We study the sensitivity of fishery management per-recruit harvest rates which may be part of a quantitative harvest strategy designed to achieve some objective for catch or population size. We use a local influence sensitivity analysis to derive equations that describe how these reference harvest rates are affected by perturbations to productivity processes. These equations give a basic theoretical understanding of sensitivity that can be used to predict what the likely impacts of future changes in productivity will be. Our results indicate that per-recruit reference harvest rates are more sensitive to perturbations when the equilibrium catch or population size per recruit, as functions of the harvest rate, have less curvature near the reference point. Overall our results suggest that per recruit reference points will, with some exceptions, usually increase if (1) growth rates increase, (2) natural mortality rates increase, or (3) fishery selectivity increases to an older age.  相似文献

Pharmacokinetics were melded with a viral dynamical model to design an optimal drug administration regimen such that the basic reproductive number for the virus was minimized. One-compartmental models with two kinds of drug delivery routes, intravenous and extravascular with multiple dosages, and two drug elimination rates, first order and Michaelis-Menten rates, were considered. We defined explicitly the basic reproductive number for the viral dynamical model melded with pharmacokinetics. When the average plasma drug concentration was constant, intravenous administration of the drug with small dosages applied frequently minimized the basic reproductive number. For extravascular administration, the basic reproductive number initially decreases to a trough point and then increases as the drug dosage increases. When a therapeutic window is considered, numerical studies indicate that the wider the window, the smaller the basic reproductive number. Once the width of the therapeutic window is fixed, the basic reproductive number monotonously declines as the minimum therapeutic level increases. The findings suggest that the existence of drug dosage and drug administration interval that minimize the basic reproductive number could help design the optimal drug administration regimen.  相似文献

The digital sharing economy is commonly thought to promote sustainable consumption and improve material efficiency through better utilization of existing product stocks. However, the cost savings and convenience of using digital sharing platforms can ultimately stimulate additional demand for products and services. As a result, some or even all of the expected environmental benefits attributed to sharing could be offset, a phenomenon known as the rebound effect. Relying on a unique dataset covering over 750,000 food items shared in the United Kingdom through a free peer-to-peer food-sharing platform, we use econometric modeling, geo-spatial network analysis, and environmentally extended input–output analysis to quantify how much of the expected environmental benefits attributed to sharing are offset via rebound effects under seven re-spending scenarios. We find that rebound effects can offset 59–94% of expected greenhouse gas (GHG) emission reduction, 20–81% of expected water depletion benefits, and 23–90% of land use benefit as platform users re-spent the money saved from food sharing on other goods and services. Our results demonstrate that rebound effects could limit the potential to achieve meaningful reductions in environmental burdens through sharing, and highlight the importance of incorporating rebound effects in environmental assessments of the digital sharing economy.  相似文献

Insects harbor a wide range of microbial symbionts, but their influence on host phenotypes is described in a limited number of biological models. One experimental approach to gain knowledge on the effects of symbionts to their hosts is to create insect lines with and without symbionts and examine their phenotypes. However, the success rate of symbiont elimination and introduction methods is dependent on several parameters that are scarcely tested or described. The pea aphid, Acyrthosiphon pisum Harris (Hemiptera: Aphididae), is a model insect of symbiosis studies. It harbors a primary symbiont that supplies the host with essential amino acids, and an array of secondary symbionts whose effects have been assessed by manipulating their presence/absence in the insect. Here, we describe the influence of key parameters on the success rate of symbiont manipulation using the pea aphid–secondary symbiont system. We compared two elimination methods differing in antibiotic treatment using several aphid–symbiont combinations. We also created new aphid host–symbiont combinations by secondary symbiont introduction and examined the effects of larval stage of recipient aphids on introduction success. Our study revealed that the aphid–symbiont combination has strong influence on both symbiont introduction and elimination success rates, and that the type of antibiotics and the larval stage of recipient aphids influence the elimination and introduction success rate, respectively.  相似文献

