电针联合天麻多糖对脑缺血大鼠海马CA3区Nesin、BDNF表达的影响 缪化春

摘    要:目的 观察电针联合天麻多糖对脑缺血大鼠海马CA3区巢蛋白(Nestin)和脑源性神经营养因子(brain derivedneurotrophic factor,BDNF)表达的影响.方法 将40只SD大鼠随机分为正常对照组、模型组、电针组、天麻多糖组和针药结合组,每组8只.以单侧大脑中动脉栓塞法制备脑缺血模型.造模后2w,天麻多糖组和针药结合组大鼠给予天麻多糖100mg/kg灌胃,每天1次,连续2w;电针组和针药结合组大鼠给予“百会”“足三里”穴电针刺激,持续30 min,每天1次,连续2w.采用免疫组织化学染色法结合图像分析检测海马CA3区Nestin和BDNF的表达.结果 与正常对照组比较,模型组缺血侧海马CA3区Nestin和BDNF阳性表达增加(P<0.05);与模型组比较,电针组、天麻多糖组和针药结合组缺血侧海马CA3区Nestin和BDNF阳性表达显著增加(P<0.05);针药结合组阳性表达显著多于电针组或天麻多糖组(P<0.05).结论 电针与天麻多糖结合可显著增加脑缺血大鼠缺血侧海马CA3区Nestin和BDNF的表达,促进内源性神经干细胞激活,且作用优于单用电针或天麻多糖.

Abstract:Obieetive To observe the effects of electroacupuncture(EA) combined with polysaccha ride of Gastrodia data Blume (PGB) on the expressions of Nestin and brain derived neurotrophic factor (BDNF) in the hippocampal CA3 region of rats with cerebral ischemia. Methods A total of 40 Sprague Dawley rats were randomly divided into the normal control group, the model group, the EA group, the PGB group and the EA + PGB group, with 8 in each group. The model was prepared by middle cerebral artery occlusion (MCAO). Two week after modeling, PGB at 100mg g/kg was given to rats in the PGB group and the EA + PGB group by gastrogavage, once dialy, for two successive weeks. EA at “Baihui” (DU20) and “Zusanli” (ST36) was performed to rats in the EA group and the EA + PGB group, lasting for 30 rain, once daily for two successive weeks. The expressions of Nestin and BDNF were detected by im munohistochemical assay group. Results Compared with those of the normal control group, the expres sions of Nestin and BDNF in CA3 of the model group increased (P〈0. 05). Compared with the model group, the expressions of Nestin and BDNF in CA3 of the EA group, the PGB group and the EA + PGB group increased significantly (P〈0.05). The expressions of Nestin and BDNF were obviously higher in the EA + PGB group than those of the EA group or the PGB group (P〈0.05). Conclusion EA combined with PGB could significantly upregulate the expressions of Nestin and BDNF in the hippocampal CA3 region of rats with cerebral ischemia, and accelerate endogenous neural stem cells. Besides, their effects are signifi-cantly higher than those of PGB or EA alone.
