Using microscopy, the gastrovascular systems of four hydroids (Eirene viridula, Cordylophora lacustris, Hydractinia symbiolongicarpus, and Podocoryna carnea) and two distantly related alcyonacean octocorals (Acrossota amboinensis and Sarcothelia sp.) were examined and compared within a phylogenetic framework. Despite a range of stolon widths (means 53–160 μm), the hydroid species exhibited similar patterns of gastrovascular flow: sequentially bidirectional flow in the stolons, driven by myoepithelial contractions emanating from the center of the colony. Unlike the hydroids, the gastrovascular system of A. amboinensis (mean stolon widths for 5 colonies, 0.57–1.21 mm) exhibited simultaneously bidirectional flow with incomplete, medial baffles (width 4–20 μm) separating the flow. Baffles visualized with transmission electron microscopy consisted of endoderm, mesoglea, and occasionally another layer of tissue. Mean flow rates of the gastrovascular fluid for seven stolons ranged from 125 to 275 μm s?1, with maximum rates of 225–700 μm s?1. In Sarcothelia sp., stolons were of comparable width (means for 13 colonies 0.55–1.4 mm) to those of A. amboinensis. These stolons, however, were divided by several partitions (width 8–25 μm), both complete and incomplete, which were spaced every 100.5±5.1 μm (mean±SE; range 27.1–283.7 μm) and appeared structurally similar to baffles. In lanes defined by these partitions, ciliary motion was visible in image sequences, and flow was unidirectional. Within a single stolon, flow moved in different directions in different lanes and changed direction by moving from lane to lane via occasional spaces between the partitions. Mean flow rates for 30 stolons ranged from 75 to 475 μm s?1, with maximum rates of 85–775 μm s?1. For both octocorals, flow rates of the gastrovascular fluid were not correlated with the width of the stolon lumen. While octocoral gastrovascular systems probably exhibit differences based on phylogenetic affinities, in all species studied thus far, gastrovascular flow is entirely driven by cilia, in contrast to the hydroid taxa.  相似文献

More and more attention is paid to diseases such as internal transfer and brain malformation which are caused by the abnormal morphogenesis of cilia. These cilia-related diseases are divided into two categories: ciliopathy resulting from defects of primary cilia and primary ciliary dyskinesia (PCD) caused by functional dysregulation of motile cilia. Cilia are widely distributed, and their related diseases can cover many human organs and tissues. Recent studies prove that primary cilia play a key role in maintaining homeostasis in the cardiovascular system. However, molecular mechanisms of cilia-related diseases remain elusive. Here, we reviewed recent research progresses on characteristics, molecular mechanisms and treatment methods of ciliopathy and PCD. Our review is beneficial to the further research on the pathogenesis and treatment strategies of cilia-related diseases.  相似文献

