摘    要:目的:合成α-芋螺毒素AnIBA11G]荧光标记物,并用于测定烟碱型乙酰胆碱受体(nAChR)α3β2在小鼠脑内的分布。方法:利用两步氧化法设计合成α3β2高选择性抑制剂AnIB突变体AnIBA11G],纯化后,用5-羧基四甲基罗丹明琥珀酰亚胺酯(5-TAMRA-SE)标记,标记物鞘内注射到小鼠脑内,用激光扫描共聚焦显微镜测定α3β2在小鼠脑内的分布。结果与结论:获得了单一5-TAMRA-AnIBA11G]荧光标记物,测定表明小鼠脑内nAChRα3β2主要分布于海马区域。

Abstract:Objective: To synthesize fluorescent analogs of α-conotoxin AnlB AllG] and determine the distribution of nAChR α3β2 in mouse brain. Methods: AnlBAllG], which target nAChR α3β2 subtype, was synthesized by the two-step oxidation and labeled with 5-TAMRA-SE in the the N-terminus. After purification by HPLC, 5-TAMRA-AnlBAllG] was injected to the mouse brain and the distribution of nAChR α3β2 was determined with confocol laser scanning microscopy. Results & Conclusion: The pure 5-TAMRA-SE fluorescent analog of α-eonotoxin AnlB AllG] was synthesized and the nAChR α3β2 in mice brain is mainly distributed on hip- pocampus.
