摘    要:目的:将重组人内皮单核细胞活化多肽Ⅱ原(pro-EMAPⅡ)作用于Saos-2细胞,研究后者相关基因表达水平的变化。方法:在大肠杆菌中表达重组人pro-EMAPⅡ,并纯化获得目的蛋白;利用基因芯片技术研究重组人pro-EMAPⅡ对Saos-2细胞基因的表达调控。结果:获得重组人pro-EMAPⅡ;基因表达芯片分析表明,重组人pro-EMAPⅡ作用于Saos-2细胞后,后者有39个基因表达上调,29个基因表达下调,在这68个基因中有12个差异基因参与多个信号通路。结论:重组人pro-EMAPⅡ作用于Saos-2细胞,使其多个基因表达水平发生改变,有助于揭示pro-EMAPⅡ的调控机制及信号通路。

Abstract:Objective:To investigate the variation of the related gene expression in Saos-2 cells induced by recombinant human pro-endothelial monocyte-activating polypeptideⅡ(pro-EMAPⅡ).Methods:The recombinant human pro-EMAPⅡ(rhpro-EMAPⅡ) was exprssed in Escherichia coli,and then was purified.The regulation of gene expression in Saos-2 cells induced by rhpro-EMAPⅡ was analyzed by gene chips.Results:rhpro-EMAPⅡ was received,and the analysis re-sults of gene chips revealed that there were 39 genes up-regulated and 29 genes down-regulated in Saos-2 cells. Among these genes there were 12 differentially expressed genes participating in cell signal pathway. Conclusion: rhpro-EMAPⅡ changes the levels in expression of several genes in Saos-2 cells, and it is helpful to reveal the regulation mechanism and signal pathway of pro-EMAPⅡ.