Primary cilia are unique sensory organelles that coordinate a wide variety of different signaling pathways to control cellular processes during development and in tissue homeostasis. Defects in function or assembly of these antenna-like structures are therefore associated with a broad range of developmental disorders and diseases called ciliopathies. Recent studies have indicated a major role of different populations of cilia, including nodal and cardiac primary cilia, in coordinating heart development, and defects in these cilia are associated with congenital heart disease. Primary cilia play an essential role in modulating signaling cascades that shape cellular responses to environmental cues to maintain proper tissue development. Mutations in primary cilium proteins have been linked to several rare developmental disorders, collectively known as ciliopathies. Together with other disorders associated with dysfunctional cilia/centrosomes, affected individuals have increased risk of developing metabolic syndrome, neurologic disorders, and diabetes. In pancreatic tissues, cilia are found exclusively in islet and ductal cells where they play an essential role in pancreatic tissue organization. Their absence or disorganization leads to pancreatic duct abnormalities, acinar cell loss, polarity defects, and dysregulated insulin secretion. Cilia in pancreatic tissues are hubs for cellular signaling. Many signaling components, such as Hh, Notch, and Wnt, localize to pancreatic primary cilia and are necessary for proper development of pancreatic epithelium and β‐cell morphogenesis. Receptors for neuroendocrine hormones, such as Somatostatin Receptor 3, also localize to the cilium and may play a more direct role in controlling insulin secretion due to somatostatin's inhibitory function. Finally, unique calcium signaling, which is at the heart of β‐cell function, also occurs in primary cilia. Whereas voltage‐gated calcium channels trigger insulin secretion and serve a variety of homeostatic functions in β‐cells, transient receptor potential channels regulate calcium levels within the cilium that may serve as a feedback mechanism, regulating insulin secretion. The olfactory epithelium of two closely related species of snowtrout—Schizothoraichthys progastus McClelland andSchizothorax richardsonii Gray (Subfamily: Schizothoracinae. family-Cyprinidae, Teleostei), from a perennial glacier-fed river Mandakini of Garhwal hills was studied by employing transmission electron microscopic method The olfactory lamella comprises two epithelia—anterior and posterior, with a stroma sandwiched in between. Both are strartified. The anterior one is thicker than posterior. InSchizothoraichithys progastus, the sensory part of epithelium has two types of receptor cells—ciliated receptor cells and rod cells whilst inSchizothorax richardsonii, there arc three types of receptor cells—ciliated receptor cells, microvillous receptor cells and rod cells in addition to sustentacular cells, basal cells and mucous cells. While inhabiting similar hillstream habitat, the differential ecological niches, feeding habits etc., account for the presence of different receptor cell types in these species, It implies the possible diversification at cellular and physiological levels so as to minimize the competition by using varied olfactory cues.  相似文献

Abstract. SEM studies of 21 species of marine bryozoans demonstrated that the abfrontal side of the tentacles bears a row of mono- or multiciliated cells, which are presumably sensory. In stenolaemates, the abfrontal cells, as well as the cells at the tentacle tips and the laterofrontal cells, are monociliated. In the 17 gymnolaemate species studied, each tentacle tip bears at least 3 multiciliated cells, each with a tuft of 5–7 stiff cilia of various lengths. On the abfrontal tentacle surface, mono- and multiciliated cells alternate, but all species studied have multiciliated cells at the base and the tip of each tentacle. In live animals, single cilia perform occasional flicks, whereas the tufts of 7–15 cilia on the multiciliated cells are immotile. Length and number of abfrontal cilia vary between species. Two types of multiciliated, putative sensory organs were found on the introvert of some gymnolaemates. One has an apical knob surrounded by a ring of cilia; the other has an apical tuft of cilia. The ultrastructure of the sensory cells of tentacles and introvert was studied in Rhamphostomella ovata . Our observations on both fixed and living material all suggest that these cells are primitive mechanoreceptors. The few species lacking ciliary structures on the introvert have long proximal ciliary tufts on the abfrontal tentacle surface.  相似文献

The epidermal organs of an undescribed Phascolion species from the Balearic Islands were investigated using SEM, TEM, LM, CLSM and μCT methods. We found axial receptor cells confirming the previously assumed sensory function of epidermal organs. Our analyses also revealed six types of secretory cells. Some secretory cells types are capable of secreting filamentous and amorphous secretion in two different ways simultaneously (bimodal secretion). The high diversity of cell types, the complex pattern of acinar units, and the absence of a common gland pore make epidermal organs of Phascolion unique amongst sipunculans (Phascolion type). Our reconstruction of the evolution of the epidermal organs of Sipuncula revealed that Phascolion‐type epidermal organs may have derived from either Golfingia‐, Sipunculus‐ or Phascolosoma‐type epidermal organs. The oldest known sipunculans were Golfingia‐like and had epidermal organs, which might resemble the architecture of the Golfingia‐type epidermal organs in extant taxa. Thus, it can be hypothesized that bimodal secretory cells (e.g. basophilic secretory cells) were part of the sipunculan ground pattern. Moreover, bimodal secretory cells of Phascolion look strikingly similar to those found in various annelid glands and thus might even be part of the ground pattern of stem species of Sipuncula + Pleistoannelida.  相似文献

