摘    要:采用mRNA差异显示技术对黄伞菌丝体、原基、菇蕾、子实体菌柄和子实体菌盖进行研究,获得了在生殖生长阶段差异表达的4条cDNA片段T_(11)G B0304、T_(11)G B0304-2、T_(11)G B0322与T_(11)C B0310,经过回收、重扩增、克隆与鉴定后进行测序分析,结果发现T_(11)G B0322的序列在GenBank中与氧类固醇结合蛋白(oxysterol-binding protein)基因序列的同源性达到41%。再经过半定量PCR方法验证,表明T_(11)G B0322基因片段是在生殖生长阶段差异表达的基因片段。

Abstract:RNA of different development periods of pholiota adiposa including mycelium,anlage,bud and fruit body(stem and pileus of mushroom)was extracted and treated by RT-PCR.Four fragments(T_(11)G B0304,T_(11)G B0304-2,T_(11)G B0322 and T_(11)C B0310)were isolated,reamplified,purified,cloned and sequenced.After blasting in GenBank,T_(11)G B0322 cDNA fragment was identified to be similar to oxysterol-binding protein.The result of semiquantitive PCR showed that the expression of the T_(11)G B0322 cDNA fragment was different in different periods.This showed that gene fragment might be associated with differentiation of Ph.adiposa.
