| 摘 要： | 凝胶过滤色谱因其操作方法简单，重复性强等特点在生物大分子的分离纯化中被广泛使用。特别在基因工程蛋白质类药物的精细纯化中起着难以替代的作用。但由于蛋白质凝胶过滤色谱要求样品上样体积较小及洗脱流速较小时才能获得理想的分辨率，这使得凝胶过滤色谱经常成为蛋白质大规模纯化工艺中的限速步骤。本介绍一种改进的上样方法，在不降低分辨率的前提下，可将单位时间内凝胶过滤柱的产量提高两倍。

| Abstract: | Gelfiltrationchromatographyhasoccupieda keypositioninthepurificationof macromoleculesforitssimplicity andhighreproducibility,especiallyinthepolishingpurificationstepof geneengineeringproteindrugs.In orderto achievea satisfactoryseparation,itis alwaysessentialto maintainlowsamplevolumeandlowelutionflowrateinproteingelfiltration chromatography,so gelfiltrationis usuallythesloweststepin large-scaleproteinpurificationprocedure.In thisarticle,an improvementof sampleapplicationmethodis introducedto increasetheproductivityof gelfiltrationby two-foldwithoutde-creasingtheresolutionof separation. |