Cilia are highly specialized organelles that extend from the cell membrane and function as cellular signaling hubs. Thus, cilia formation and the trafficking of signaling molecules into cilia are essential cellular processes. TULP3 and Tubby (TUB) are members of the tubby-like protein (TULP) family that regulate the ciliary trafficking of G-protein coupled receptors, but the functions of the remaining TULPs (i.e., TULP1 and TULP2) remain unclear. Herein, we explore whether these four structurally similar TULPs share a molecular function in ciliary protein trafficking. We found that TULP3 and TUB, but not TULP1 or TULP2, can rescue the defective cilia formation observed in TULP3-knockout (KO) hTERT RPE-1 cells. TULP3 and TUB also fully rescue the defective ciliary localization of ARL13B, INPP5E, and GPR161 in TULP3 KO RPE-1 cells, while TULP1 and TULP2 only mediate partial rescues. Furthermore, loss of TULP3 results in abnormal IFT140 localization, which can be fully rescued by TUB and partially rescued by TULP1 and TULP2. TUB’s capacity for binding IFT-A is essential for its role in cilia formation and ciliary protein trafficking in RPE-1 cells, whereas its capacity for PIP2 binding is required for proper cilia length and IFT140 localization. Finally, chimeric TULP1 containing the IFT-A binding domain of TULP3 fully rescues ciliary protein trafficking, but not cilia formation. Together, these two TULP domains play distinct roles in ciliary protein trafficking but are insufficient for cilia formation in RPE-1 cells. In addition, TULP1 and TULP2 play other unknown molecular roles that should be addressed in the future.  相似文献

We have examined the subcellular localization of transient receptor potential (TRP) ion channels and the potential sensory role of cilia in murine female reproductive organs using confocal laser scanning microscopy analysis on ovary and oviduct tissue sections as well as on primary cultures of follicular granulosa cells. We show that the Ca2+ permeable cation channel, polycystin-2, as well as polycystin-1, a receptor that forms a functional protein complex with polycystin 2, distinctively localize to primary cilia emerging from granulosa cells of antral follicles in vivo and in vitro. Both polycystins are localized to motile oviduct cilia and this localization is greatly increased upon ovulatory gonadotropic stimulation. Further, the Ca2+ permeable cation channel, TRP vaniloid 4 (TRPV4), localizes to a sub-population of motile cilia on the epithelial cells of the ampulla and isthmus with high intensity in proximal invaginations of the epithelial folds. These observations are the first to demonstrate ciliary localization of TRP ion channels and their possible receptor function in the female reproductive organs. We suggest that polycystins 1 and 2 play an important role in granulosa cell differentiation and in development and maturation of ovarian follicles. In the oviduct both TRPV4 and polycystins could be important in relaying physiochemical changes in the oviduct upon ovulation.  相似文献

With few exceptions, the non-cycling cells in a vast range of animals including humans have a non-motile primary cilium that extends from the mother centriole of the pair of centrioles in their centrosomes located between their Golgi apparatuses and nuclei. It has very recently been shown that the primary cilium of a dog or a mouse embryonic kidney cell is a fluid flowmeter studded with heterodimeric complexes of mechanoreceptors linked to Ca(2+)-permeable cation channels that when the cilium is bent can send Ca(2+) signals into the cell and beyond to neighboring cells through gap junctions. More than 30 years ago, osteocytes were reported also to have primary cilia, but this was promptly ignored or forgotten. Osteocytes are the bones' strain sensors, which measure skeletal activity from the effects of currents of extracellular fluid caused by their bones being bent and squeezed during various activities such as walking and running. Since bending a kidney cell's primary cilium can send a Ca(2+) wave surging through itself and its neighbors, the bending of an osteocyte's primary cilium by sloshing extracellular fluid is likely to do the same thing and thus be involved in measuring and responding to bone strain.  相似文献

