摘    要:本工作建立了CIC的PEG差示扫描谱线分析技术.其方法是在PEG沉淀比浊法基础上应用双光束分光光度计从600nm到240nm连续读取△OD值.得到一条差示扫描谱线.该谱线反应了CIC分子结构的差异,因而在一定程度上反映了疾病的特异性,具有较广泛的临床应用价值.


A NEW METHOD OF DETECTING CIRCURLATING IMMUNE COMPLEXES ANALYSIS OF PFG DIFFERENTIAL SCANNING SPECTRUM
Abstract:A new method of detecting Circulating Immune Complexes (CIC) has been established. The method is of both nonspecific and specific advantages and able to distinguish heterogeneity of CIC in sera from patients with diverse diseases by means of analyzing polyethylene Glycol Differential Scanning Spectrum (PEG-DSS).The method comes from conventional Polyethylene Glycol Turbidity Assay for detection of CIC. The conventional method is easy to perform and reproducible, but in want of specificity and unable to distinguish the heterogeneity of CIC. However, in accordance with the theory of molecular orbit, when several peptide chains combine, -electrons could move all over the macro- molecule. So the combination of PEG with CIC changes not only the turbidity of solution, but also the energy levels of molecular orbits that determine the absorb spectrum of the solution. Analysis of the spectrum could distinguish the heterogeneity of CIC.This paper reports the observations of PEG-DSS of sera from normal mice, mice bearing Ehrlich Ascites Carcinoma and S180 Sarcoma and from normal persons, patients with Gastric Carcinoma, Hepatoma and Leukemia.