Flow cytometry and Northern blotting were used to examine the effects of hydrodynamic forces in stirred tank bioreactors on CD13 receptor surface content and mRNA levels of HL60 (human promyelocytic leukemia) cells. A step increase in agitation rate from 80 to 300 or 400 rpm reduced the apparent HL60 growth rate in a dose-dependent manner. This step increase in agitation rate (to 300 or 400 rpm) also increased the CD13 receptor surface content on averge by 30% and 100%, respectively. This increase in CD13 receptor surface content was correlated with a 10% and a 60% increase in CD13 mRNA levels. We also observed a significant and very reproducible drop in CD13 expression over the course of a batch bioreactor run (80 rpm). Although we have no explanation for this, we show that the decrease in CD13 receptor surface content can be (at least partially, if not fully) explained by the corresponding decrease in CD13 mRNA. HL60 cell cultures agitated at 300 and 400 rpm exhibited glucose consumption and lactate production rates that were approximately 40% and 90% greater than values of the cultures agitated at 80 rpm. The physiological and practical implications of these results are discussed.  相似文献

A model is developed to explain the following experimental observations: oligomers of the alternating DNA copolymer dAT serve as templates for the enzymatic synthesis of macromolecular dAT; the rate of polymer synthesis with a template oligomer of given length increases to a maximum and then decreases as the temperature increases; the temperature for the maximum rate of synthesis increases with the length of the template. These observations were interpreted in terms of the repeated “slipping” of the oligomer along the product strand to expose template sites. This communication extends this idea. We suppose that the rate-limiting step in the polymerization is either the attachment of the substrate to the exposed template site or the synthesis of the phosphodiester bond. Then the rate of synthesis is proportional to the equilibrium concentration either of exposed template sites or of template sites occupied by substrate molecules. These concentrations are evaluated in terms of a simple theory of the helix–coil transition. The model qualitatively reproduces the dependence of the rate of synthesis on template length and temperature and predicts rates of synthesis in approximate agreement with experiment.  相似文献

Alpha-thujone (1alpha) and beta-thujone (1beta) were used to investigate the mechanism of hydrocarbon hydroxylation by cytochromes P-450(cam) (CYP101) and P-450(BM3) (CYP102). The thujones are hydroxylated by these enzymes at various positions, but oxidation at C-4 gives rise to both rearranged and unrearranged hydroxylation products. Rearranged products result from the formation of a radical intermediate that can undergo either inversion of stereochemistry or ring opening of the adjacent cyclopropane ring. Both of these rearrangements, as well as a C-4 desaturation reaction, are observed. The ring opening clock gives oxygen rebound rates that range from 0.2 x 10(10) to 2.8 x 10(10) s(-1) for the different substrate and enzyme combinations. The C-4 inversion reaction provides independent confirmation of a radical intermediate. The phenol product expected if a C-4 cationic rather than radical intermediate is formed is not detected. The results are consistent with a two-state process and provide support for a radical rebound but not a hydroperoxide insertion mechanism for cytochrome P-450 hydroxylation.  相似文献

Depuration or elimination of diarrhetic shellfish toxins (DST) was followed for 73 days in 1- and 2-year-old mussels. The age groups also differed in size, providing a broad approach to studying the effect of the differences in physiology accompanying the differences in size. Content of DST was analysed both on groups and individual mussels. Environmental variables were measured to evaluate their effect on depuration.We found no significant differences in elimination rate of DST between 1- and 2-year-old mussels under natural conditions. This suggests that size and age do not affect the elimination rate of the DST. The present study is the first study on the effect of age and size on the elimination rate of algal toxins in bivalves. The natural variations in food levels and temperature were not found to affect the elimination rate of DST.The digestive gland weights in the 1-year-old mussels increased four times while the DST content per individual decreased eight times. This demonstrated that dilution of toxins due to tissue growth could have an important contribution to declines in toxin concentrations. Changes in tissue mass are affected by environmental variables via growth or starvation, and when such changes lead to concentration or dilution of toxins this does not reflect the accumulation or removal of toxins from the tissues. We hence suggest that when evaluating the actual elimination capacity of the mussels, as in the present study, the total content of toxins per individual should be used, rather than toxin concentrations.The 1-year-old mussels had faster growth compared to the 2-year-old mussels in both total soft tissue and digestive glands. The mechanism of DST elimination is still unknown. If this process involves metabolism of the toxins, one could expect the rates of elimination to follow overall metabolic rates. However, the results from the present study suggest that large differences in growth rates, which also include difference in feeding and metabolic rates, do not affect the elimination rate of DST. Our results support the assumption that the depuration rates cannot be accelerated, even in artificial systems, as a cost-effective way to solve the problem with toxic mussels for the industry.  相似文献

