摘    要:热水提取猪肚菇粗多糖中的酸性多糖WPG1,经凝胶纯化后用高效液相色谱(HPLC)测定其相对分子质量,用气相色谱对其单糖组成进行分析,用原子力显微镜(AFM)对其形态结构进行观测。结果表明:酸性多糖WPG1经SephacrylS-300凝胶层析纯化后得到WPG1-1和WPG1-2这2个组分; 经HPLC分析可知WPG1-1与WPG1-2均为单一组分,其相对分子质量分别为1.7×106和1.6×104; WPG1-1与WPG1-2的单糖组成主要是糖醛酸、甘露糖、葡萄糖和半乳糖; 原子力显微镜观测图分析表明WPG1-1为网链状聚集体结构,WPG1-2呈梭状聚集体,两者均非单链存在。

Abstract:An acidic polysaccharide WPG1 was extracted from Panus giganteus by hot water. Its molecular weight was measured by using high performance liquid chromatography and monosaccharides composition was analyzed by gas chromatography (GC), a atomic force microscopy (AFM) was used to analyze its morphological structure. The acidic polysaccharide WPG1 divided into two components, called the WPG1- 1 and WPG1-2 after Sephacryl S-300 gel chromatography purification, WPGI-1 and WPG1-2 were both simple components after analyzed by high performance liquid chromatography, their molecular weights were 1.7 × 106 and 1.6 × 104, respectively. Uronic acid, Man, Glu, and Gal were determined to be the components of WPGI-1 and WPG1-2. It was found that WPGI-1 on mica formed reticulate body aggregates by AFM,while WPG1-2 existed in the form of fusiform aggregates structure. The results showed that they were not single strand.
