spin hexacoordinated heme iron with a water molecule and a protonated and/or hydrogen-bonded imidazole of his-18 as the two axial ligates; and state I s has a high-spin pentacoordinated heme iron, which is about 0.49 Å out of the porphyrin plane, with a protonated and/or hydrogen-bonded imidazole nitrogen as the only axial ligate. The addition of anions causes the stabilization of the protein secondary structures and the state III a state II transition. The mode of effectiveness of anions appears to be nonspecific (i.e., because of electrostatic shielding and/or disruption of salt bridges).  相似文献

Studies were conducted using a novel in vitro approach to investigate the efficacy of acetamidine hydrochloride (ACE) and guanidine hydrochloride (GUAN), previously shown to block gramicidin D (GRAM) channels in artificial membranes, in preventing the toxic effects of GRAM in NG108-15 (neuroblastoma×glioma hybrid) cells. Specifically, intracellular microelectrode techniques were employed to examine changes in membrane resting potential (V m) and input resistance (R in). At 1 mol/L, ACE significantly reduced loss of V m induced by 1 or 10 g/ml GRAM, although higher concentrations of ACE did not afford enhanced antagonism. GUAN, in contrast, produced a concentration-dependent antagonism of GRAM-induced V m and R in loss, with high concentrations (10 or 100 mol/L) completely preventing diminutions in both V m and R in. In control cells superfused without GRAM, ACE produced a direct, concentration-dependent reduction in V m and R in, whereas GUAN hyperpolarized NG108-15 cells but did not alter R in. These data represent the initial demonstration of the reversal of GRAM toxicity in an intact cell system.  相似文献

Structural differences have been reported in the glycosylation patterns of cystic fibrosis glycoproteins. Although the gene mutated in cystic fibrosis (CFTR) has been cloned and characterized as a chloride channel, its relationship to the highly viscous mucus and structural glycoprotein and mucin abnormalities in cystic fibrosis still remains to be defined. We have evaluated O-glycan biosynthesis in CHO and BHK cells that express CFTR and F508 CFTR as in vitro models, and utilized the cftr knockout mouse as an in vivo model of CFTR dysfunction. Activities of glycosyltransferases and sulfotransferases synthesizing mucin type O-glycan chains were determined in these models. Differences in transferase activity levels were found between tissues and cell types and during mouse development. No specific patterns of activities were associated with the lack of CFTR or with F508CFTR expression. This suggests that it is not the presence or absence of normal CFTR, or the presence of mutant CFTR alone, but rather cell specific additional factors or pathophysiological consequences that determine the changes in mucin glycosylation in cystic fibrosis.  相似文献

Cardiac hypertrophy is a compensatory response of myocardial tissue upon increased mechanical load. Of the mechanical factors, stretch is rapidly followed by hypertrophic responses. We tried to elucidate the role of angiotensin II (AII), endothelin-1 (ET-1) and transforming growth factor- (TGF-) as autocrine/paracrine mediators of stretch-induced cardiomyocyte hypertrophy. We collected conditioned medium (CM) from stretched cardiomyocytes and from other stretched cardiac cells, such as cardiac fibroblasts, endothelial cells and vascular smooth muscle cells (VSMCs). These CMs were administered to stationary cardiomyocytes with or without an AII type 1 (AT1) receptor antagonist (losartan), an ET-1 type A (ETA) receptor antagonist (BQ610), or anti-TGF- antibodies. By measuring the mRNA levels of the proto-oncogene c-fos and the hypertrophy marker gene atrial natriuretic peptide (ANP), the molecular phenotype of the CM-treated stationary cardiomyocytes was characterized.Our results showed that c-fos and ANP expression in stationary cardiomyocytes was increased by AII release from cardiomyocytes that had been stretched for 60 min. Stretched cardiomyocytes, cardiac fibroblasts and endothelial cells released ET-1 which led to increased c-fos and ANP expression in stationary cardiomyocytes. ET-1 released by stretched VSMCs, and TGF- released by stretched cardiac fibroblasts and endothelial cells, appeared to be paracrine mediators of ANP expression in stationary cardiomyocytes.These results indicate that AII, ET-1 and TGF- (released by cardiac and vascular cell types) act as autocrine/paracrine mediators of stretch-induced cardiomyocyte hypertrophy. Therefore, it is likely that in stretched myocardium the cardiomyocytes, cardiac fibroblasts, endothelial cells and VSMCs take part in intercellular interactions contributing to cardiomyocyte hypertrophy.  相似文献