Studies were conducted in anesthetized paralyzed dogs to determine how the dynamic and proportional sensitivity of pulmonary stretch receptors change during lung inflation. The firing of each receptor was examined at multiple levels of static transpulmonary pressure and during multiple identical inflations at each of several rates. The averaged response of the receptor was computed and receptor activity related to transpulmonary pressure. On the basis of a quantitative criterion, employed to distinguish type I from type II receptors, the receptors could not be divided into distinct subpopulations. Thus all receptors were treated as coming from a single population. For all receptors we observed that their proportional sensitivity (increases in firing produced by increases in lung expansion at a constant rate of inflation) declined as the lung was inflated. In contrast, the dynamic sensitivity (increases in firing produced by increased rates of inflation at constant transpulmonary pressure) increased or remained relatively constant with increasing lung expansion. Thus, as inflation volume increases, the pulmonary stretch receptor acts increasingly as a rate receptor. The rate of inflation may have a more important role in control of the inspiratory duration than previously realized.  相似文献

It is assumed that certain drug receptors are so coupled with certain physiological receptors that stimulation of either receptor increases the sensitivity of the other. If the drug receptor suffers tolerance (i.e. slow desensitization) and if insensitivity of the drug receptor also makes the physiological receptor insensitive, then tolerance must be responsible for a physiological deficiency. This may be remedied by increased drug administration which will raise the sensitivity of the remaining physiological receptors so that a normal or near-normal physiological situation is achieved. Thus the organism is not only tolerant to the drug but also dependent on it. If such theoretical considerations apply to opiate receptors (as drug receptors) and to catecholamine receptors (as physiological receptors), then the theory predicts that acute morphine administration increases the sensitivity of dopamine receptors, that sympathetic stimulation decreases pain sensitivity, that opioid tolerance provokes increased catecholamine activity, that alpha-receptor stimulants attenuate and alpha-receptor antagonists exacerbate morphine abstinence, and that catecholaminergic inhibition results in increased morphine toxicity. All of these predictions have been verified experimentally.  相似文献

From foraging theory, generalist predators should increase consumption of prey if prey availability increases. Pulsed resource events introduce a large influx of prey to predators that may exhibit a functional response of increased consumption rate on, or specialization to, this abundant food resource. We predicted that coyotes (Canis latrans) would respond functionally to numerical increases of neonate white-tailed deer (Odocoileus virginianus) during the pulsed resource event of parturition. We used howl surveys and deer camera surveys with occupancy modeling to estimate densities for coyotes, adult deer, and fawns, respectively, in Upper Peninsula of Michigan, USA, 2009–2011. We estimated biomass of adult and fawn deer consumed by coyotes during 2 periods [fawn limited mobility period (LMP) and social mobility period (SMP)] in May–August each year. Coyote densities were 0.32 and 0.37/km2 for 2010–2011, respectively. Adult deer densities (3.7–3.9/km2) and fawn densities (0.6–1.3/km2) were similar across years. Overall, fawn hair occurrence in coyote scats was 2.3 times greater in LMP than SMP. Estimated consumption of fawns between periods (n = 157–880) by coyotes varied, suggesting a functional response, with increasing consumption of fawns relative to their availability. Coyotes, on average, consumed 2.2 times greater biomass of fawns than adults across years, and consumed 1.5 times greater fawn biomass, on average, during LMP than SMP. We suggest that consumption rates of coyotes is associated positively with increases in fawn density, and fawn consumption by coyotes follows predictions of optimal foraging theory during this pulsed resource event.  相似文献

