药用植物红芽大戟的组织培养

摘    要:针对药用植物红芽大戟结实率和种子萌发率较低的情况,探讨了利用组织培养方法解决生产中繁殖的问题。结果表明,在茎、叶和芽3种外植体中,芽的出愈率最高,愈伤组织褐化率低,生长最好,且易被诱导出不定芽,是较理想的快速繁殖材料;而茎和叶的愈伤组织褐化率高,难以诱导出不定芽。较适宜的诱导愈伤组织的激素组合是6-BA2.0mg/L+NAA0.4mg/L,诱导不定芽的适宜激素组合是6-BA5.0mg/L+NAA0.05mg/L,根的诱导则在1/2MS+NAA0.2+庶糖1.5%+琼脂0.8%培养基上进行

Abstract:Because the fruiting rate and germinative percentage of Knoxia Valerianoides are lower, tissue culture was explored to solve the propagating problem in the production. The results show that the bud is the best material in speeding propagation among the three explants (stem, leaf and bud) as it has the biggest inductivity, the best growth, the lowest brown percentage and it is easy to be inducted adventitious buds. The brown percentage of the callus from stem and leaf is quite high and the callus is difficult to be inducted adventitious buds in our experiment. The suitable phytohormone compositions to induct callus, adventitious buds and roots are respectively 6-BA 2.0 mg/L+NAA 0.4 mg/L, 6-BA 5.0 mg/L+NAA 0.05 mg/L, 1/2 MS+NAA 0.2 mg/L+sugar 1.5%+agar 0.8%.