A statistical analysis of protein conformations in terms of the distance between residues, represented by their C atoms, is presented. We consider four factors that contribute to the determination of the distanced i,i+k between a given pair ofith and(i+k)th residues in the native conformation of a globular protein: (1) the distancek along the chain, (2) the size of the protein, (3) the conformational states of theith to(i+k)th residues, and (4) the amino acid types of the and(i+k)th residues. In order to account for the dependence on the distancek along the chain, the statistics are taken for three ranges, viz., short, medium, and long ranges (k8; 9k20; andk21; respectively). In the statistics of short-range distances, a mean distanceD k and its standard deviationS k are calculated for each value ofk, with and without taking into account the conformational states of all residues fromi toi+k (factors 1 and 3). As an Appendix, the relations for converting from the distances between residues into other conformational parameters are discussed. In the statistics of long-range distances, a reduced distanced* ij (the actual distance divided by the radius of gyration) is used to scale the data so that they become independent of protein size, and then a mean reduced distanceD l (a, a) and its standard deviation l (a, a) are calculated for each amino acid pair (a, a) (factors 2 and 4). The effect of the neighboring residues along the chain on the value of the distanced* ij is explored by a linear regression analysis between the actual reduced distanced* ij and the mean value over theD l for all possible pairs of residues in the two segments of the (i–2)th to the (i+2)th and the (j–2)th to the (j+2)th residues. The effect is assessed in terms of the tangentA l (a, a) of the calculated regression line for each amino acid pair (a, a). In the statistics of medium-range distances, only factors 1 and 4 are considered, to simplify the analysis. The scaled distanced i,i+k =(d i,i+k -D k )/S k is used to eliminate the dependence onk, the distance along the chain. The propertiesD m (a, a), m (a, a) andA m (a, a) corresponding toD l (a, a), l (a, a), andA l (a, a), and also calculated for each amino acid pair (a, a). The results are interpreted as follows: the smaller values ofD l (a, a) andD m (a, a) indicate a preference of the pair (a, a) for a contact (e.g., pairs between hydrophobic amino acids, and pairs of Cys with aromatic amino acids), and the larger values of these quantities indicate a preference for distant mutual location (e.g., pairs between strong hydrophilic amino acids); the smaller values of l (a, a) and m (a, a) indicate a strong preference for either contact or noncontact (e.g., pairs between hydrophobic amino acids, and pairs between strong hydrophobic and hydrophilic amino acids, respectively), and the larger values of these quantities indicate the ambivalent/neutral nature of the preference for contact and noncontact (e.g., pairs containing Ser or Thr); the smaller values ofA l (a, a) andA m (a, a) indicate that the distance of an (a, a) pair is determined independently of the amino acid character of the neighboring residues along the chain (e.g., some pairs of Cys or Met with other amino acids) and the larger values of these quantities indicare that such amino acid character contributes strongly to the determination of the distance (e.g., pairs containing Ser or Thr, and pairs between amino acids with small side chains). The difference between the statistics for the long- and medium-range distances is also discussed; the former reflect the difference between the hydrophobic and hydrophilic character of the residues, but the latter cannot be easily interpretable only in terms of hydrophobicity and hydrophilicity. The data analyzed here are used in the optimization of an object function to compute protein conformation in a subsequent paper.  相似文献

This paper considers the composition and function of sensory systems monitoring H2O2 level by the lung neuroepithelial cells and carotid bodies. These systems are localized in the plasma membrane of the corresponding cells and are composed of O 2 -generating NADPH-oxidase and an H2O2-activated K+ channel. This complex structure of the H2O2 sensors is probably due to their function in antioxidant defense. By means of these sensors, an increase in the H2O2 level in lung or blood results in a decrease in lung ventilation and constriction of blood vessels. This action lowers the O2 flux to the tissues and, hence, intracellular [O2]. The [O2] decrease, in turn, inhibits intracellular generation of reactive oxygen species. The possible roles of such systems under normal conditions (e.g., the effect of O 2 in air) and in some pathologies (e.g., pneumonia) is discussed.  相似文献

