麦迪霉素产生菌酮基还原酶基因在大肠杆菌中的表达

摘    要:用PCR法扩增麦迪霉素产生菌酮基还原酶(MKR)基因,得到约0.8kb的DNA片段,扩增片段重组到利用依赖T7RNA聚合酶的高效表达载体pT7-7中,在大肠杆菌中表达出28.9kD的蛋白质。表达的蛋白质具有生物活性。

Abstract:Two primers were designed and synthesized accoding to the polyketide ketoreductase from widecamycin producing strain gene(MKR) sequence data which has been reported, MKR gene was amplified using PCR technique. The amplified MKR gene was subeloned into Ndel and BamHI sites of the T7RNA polymerase-dependent pT7-7 expression vector, and introduced into E coli K38 / pGP1-2. Proteins were isolated from transformant The result of exclusive labelingby L-35S-memionine of plasmid-encoded proteins SDS-PAGE analysis showed that the MKR was produced in E. coli. The expessed MKR has bioachvity.