The elimination of polyneuronal innervation (synapse elimination) that occurs following reinnervation was studied in sartorius muscles of adult Rana pipiens. The percentage of neuromuscular junctions that were polyneuronally innervated declined from 47% at 40–80 days after nerve crush to 22% at greater than 250 days after nerve crush. We measured the size, synaptic strength, and position of competing nerve terminals at identified dually innervated neuromuscular junctions at these two different periods of synapse elimination. Our goal was to determine if any of these parameters play a role in the competition between nerve terminals that ultimately results in the elimination of polyneuronal innervation. Our data support the hypothesis that polyneuronal innervation will persist if competing nerve terminals are of similar synaptic efficacies but will be eliminated if the competing terminals are of different synaptic efficacies. We also tested, but failed to find any evidence, that the spatial proximity of competing nerve terminals at the same synaptic site influences the elimination of polyneuronal innervation.  相似文献

We studied the effect of catheter position and flow rate on gas exchange during constant-flow ventilation (CFV) in eight anesthetized, paralyzed dogs. The distal tips of the insufflation catheters were positioned 0.5, 2.0, 3.5, and 5.0 cm from the tracheal carina. Flow rates were varied between 10 and 55 l/min and steady-state arterial blood gases were measured. At a given flow rate, arterial CO2 pressure (PaCO2) decreased as CFV was administered further into the lung up to a distance of 3.5 cm from the carina; there were no significant differences in PaCO2 at 3.5 and 5.0 cm. For a given catheter position, PaCO2 decreased with increasing flow rate up to a flow rate of 40 l/min. Further increases in flow rate had no significant effect on PaCO2. Arterial O2 pressure (PaO2) was relatively constant at all flow rates and catheter positions. We conclude that, up to a point, CO2 elimination can be improved by positioning the catheters further into the lung; advancing the catheters further than 3.5 cm from the carina may cause over-ventilation of specific lung regions resulting in a relative plateau in CO2 elimination and relatively constant PaO2's. Positioning the catheters further into the lung permits the use of lower flow rates, thus potentially minimizing the risk of barotrauma.  相似文献

The kinetics of drug elimination of interactive drug systems is stimulated by a set of differential equations based on mass balances, the mass of organs and blood flow rates. Experimentally determined concentration profiles of the drugs in the plasma and bile are used to evaluate clearance rate parameters. An example is shown in which the clearance of the anticoagulant warfarin is reduced to less than 50% of its normal rate due to the interference by BSP.  相似文献

The rates of germination of Bacillus subtilis spores with L-alanine were increased markedly, in particular at low L-alanine concentrations, by overexpression of the tricistronic gerA operon that encodes the spore's germinant receptor for L-alanine but not by overexpression of gerA operon homologs encoding receptors for other germinants. However, spores with elevated levels of the GerA proteins did not germinate more rapidly in a mixture of asparagine, glucose, fructose, and K(+) (AGFK), a germinant combination that requires the participation of at least the germinant receptors encoded by the tricistronic gerB and gerK operons. Overexpression of the gerB or gerK operon or both the gerB and gerK operons also did not stimulate spore germination in AGFK. Overexpression of a mutant gerB operon, termed gerB*, that encodes a receptor allowing spore germination in response to either D-alanine or L-asparagine also caused faster spore germination with these germinants, again with the largest enhancement of spore germination rates at lower germinant concentrations. However, the magnitudes of the increases in the germination rates with D-alanine or L-asparagine in spores overexpressing gerB* were well below the increases in the spore's levels of the GerBA protein. Germination of gerB* spores with D-alanine or L-asparagine did not require participation of the products of the gerK operon, but germination with these agents was decreased markedly in spores also overexpressing gerA. These findings suggest that (i) increases in the levels of germinant receptors that respond to single germinants can increase spore germination rates significantly; (ii) there is some maximum rate of spore germination above which stimulation of GerA operon receptors alone will not further increase the rate of spore germination, as action of some protein other than the germinant receptors can become rate limiting; (iii) while previous work has shown that the wild-type GerB and GerK receptors interact in some fashion to cause spore germination in AGFK, there also appears to be an additional component required for AGFK-triggered spore germination; (iv) activation of the GerB receptor with D-alanine or L-asparagine can trigger spore germination independently of the GerK receptor; and (v) it is likely that the different germinant receptors interact directly and/or compete with each other for some additional component needed for initiation of spore germination. We also found that very high levels of overexpression of the gerA or gerK operon (but not the gerB or gerB* operon) in the forespore blocked sporulation shortly after the engulfment stage, although sporulation appeared normal with the lower levels of gerA or gerK overexpression that were used to generate spores for analysis of rates of germination.  相似文献

