己酮可可碱对非酒精性脂肪性肝炎大鼠肝脏超微结构和酶组织化学的影响

摘    要:目的研究己酮可可碱(PTX)对非酒精性脂肪性肝炎(NASH)大鼠肝脏超微结构和酶组织化学的影响。方法高脂饮食建立大鼠非酒精性脂肪性肝炎模型。取SD大鼠40只,分为对照组、12w模型组、16w模型组和治疗组,每组10只。腹主动脉采血,测ALT、AST、血糖等水平。取肝组织做电镜和SDH、CCO、ATPase、LDH的酶组织化学染色。结果两个模型组均比对照组ALT、AST、血糖升高,LDL降低(P<0.05)。治疗组血糖、AST比16w模型组降低(P<0.05)。电镜显示16w模型组线粒体肿大,嵴排列紊乱,基质密度降低。治疗组结构改善。酶组织化学显示四种酶的活性16w模型组均较对照组降低(P<0.05),治疗组活性均较16w模型组升高(P<0.05)。结论NASH时存在肝细胞能量代谢障碍,经PTX治疗后能量代谢障碍得到改善。

Abstract:Objective To study the effects of pentoxifylline(PTX) on ultrastructure and enzyme histochemistry of nonalcoholic steatohepatisis(NASH) in rats. Methods Rat model of NASH was established by giving a fat-rich diet. These rats were randomly divided into four groups: control group(n=10),12w model group(n=10), 16w model group(n=10) and treatment group(n=10). The two model groups and the treatment group was given fat-rich diet while the normal group was given normal diet. After 12 weeks, the 12w model group were sacrificed and the treatment group were given PTX (16mg kg-1 d-1).After 16 weeks, the rats of three groups were sacrificed. The body weight, liver weight, liver index(liver/body weight ratio), and serum activities of liver alanine aminotransferase(ALT),aspartate aminotransferase(AST), blood glucose(GLU),total cholesterol(TC), triglycerid (TG), high density lipoprotein(HDL) and low density lipoprotein(LDL) of all rats were measured. The liver histology, enzyme histochemistry and ultrastructure were analyzed. Results The body weight and the liver index of the 16w model group increased significantly than the control group(P<0.05) while the treatment group decreased significantly than the 16w model group(P<0.05). ALT,AST and GLU in two model groups increased significantly while LDL decreases significantly than the control group (P<0.05). TC in the 16w model group increased significantly than the control group (P<0.05). AST and GLU in the treatment group were decreased significantly than the 16w model group (P<0.05). The change of mitochondria swelling, disordered cristae could be observed in 16w model group while the change of the treatment group were alleviated. The activities of SDH, CCO, ATPase and LDH were decreased significantly in the two model groups as compared to the control group and elevated significantly in the treatment group as compared to two model groups (P<0.05). Conclusion PTX can play a role in the treatment of NASH by relieving the disorder of energy meuabolism.