Two characteristic temperatures were identified from measurements of the temperature dependence of O2 evolution by Chlorella vulgaris and Anacystis nidulans: T1, the threshold temperature for inhibition of O2 evolution under saturating light conditions, and T2, the upper temperature limit for O2 evolution. Measurement of delayed light emission from photosystem II (PSII) showed that it passed through a maximum at T1 and was virtually eliminated on heating the samples to T2. Related changes were observed in low-temperature (77K) fluoresence emission spectra. Heat-stress had little effect on the absorption properties of the cells at temperatures below T1 but incubation at higher temperatures, particularly under high-light conditions, resulted in extensive absorption losses. An analysis of these measurements suggests that this increased susceptibility to photobleaching is triggered by an inhibition of the flow of reducing equivalents from PSII that normally serves to protect the light-harvesting apparatus of the cells from photo-oxidation. Summary P730, the active phytochrome, increases the rate of RNA synthesis (Table) and the RNA contents in the cotyledons of the mustard seedling (Sinapis alba L.) (Fig. 1) whereas the RNA contents in the hypocotyl is decreased under the influence of P730 (Fig. 2).—It takes about 6 hours until changes in the RNA contents-which must be attributed to the formation of P730—can be measured after the onset of light (continuous far-red). Since the lag-phases of positive photoresponses in the cotyledons and negative photoresponses in the hypocotyl (Mohr, 1966) are in general much shorter than 6 hours, the changes of the RNA contents of the organs cannot be regarded as being directly connected with the formation of characteristic positive photoresponses such as anthocyanin synthesis, induced enzyme synthesis, ascorbic acid synthesis, etc., or negative photoresponses such as inhibition of hypocotyl lengthening.We have rather to conclude that the changes of RNA contents are secondary adaptations of the organs which lead to an increase (cotyledons) or decrease (hypocotyl) of protein synthesizing capacity of the cells and tissues. The P730-dependent increase of bulk RNA in the cotyledons is probably due to a differential gene activation and the P730-dependent decrease of bulk RNA in the hypocotyl is due to a differential gene repression. The causalities of these processes are possibly complex.The hypothesis of differential gene activation or repression by P730 (Mohr, 1966; Schopfer, 1967a, b) is not disproved by these results. We have rather to reach a conclusion which has already been suggested by other data (e.g. Karow and Mohr, 1966), namely, that positive as well as negative photoresponses are due to changes in the activity of a limited (possibly small) number of enzymes. Correspondingly changes in only a minute amount of the total RNA are directly involved in the formation of photoresponses. These changes cannot be detected by following RNA contents.—It seems to be of great interest, however, that P730 eventually brings about strong tissue specific changes in the RNA contents per cell as described in the present paper.  相似文献

The structural gene locus specifying subunits of the cytoplasmic isozymes of phosphoglucose isomerase (PGI) is present in duplicate in many diploid species of Clarkia (Onagraceae), a genus of annual plants native to California. We studied the kinetic properties and molecular weights of a large number of genetically defined and highly purified PGIs in C. xantiana, a species with the duplication, as a means of examining the biochemical consequences of the evolution of a new gene locus. This species is primarily outcrossing, but also includes several previously described predominantly self-pollinating populations. Both cytoplasmic PGI loci in the outcrossing populations are polymorphic and their enzyme products are readily separated by electrophoresis. The PGIs from the outcrossing populations were generally closely similar in molecular weight, pH optimum, heat sensitivity, energy of activation, and apparent K m (fructose-6-phosphate). The PGI loci in the selfing populations are monomorphic and specify enzymes having identical electrophoretic mobilities to those coded by the most frequent alleles of the outcrosser. The PGI isozymes in the selfers differed fivefold in K m , suggesting that they have a very different catalytic effectiveness. The high K m of the PGI-3A isozyme (1.1mm) was anomalous among the examined and would likely be disadvantageous in a species which lacked other more normally functioning PGIs. But in the cytoplasm of the selfing plants, it is present with other PGIs that have low K m values. The PGI-3A enzyme is a good candidate for a gene product coded by a forbidden mutation that could not have been established in the absence of the duplication. The flash-induced kinetics of various characteristics of Photosystem II (PS II) in the thylakoids of oxygenic plants are modulated by a period of two, due to the function of a two-electron gate in the electron acceptor side, and by a period of four, due to the changes in the state of the oxygen-evolving complex. In the absence of inhibitors of PS II, the assignment of measured signal to the oxygen-evolving complex or to quinone acceptor side has frequently been done on the basis of the periodicity of its flash-induced oscillations, i.e. four or two. However, in some circumstances, the period four oscillatory processes of the donor side of PS II can generate period two oscillations. It is shown here that in the Kok model of oxygen evolution (equal misses and equal double hits), the sum of the concentrations of the S 0 and S 2 states (as well as the sum of concentrations of S 1 and S 3 states) oscillates with period of two: S 0+S 2S 1+S 3S 0+S 2S 1+S 3. Moreover, in the generalized Kok model (with specific miss factors and double hits for each S-state) there always exist such 0, 1, 2, 3 that the sum 0[S0] + 1[S1] + 2[S2] + 3[S3] oscillates with period of two as a function of flash number. Therefore, the decomposition of the flash-induced oscillations of some measured parameters into binary oscillations, depending only on the acceptor side of PS II, and quaternary oscillations, depending only on the donor side of PS II, becomes practically impossible when measured with techniques (such as fluorescence of chlorophyll a, delayed fluorescence, electrochromic shift, transmembrane electrical potential, changes of pH and others) that could not spectrally distinguish the donor and acceptor sides. NonenergizedStreptococcus bovis cells, which were washed in potassium-phosphate buffer and incubated in Tris buffer containing 200mm potassium chloride (pH 6.5), did not take up tetraphenylphosphonium ion (TPP+), but the same cells took up TPP+ when they were incubated in Tris buffer lacking potassium. This result indicated that passive potassium diffusion was creating an electrical potential () across the cell membrane. Neither cells took significant amounts of 9-aminoacridine (9-AA), an intracellular pH marker. Cells that were incubated in Tris buffer and treated with carbonyl cyanidem-chlorophenylhydrazone (CCCP) took up 9-AA, and this result indicated that this protonophore was facilitating proton influx. The ionophores monensin and lasalocid also caused 9-AA uptake, and it appeared that they were responsible for or responsive to potassium/proton antiport. However, there was also a rapid accumulation of 9-AA when the cells were treated with valinomycin, a potassium uniporter that cannot translocate protons. This latter result indicated that potassium efflux was associated with another avenue of proton influx (e. g., potassium/proton symport). Because cells treated with dicyclohexyl carbodiimide (DCCD) also exhibited valinomycin-dependent 9-AA uptake, it is unlikely that the F1F0ATPase or ATP formation was responsible for proton flux across the cell membrane.  相似文献

