微型离心柱法快速分离纯化神经节苷脂

摘    要:神经节苷脂的分离与纯化是研究组织细胞Gls组成、含量及代谢的基本手段.但是以往的方法周期长、溶剂用量大,为此建立了一种新的微型离心柱快速分离Gls纯化法,标准Gls的回收率为97.64%,n=7,CV<3%,HPTLC图谱显示无丢带现象.纯化样品只需十几个小时,操作简单,溶剂用量小,重复性好,为微量样品组分的研究提供了一个有效的手段.

Abstract:Asimpleandrapidmethodforisola tionandpurificationofgangliosides (Gls)from theratbrainisdescribed .Thegangliosideswere isolatedandpurifiedbycentrifugalmini column chromatography.Therateofrecoveryofstan dardGlswasupto 97 6 4% ,n =7,CV <3% . NodetectivelossofGlswasdemonstrated .The novelmethodoffersafewadvantages:1 Itis simple,rapidandefficient.2 Thethinlayer chromatographicpatternsoftotalGlsofsamples obtainedareclearanddistinct.3 Lesssolvent wasconsumed .Thus,themethodisespecially applicabletocomparativeandqualitativestudies ofgangliosidesofmicrosamples,andtoganlio sidemetabolismresearchoftissuesandcellsin vitro.
