植物绿叶粗蛋白在SDS—PAGE中向负极泳动的富含苯丙氨酸的碱性蛋白的测定

摘    要:提取菜心(BrassicacampestrisL.ssp.chinensis(L.)Makinovar.utillisTsenetLee)和生菜(LactucasativaL.)绿叶粗蛋白,经SDS-变性后作醋酸纤维素薄膜电泳,证实这两种蔬菜含有分别向负极泳动和向正极泳动的蛋白。菜心绿叶粗蛋白经SDS-PAGE电泳后,从负极电泳缓冲液中获得向负极泳动的蛋白,其苯丙氨酸含量为57.05%,碱性/酸性氨基酸残基的比例为3.04,表明是富含苯丙氨酸的碱性蛋白。

Abstract:Crude proteins from green leaves of Brassica campestris L.ssp.chinensis(L.) Makino var.utillis Tsen et Lee and Lactuca sativa L.were subjected to acetate cellulose membrane electrophoresis under SDS-denatured conditions.Two groups of protein were observed,one migrated towards the anode,and the other,the cathode.The cathode protein from Brassica campestris ssp.chinensis var.utillis was collected from cathode reservoir after SDS-PAGE,and its amino acid components were determined to have 57.0% phenylalanine,the ratio of basic/ acidic amino acid residues was 3.04,indicating the protein towards cathode was a phenylalanine rich basic protein.It must be emphasized that the protein migrated towards the cathode had never been reported previously.