Summary Although an outwardly rectifying K+ conductance (I K, A) is prominently expressed in human alveolar macrophages, the expression of this conductance in human monocyte-derived macrophages (HMDMs) is rare. We have analyzed the induction of the expression of I K, A in voltage-clamped, in vitro differentiated HMDMs by a number of stimuli which produce either priming or activation of macrophages. Cultures were stimulated with lipopolysaccharide (LPS, 2 g/ml), interleukin 2 (IL-2, 100 U/ml), or combinations of LPS and either recombinant interferon-gamma (-IFN, 10 U/ml), phorbol myristate acetate (PMA, 0.01 or 1 g/ ml) and platelet activating factor (PAF, 20 ng/ml) for periods of up to 24 hr. Treatment of the cells with either LPS or IL-2 greatly enhanced the frequency of current expression. Treatment with either PMA or -IFN alone did not induce current expression; treatment of the cells with a combination of LPS and either PMA, -IFN, or PAF did not enhance current expression over that observed with LPS alone. The expression of the outwardly rectifying K+ current was observed in 36% (n=321) of the cells for cultures treated with LPS and 33% (n=55) of the cells for cultures treated with IL-2. The inactivating outward K+ current was absent in cells which were not treated with either LPS or IL-2. The kinetics of current activation and inactivation appeared identical to that previously described for the transient-inactivating outward current of the human alveolar macrophage. Cycloheximide (1 g/ml), an inhibitor of protein synthesis, completely suppressed LPS-induced current expression. No correlation was found between peak current amplitude and cell size in LPS-activated cells expressing the outwardly rectifying K+ current, indicating that current density was not held constant from cell to cell. The coupling of ion channel expression and secretion in individual HMDMs was studied using the reverse hemolytic plaque assay. Although an enhancement of K + current expression was observed following either LPS or IL-2 treatment, a quantitatively similar and uniform increase in the percentage of either IL-1 or lysozyme-secreting cells was not observed. The frequency of current expression in cells identified as secreting tumor necrosis factor- (TNF-), interleukin 1 (IL-1), or lysozyme was the same or decreased over that observed for nonsecreting cells. Thus, LPS treatment increases the number of K+ channels on HMDM membranes; however, K+ channel expression alone was not sufficient to give rise to enhanced secretion in LPS-activated macrophages. Enhanced K+ channel expression appears to be a part of the primary activation signal. K+-channel activation would hyperpolarize the membrane potential, potentially providing the driving force for calcium entry through voltage-independent pathways activated by the subsequent binding of soluble substances to membrane surface receptors, the secondary signal linked to secretion.This work was supported by NIH grant RO 1 GM36823.  相似文献

A model is presented in which the potassium conductance (G K ) changes responsible for the afterhyperpolarization (AHP) in motoneurones are postulated to follow a kinetics coherent with Hodgkin-Huxley membrane model. Such G K kinetics, which operates as a bistable system switched from one state to the other by the action potential, can be expressed by simple analytical expressions if the spike is approximated to a rectangular pulse. Accumulation of G K by repetitive activation of the model accurately describes the different features of AHP summation in motoneurones; moreover, this accumulation process enables a model for repetitive firing of motoneurones to display secondary range firing at steady state.  相似文献
