摘    要:利用流式细胞光度术测定了苹果12个二倍体,5个三倍体细胞DNA含量。结果表明:二倍体细胞核DNA含量平均为2.27pg,三倍体细胞核DNA含量平均为3.13pg。

Abstract:Content of nuclear DNA for apple cultivars including 12 diploid and 5 triploid was measured by flow cytometry.The average nuclear DNA content of diploid cultivars was 2.27 pg and that of triploid cultivars was 3.13 pg,which indicated that variation of nuclear DNA content between diploid and triploid was significant.Nuclear DNA content of 6 colchicine induced plants were also measured.That these induced plants had two groups of cells which nuclear DNA content diverged almost one times,which suggested that these inducing plant were chimerals.The result shows that flow cytometry could be used for identifying ploidy.Compared to count chromosome number for identifying ploidy,using flow cytometry for identification of ploidy was ideal method because of simple preparation of sample,rapid and accurate measurement.
