摘    要:随首微生物核糖体数据库的日益完善,16S rRNA序列分析技术已应用于海洋、湖泊和土壤等环境微生物多样性的分析,但尚未见其在大气微生物菌群分析中的应用报道。本研究选择5株大气中采集分离的菌株,通过细胞16S rRNA通过引物PCR扩增其对应序列,直接对PCR产物进行测序,分析鉴定其对应细胞的种属,并将该结果同细胞表型鉴定、全自动微生物分析仪以及相色谱分析结果加以比较。结果表明16S rRNA序列分

Abstract:The rapid increasing data of microbial small subunit rRNA sequences make it possible to use 16S rRNA sequence analysis for study the diversity of microbial populations in the natural environment such as marine, lake and soil. However, there is no report of using it to study bacteria in the atmosphere as far as I know. The genome DNA of 5 strains bacteria sampled and isolated from atmosphere are amplified using the primers targeted to 16S rRNA sequence. Sequences of the PCR products without cloning are analyzed to identify the identity of the correspondent strains. The results are compared with those from morphological characteristics and gas chromotography analysis. It reveals that the three methods give essentially the same results. The 16S rRNA method provides the advantages of accuracy, simplicity and independence of the growth status of the bacteria. It may be a powerful alternative analysis method for the study of atmospheric bacteria.
