不同脂肪高脂日粮诱发胰岛素抵抗综合征大鼠模型的比较

摘    要:目的分析猪油、豆油、氢化椰子油、乳脂四种不同脂肪的高脂日粮分别诱发胰岛素抵抗综合征(IRS)大鼠的血液生化指标差异,为此类模型的建立及实验研究提供参考。方法雄性SD大鼠随机分为5组,对照组给予普通日粮,高脂组给予脂肪热量比相同的高脂日粮。喂养6周,每两周测定空腹血糖、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-c)、总胆固醇(TC)、胰岛素,根据胰岛素敏感性指数(ISI)=ln1/(FPG×FINS)评定大鼠的胰岛素敏感性。结果6周后,猪油组、乳脂组、豆油组血清胰岛素均显著高于对照组(P〈0.05);乳脂组血清TG显著高于其它高脂组(P〈0.05);高脂组血清HDL-c均显著低于对照组(P〈0.05)并以豆油组下降幅度最大;猪油组、乳脂组ISI显著低于对照组(P〈0.05);而各组间血清总胆固醇、血糖及体重无明显差异(P〉0.05)。结论4种高脂日粮诱发IRS大鼠模型的综合效果依次为乳脂、猪油、豆油、氢化椰子油。

Abstract:Objective To analyze the changes of blood biochemical parameters in rats with insulin resistant syndrome (IRS) induced by four types of dietary fat : lard, bean oil, hydrogenated-coconut oil and butterfat, respectively, to provide reference for establishment and related studies of this animal model. Methods Normal male SD rats were fed on standard laboratory chow (10.97 % of calories as fat) or isocaloric high fat diets (46.15 % of calories as fat) for 6 weeks. Serum glucose (GLU),insulin, triglyeride (TG), cholesterol (CHO) and HDL-cholesterol were measured fortnightly interval. The insulin sensitivity was estimated by ISI index, ISI = lnl/( FPG x FINS). Results After 6 weeks, the serum insulin was significantly increased in the lard, butterfat and bean oil groups compared to that in the normal group (P 〈 0.05). The serum TG was significantly higher in the butterfat group compared with that in other high-fat groups (P 〈 0.05). High-fat feeding groups exhibited significantly lower HDL- cholesterol ( P 〈 0.05 ) than that in the normal group, especially the bean oil group. As for ISI, butterfat and lard groups were both lower than the normal group ( P 〈 0.05 ). However, there were mild differences in TG, GLU and body weight among the five groups. Conclusion High butterfat chows have the best effect of the four high fat feeds on establisment of IRS rat model, and lard is better than bean oil, but hydrogenated-coconut oil is the worst.
