应用RT-PCR检测香蕉分化芽中黄瓜花叶病毒

摘    要:通过改进的异硫氰酸胍法提取香蕉分化芽中的总RNA,既经济又满足了RT-PCR的要求。运用RT-PCR两步法和一步法都可从带毒的香蕉分化芽提取的RNA中扩增出739 bp的特异性条带,而阴性对照未扩增出任何条带,并对RT-PCR一步法进行了优化及应用。本研究建立了经济简便的检测香蕉分化芽中黄瓜花叶病毒(cucumber mosaic virus,CMV)的RT-PCR方法,为灵敏高效的RT-PCR方法在香蕉分化芽检测中的应用奠定了基础。

Abstract:It is not only economic but also capable of meeting the requirements of RT-PCR to extract the total RNA from the in vitro differentiated shoots of banana by the modified Guanidine Thiocyanate method. A specific band of 739 bp could be amplified form the RNA extracted from the in vitro differentiated and CMV-infected shoots of banana by two-step RT PCR and one-step RT-PCR, while no bands were amplified from the negative control. At the same time, one-step RT PCR was optimized and put into use. The study presented in this paper established an economic and simple RT-PCR to detect CMV in the in vitro differentiated shoots of banana thereby laying down the basis for the application of sensitive and efficient RT-PCR to testing the in vitro differentiated shoots of banana.
