百脉根愈伤组织原生质体再生植株

摘    要:百脉根无菌苗幼茎在含2.0mg/L-,2,4-D,0.1mg/L2-ip的MS培养基上诱导和继代培养愈伤组织。选取绿色松散颗粒愈伤组织分离原生质体。原生质体培养在调整珠KM8P,V-KM,MS和SH培养基上「含300mg/L,CH,2%CW,2%蔗糖,6%葡萄糖,2.0mg/L,2,4-D,0.5mgg/L,BA,5mmol/L MES」,原生质体再生细胞均能分裂,并形成小愈伤组织,但以KM80为

Abstract:Calli from young stems of aseptic young plants of Lotus corniculatus L.was induced and maintained on Murashige ans Skoog's medium containing 2. Omg/L 2, 4-D, 0.lmg/L 2-ip. Protoplasts which were isolated from friable,rasy dispersed green calli were cul-tured on modified KM8P,V-KM,MS and SH media supplemented with 300mg/L casein hydrolysate,2 % coconuk, 2% sucrose, 6% glucose, 2. Omg/L 2, 4-D, 0.5mg/L 6-benzyl-aminopurine,5mmol/L 2- (N-morpholino )-ethanesulfonic acid. Division of regenerated cells and small callus formation could be seen for the four media. KM8P was the best one of the four media.The frequency of cell colone formation was 3. 4% on this medium.Calli regenerated plants with the frequency of 100 %.
