Abstract:Seventeen monoclonal antibodies ( McAbs ) specific for the nucleocapsid protein ( NP ) of epidemic hemorrhagic fever virus ( EHFV ) strains L99 and C4 have been obtained. Group-specific and type-specific McAbs were identified by antigenic reactivity analysis. It was demonstrated that there were not only group-specific but also type-specific antigenic determinants on the NP of EHFV. An analysis of the antigenic epitopes of strains L99 and C4 NP was carried out by competitive ELISA using these McAbs and NP purified by affinity chromatography as antigens. At least seven. epitopes common to both the Rattus-type and Apodemus-type exist. In addition, two epitopes specific for the Rattus-type virus and three specific for the Apodemus-type virus were defined on the NP of both strains. Group specific antigenic epitopes common to both types were distributed in two major regions, its conformation and topological distribution being some what different for different EHFV strains. The type-specific antigenic epitopes of either the Rattus-type virus or the Apodemus-type virus were localized in a distinct region.
