激光照射对角膜蛋白质二级结构影响的光谱分析

摘    要:为了研究激光照射后角膜蛋白质化学组成的改变,探讨激光角膜损伤的发生机制,将日本大耳白兔随机分为正常对照组和激光损伤组,选取角膜上皮层和基质层作为研究对象,通过傅里叶变换红外光谱技术(Fou-rier transform infrared spectroscopy,FT-IR)对角膜组织酰胺I带中蛋白质二级结构各吸收峰进行定量分析,观察激光照射后蛋白质分子结构的改变情况。结果显示,激光照射后出现蛋白质二级结构吸收峰的位移和积分百分比的改变。激光照射可使角膜上皮层和基质层蛋白质构象发生改变,从而导致蛋白质结构稳定性下降和蛋白质生物功能的破坏。

Abstract:To study the effects of laser on animal corneal proteins structure and explore the molecular mechanism of la- ser induced cornea injury, the rabbits were divided randomly into injury group and normal group, and fourier transform infrared spectroscopy (FT-IR) was applied to observe the change of proteins structure in the corneal epithelium and stro- ma. It reveals that there are significant spectral differences of the peak position and the percentages of absorption value for secondary protein structure after laser exposure. These results suggest that the secondary structure of corneal epitheli- um and stroma proteins have changed since the laser irradiation, which may lead to the decrease in proteins structural stability and the loss of proteins function.
