Abstract:
A protein-DNA complex which is stable in concentrated salt solutions and in the presence of sodium dodecyl sulfate has been extracted from purified viruses and is found in the nicked circular DNA fraction. The protein is visualized as a “dot” on the DNA molecule by electron microscopy using a modified version of the ethidium bromide mounting technique. The position of the dot is at 0.67 genome units clockwise from the ecoRI restriction site on the SV40 DNA map.
