甘草酸二铵脂质配位体对非酒精性脂肪肝大鼠白细胞浸润的影响

摘    要:目的研究甘草酸二铵脂质配位体(DGLL)对非酒精性脂肪肝(NAFLD)大鼠白细胞浸润的影响及机制。方法用高脂乳剂灌胃诱导大鼠NAFLD模型并给予DGLL干预,6周后观察肝脏组织中髓过氧化物酶(MPO)的表达,酶标仪法测定肝脏组织MPO活性的变化,流式细胞技术测定外周血白细胞粘附分子表达的变化,Western blot技术检测肝脏组织内皮细胞间粘附分子-1(ICAM-1)的表达水平。结果与高脂模型组相比,DGLL能明显降低NAFLD大鼠肝脏组织中白细胞MPO的表达和活化,下调大鼠外周血中单核细胞和粒细胞粘附分子的表达,同时显著地抑制肝脏组织中内皮细胞ICAM-1的表达。结论 DGLL能显著降低高脂饮食诱导的NAFLD大鼠肝脏组织中白细胞的活化和浸润,这种作用可能与抑制白细胞与内皮细胞粘附分子的表达相关。

Abstract:Objective To study the therapeutic effects of diammonium glycyrrhizinate lipid ligand (DGLL) on leukocyte infiltration in rat liver with nonalcoholic fatty liver disease (NAFLD) and underlying mechanisms. Methods The rats of the model group were fed with high fat forage. At the same time, the rats of DGLL groups were administrated with relative dose of medicine. After six weeks, the myeloperoxi- dase (MPO) level and activity in liver tissues were measured by immunohistochemistry and ELISA, respectively. The expressions of leukocyte adhesion molecules in whole blood were determined by flow cytornetry. The expressions of intercellular adhesion molecule-1 (ICAM-1) in liver tissues was detected by Western blotting. Results Compared with NAFLD model groups, the expression and activity of MPO in liver tissues were both ameliorated remarkably by administration of DGLL. Meanwhile, the up-regulation of adhesion molecules expressed on monocytes and neutrophils in whole blood, as well as endothelial ICAM-1 expression were down-regulated by DGLL treatment. Conclusion DGLL ameliorated leukocyte activation and infiltration in rat liver with NAFLD, which might be related to suppression of adhesion mole- cules expression on leukocyte and endothelium.
