BT型细胞质雄性不育水稻及其三系的线粒体DNA研究

摘    要:用RAPD技术对BT型水稻胞质雄性不育系秀A及其保持系秀B、恢复系湘晴以及杂种F1代的线粒体DNA进行了比较分析。结果表明不育系与其保持系间存在显著差异;不育系与其F1之间mtDNA也存在差异。在引物OPJ-08的扩增产物中,秀A扩增出一条分子量为800bp的多态性片段,在引物OPK-10的扩增产物中,杂种F1扩增出一条分子量为900bp的片段。把这两片段回收、克隆并制备探针,OPJ-08800的Southern杂交结果显示不育系与其F1杂交图谱存在多态性;OPK-10900的Suthern杂交结果显示不育系与其保持系同存在差异。推测这两片段与育性可能有一定的联系。

Abstract:BT type rice cytoplasmic male sterility line were compared with its maintainer,restorer and F 1 hybrid using mitochondrial DNA (mtDNA) RAPD analysis.The results showed that difference was between not only the cms line and its maintainer,but also cms line and its F 1 hybrid.Primer OPJ-08 generated a polymorphic product of 800 bp in sterile line,and primer OPK-10 amplified a specific fragment of 900 bp in F 1 hybrid.The two fragments were then cloned into plasmid vector and converted to RFLP probes.Southern hybridization patterns revealed polymorphism between sterile line and F 1 hybrid using OPJ-08 800 fragment as probe,and between sterile line and maintainer line using OPK-10 900 fragment as probe.We infered that the two mtDNA fragments may be associated with cms in rice.
