摘    要:目的:建立表达MICA* 008和MICA* 001蛋白的人包皮成纤维(human foreskin fibroblast,HFF)细胞,为研究MICA基因的免疫功能提供生物学材料。方法:组织块法原代培养HFF细胞;用脂质体将质粒pLXSN,pLXSN-MICA* 008和pLXSN-MICA* 001转染PA317包装细胞后收集病毒颗粒,再将病毒颗粒感染HFF细胞,G418筛选14天,扩增耐药细胞,westernblot检测MICA* 008和MICA* 001蛋白表达情况。结果:逆转录病毒载体pLXSN,pLXSN-MICA* 008和pLXSN-MICA* 001经包装细胞PA317包装可产生有感染能力的病毒,该病毒感染HFF细胞后,westernblot检测出外源蛋白MICA* 008和MICA* 001。结论:建立了表达MICA* 008和MICA* 001蛋白的人成纤维细胞。

Abstract:Objective : Construct human foreskin fibroblast (HFF) cell expression protein MICA * 008, MICA * 001 to provide biotic material to research immune function of gene MICA. Methods : Cell culture in vitro technique was used ; The retroviral vector pLXSN, pLXSN-MICA * 008 and pLXSN-MICA * 001 transfect packaging cell PA317 with lipo- somes to gain virus particles. HFF cells are infected by virus particles and screened with G418 resistance for 14 days. The expression protein MICA * 008, MICA * 001 were dentified by westerublot. Results:Packaged by packaging cell, the ret- roviral vector pLXSN, pLXSN-MICA * 008 and pLXSN-MICA * 001 form virus particles with infection ability which are able to infect HFF cell and protein MICA * 008, MICA * 001 was expressed in HFF cell indicated by westernblot. Conclusion: HFF cell expression protein MICA * 008, MICA * 001 was constructed successfully.
