聚磷茵JPA1菌株的生长特性和聚磷条件的优化

摘    要:以YG培养基为基础,考察不同条件对聚磷菌JPA1菌株生长及聚磷效率的影响。结果表明:JPA1菌株生长的对数期为3-9h,9h后进入稳定期,12h到达衰亡期。在单因素试验中,麦芽糖为JPA1菌株最适生长和聚磷的C源,最适生长温度为30-35℃,pH为8,Na+、Ca2+、Mg2+、Mn2+和K+等离子有利于JPA1菌株的生长;最适聚磷温度为30-35℃,pH为7,Fe2+、Na2+、Mg2+、Mn“和K+等离子有利于JPA1菌株的聚磷。正交试验结果表明:JPA1菌株去磷培养基的最优组合为麦芽糖1.5g/L,温度37℃,pH6.5,装液量50mL(300mL摇瓶);菌株最适扩增培养基为麦芽糖1.5g/L,温度37℃,pH6.5或7.5,装液量150mL(300mL摇瓶)。

Abstract:Based on the YG media, effects of different conditions to the growth and phosphorus accumula- tion rate of JPA1 strain were discussed. Result showed that the logarithmic phase of JPA1 was from 3 h to 9 h, it turned into stationary phase after 9 h, and then entered the state of death phase. Single factor ex- periment showed that maltose was the optimal carbon source for JPA1. The optimal growth temperature was 30 ℃ to 35 ℃. The best pH was 8. Metal ions of Na + , Ca2 + , Mg2 + , Mn2 + , and K + were benefical to the growth of JPA1. The optimal phosphorus accumulation temperature was among 30 ℃ to 35 ℃, op- timal phosphorus accumulation pH was 7, Metal ions of Fe2+ , Na + , Mg2+ , Mn2+ , and K + were good for phosphorus accumulation. Based on the YG media, using maltose to replace glucose, orthogonal experi- ment results showed that the best phosphorus accumulation condition was maltose 1.5 g/L, 37 ℃, pH 6.5, volume of liquid 50 mL ( shake flask culture in 300 mL) ; the best condition of culture was maltose 1.5 g/L, 37 ℃, pH 6.5 or 7.5, volume of 150 mL (shake flask culture in 300 mL).