In the past half century, beginning with electron microscopic studies of 9 + 2 motile and 9 + 0 primary cilia, novel insights have been obtained regarding the structure and function of mammalian cilia. All cilia can now be viewed as sensory cellular antennae that coordinate a large number of cellular signaling pathways, sometimes coupling the signaling to ciliary motility or alternatively to cell division and differentiation. The primary cilium is a non‐motile cilium whose structure is 9+0. It is involved in co‐ordinating cellular signal transduction pathways, developmental processes and tissue homeostasis. Defects in the structure or function of the primary cilium underlie numerous human diseases, collectively termed ciliopathies. The presence of single cilia in the central nervous system (CNS) is well documented, including some choroid plexus cells, neural stem cells, neurons and astrocytes, but the presence of primary cilia in differentiated neurons of the enteric nervous system (ENS) has not yet been described in mammals to the best of our knowledge. The enteric nervous system closely resembles the central nervous system. In fact, the ultrastructure of the ENS is more similar to the CNS ultrastructure than to the rest of the peripheral nervous system. This research work describes for the first time the ultrastructural characteristics of the single cilium in neurons of rat duodenum myenteric plexus, and reviews the cilium function in the CNS to propose the possible role of cilia in the ENS cells.  相似文献

Summary Single primary cilia are found in developing as well as mature ciliated cells of guinea-pig tracheal epithelium. A few biciliated cells were observed, and in a rare case one cell had developed four such processes. Primary cilia are characterized by a 9 + 0 microtubular arrangement near the base, while a transition to an 8 + 1 pattern occurs at a slightly more distal position. Spokes are lacking, and dynein arms are absent or incompletely developed. The function, if any, of primary cilia remains unknown.In the population of the motile 9 + 2 cilia atypical forms are very rare, i.e. <0.1%. Of the various abnormalities cilia with supernumary microtubules are most common. Only one atypical basal body was observed. Although some of the aberrant forms undoubtedly are non-motile, their very low number suggests that they have no practical influence on the muco-ciliary clearance.Local extrusions of the ciliary membrane, here named ciliary knobs, are believed to be fixation artefacts. It is suggested that they represent circumscribed regions of the ciliary membrane which are sensitive to changes in the environmental osmotic pressure.  相似文献

Cilia from the protozoan Tetrahymena pyriformis were demembranated and then extracted for 5 min with a buffer containing 0.5 M NaCl. The briefly extracted axonemal pellet was then reextracted for about 20 hr. The soluble material obtained from each extraction was resolved into 14S and 30S dynein ATPases by sedimentation on sucrose density gradients and tested for sensitivity to added calmodulin. The 14S dynein obtained by a 5-min extraction was generally insensitive to added calmodulin, whereas that obtained by 20-hr extraction of the 5-min extracted axonemes was activated by calmodulin, the activation being much larger in the “light” 14S fractions than in the “heavy” fractions. The 30S dynein ATPase obtained by a 5-min extraction was generally activated over 1.6-fold by added calmodulin, whereas that obtained by the subsequent long extraction was usually activated only 1.3-fold. After further purification of the 5-min extracted 30S dynein and of the 5-min to 20-hr-extracted 14S dynein on DEAE-Sephacel, these dyneins retained much of their calmodulin activatability. The ATPase activity of both 14S and 30S dyneins was inhibited more strongly by erythro-9-[3-(2-hydroxynonyl)] adenine and by vanadate in the presence of added calmodulin than in its absence. These data suggest that the only ATPase activity present in the fractions studied is that of the dyneins and demonstrate that both the 14S and 30S dynein ATPases may be obtained in forms mat are activated by added calmodulin as well as in forms that are insensitive to added calmodulin.  相似文献

Rohde K. 1973. Ultrastructure of the protonephridial system of Polystomoides malayi Rohde and P. renschi Rohde (Monogenea : Polystomatidae). International Journal for Parasitology3: 329–333. Polystomoides malayi and P. renschi have three types of protonephridial flames. The first type is a typical flame cell with internal and external ribs connected by a weir membrane without nephrostomes, and with internal and external leptotriches. The second type is a flame cell complex consisting of at least two flames reaching into a common cavity. The third type is a non-terminal (= lateral) flame in the protonephridial ducts, consisting of loosely arranged cilia many of which have lateral tube-like extensions and whose tips have irregularly arranged filaments gradually decreasing in number. The number of cilia in all types of flames varies. The smallest capillaries are strongly convoluted and have a smooth or slightly reticulated surface, the larger ducts have strongly reticulated walls and single cilia may be found in the cavities of the reticulum.  相似文献