Species diversity patterns are the product of diversification rate variation, but the factors influencing changes in diversification rates are poorly known. Radiation is thought to be the result of ecological opportunity: the right traits in the right environment at the right time. We test this in the Cape Floristic Region (CFR) of South Africa, in which pyrophytic heathland (fynbos) and non-pyrophytic Afromontane forest occur interdigitated. We infer transitions from forest to fynbos in three Cape clades (Penaeaceae, Phyliceae and Diosmeae) and test if they are associated with diversification rate shifts and the evolution of functional traits linked to fire, high insolation and seasonal drought. We estimate diversification rate shifts using maximum likelihood and use phylogenetic comparative methods to show that forest to fynbos shifts were associated with decreases in leaf area and specific leaf area and preceded or coincided with increases in diversification rates. Furthermore, we show that Penaeaceae, Phyliceae and Diosmeae species are typical members of their vegetation types in terms of their traits. The diversification rate shifts of Penaeaceae and Phyliceae are dated to the Miocene, when postulated aridification-driven changes in the CFR fire regimes may have triggered expansion of the fynbos at the cost of forest, providing an ecological opportunity for the diversification of fynbos lineages.  相似文献

In this paper, we propose a theoretical framework within which a unified treatment of the key sources of size-at-age variability—size dependence of growth rate, stochastic growth rate variations and individual-to-individual variability in growth performance—is possible. We use this framework to develop a general criterion for growth depensation in cohorts, which we define as the increase of the coefficient of variation of size-at-age, with increasing age. We use this criterion to show that size dependence of growth rate, acting alone, is depensatory only if the growth rate increases faster than linearly with size (that is, if growth is faster than exponential), while stochastic growth rate variation is invariably depensatory. Many species exhibit growth rates that scale less than linearly with size; indeed the commonly used von Bertalanffy model shows growth rates which actually decrease with size. In such a species, the size dependence of growth rate acts compensatorily, while stochastic growth rate variability is depensatory. We show that the tension between these two mechanisms leads to quasi-stationary size-at-age variability, which we can calculate analytically in some special cases and obtain by a simple numerical procedure where analysis is impractical.  相似文献

Insulin-like growth factor binding proteins (IGFBP) can inhibit or accentuate the mitogenic activities of insulin-like growth factor 1 (IGF-1) depending upon the experimental model employed. Inhibitory effects may be attributed to sequestration of IGF-1 onto IGFBP rather than the type I IGF receptor. We have demonstrated that the presence of IGFBP in a simple equilibrium binding assay significantly reduces the total amount of IGF-1 bound to the type I IGF receptor and increases the IC50 for IGF-1 binding. On the basis of such an experiment, performed at equilibrium, IGFBP should reduce the mitogenic activity of IGF-1. Recent work has demonstrated an inverse correlation between the dissociation rate of insulin-like molecules from their receptors and their mitogenic activity. It has also been suggested that the increased rate of dissociation of insulin and IGF-1 from their receptors at increased ligand concentrations serves as a ‘dampening’ mechanism to decrease mitogenic signalling. We have demonstrated increased rates of dissociation of IGF-1 from the type I IGF receptor with increasing concentrations of IGF-1. Furthermore, IGFBP-3 inhibits the acceleration of dissociation rates due to increased IGF-1 levels. Thus, under receptor saturating conditions IGFBP-3 may act to increase mitogenesis by increasing the residence time of individual molecules of IGF-1 upon the type I IGF receptor.  相似文献
