急性弛缓性麻痹病例中ECHO11病毒的基因特征分析

摘    要:该文首次分析了我国ECHO11病毒的分子流行病学资料。1999~2004年,ECHO11病毒是山东省急性弛缓性麻痹(AFP)病例中分离到的优势毒株,2003年从山东省482例AFP病例中共分离到11株ECHO11病毒,其中相关的10例病例分布跨越山东省大部分地区,但发病日期集中在7月和12月。该研究试图通过对VP1编码基因全序列的测定和分析,为探讨ECHO11病毒与AFP之间的病因关系提供线索。分子流行病学研究提示,11株E-CHO11毒株都位于同一传播链,核苷酸同源性为97.2%~100%,氨基酸同源性为99.6%~100%,其中7月和12月的分离株之间相差8~9个核苷酸,氨基酸序列一致。这说明山东省2003年7月和12月分别发生了ECHO11病毒流行,但这些毒株与AFP的病因关系尚需进一步研究。11株病毒组成了A基因型中的一个新亚型,在进化树上单独呈密切相关的一簇,与同基因型内的其它亚型的核苷酸同源性为82.2%~84.7%,氨基酸同源性为94.8%~97.6%。

Abstract:This research firstly reported the molecular data of ECHO11(Echovirus type 11) in China.During 1999-2004,ECHO11 was the most frequently isolated non-polio enterovirus from AFP cases in Shandong Province.10 of the 11 ECHO11 strains isolated in 2003 were in July and December,distributing across the most parts of Shangdong Province.We sequenced the complete VP1 coding region of these 11 ECHO11 strains,and the results suggested that all these strains belong to a single transmission chain,the nucleotide sequence identity among them was 97.2%-100%,and that of the amino acid sequence was 99.6%-100.0%.There were 8-9 nucleotide differences among the strains isolated in July and in December,but the amino acid sequences were identical.Thess indicated that there were two ECHO11 epidemic in Shandong in 2003,in July and in December individually,but how these strains associated with AFP was yet uncertain.All the 11 ECHO11 strains belong to a new sub-genotype of genogroup A.The nucleotide and amino acid sequence identities among them and the other subgenotypes were 82.2%-84.7% and 94.8%-97.6% respectively.
