菠萝种质的离体保存研究

摘    要:以菠萝品种巴厘"的组培苗为试验材料,在常温下通过改变MS培养基中无机盐和蔗糖含量,选择适合菠萝种质保存的培养基.对不同保存培养基中菠萝苗叶色的变化、小苗的生长及存活等进行了调查分析.结果显示,菠萝种质在M2(1/2MS 0.7%琼脂)中保存12个月存活率为92.2%,小苗的平均生长量只有1 cm左右;保存苗转入分化培养基后,很快恢复生长并分化出丛芽.在M2中保存18个月后仍有较高存活率(85.7%).表明M2是菠萝种质保存的最佳培养基.

Abstract:The aim of this study was to establish a suitable medium for the conservation of pineapple germplasm 'Bali' at norma1 temperature in vitro.The pineapple plantlets were conserved on five different media with different concentrations of the mineral nutrition and sucrose.During the conservation,the color of the leaves,the growth of plantlets and roots,the survival rate and so on were investigated.The results showed that survival rate of the pineapple germplasm in M2(1/2MS 0.7% agar)was 92.2% and the plantlets grow only 1 cm after conservation for 12 months.Plantlets were growing fast and multiple shoots were differentiated when transferred to the medium for differentiation.The survival rate of the pineapple germplasm in M2 was 85.7% after conservation for 18 months.So in this study,the medium M2 was the best one for the conservation of pineapple germplasm.
