| 摘 要： | 用人肝癌细胞系SMMC7721免疫小鼠，提取脾细胞总RNA，构建单链抗体库，从中筛选到1个与人肝癌细胞系HepG2特异性结合的噬菌体－单链抗体。此单链抗体与HepG2细胞结合滴度比正常肝细胞低100倍以上。构建表达质粒pTrx-scFv5-56，单链抗体scFv5-56在大肠杆菌BL21（DE3）中成功地进行了可溶性表达。

| Abstract: | The recombinant phage antibody system was used to construct a single-chain Fv fragment (scFv)cDNA library from the totol RNA of mice immunized with cell line SMMC7721.a phage-scFv named5-56which can bind HepG2cells specifically was selected from the library;the titer of scFv5-56for binding HepG2cells was more than100times lower than for binding the normal hepatocyte HL02.By constructing the expression plasmid pTrx-scFv5-56,scFv5-56was expressed as soluble form in E.coli BL21(DE3). |
