摘    要:以柳杉种子在无菌条件下萌发的幼苗为外植体,研究3种基本培养基和3种植物生长调节剂对不定芽诱导、增殖及生根的影响,建立了完整的组培快繁体系。结果表明,柳杉种子经预处理后,用75%酒精消毒20 s,再用0.1%HgCl2浸泡600 s的消毒效果较好,污染率为10.00%,成活率为82.33%;丛生芽诱导的最适培养基配方为DCR+6-BA 0.2 mg·L-1,诱导率较高,达66.67%;丛生芽增殖的最适培养基配方为DCR+6-BA 0.2 mg·L-1+IBA 0.05 mg·L-1,增殖倍数达到11.00;适宜的生根培养基为DCR+NAA 0.01 mg·L-1+IBA 0.2 mg·L-1,生根率为90.00%,平均生根数为4.09。

Establishment of Rapid Propagation for Sterile Seedlings of Cryptomeriafortunei by Tissue Culture

Abstract:The seedlings sprouted from Cryptomeriafortunei's seeds under sterile conditions acted as explants. The effects of 3 types of basic mediums and plant growth regulators on buds induction, multiplication and root formation were studied and the rapid propagation technique by tissue culture was established. The results showed that rapid immersed with 75% alcohol for 20 s plus 0.1% HgCl2 for 600 s was the best disinfection method for C fortunei's seeds after pretreatment with the contamination rate of 10.00% and the survival rate was 82.33%; the best medium for the buds induction was DCR+6-BA 0.2mg·L-1 with the induction rate of up to 66.67%; DCR+6-BA 0.2mg·L-1+IBA 0.05mg·L-1 was the most suitable for buds multiplication while value ratio reaches 11.00. The optimum for rooting which rooting rate achieved 90.00% was DCR+NAA 0.01 mg·L-1+IBA 0.2 mg·L-1 and the average number of roots was 4.09.
