TDX缓冲溶液浓度对血药浓度测定的影响

摘    要:目的考察在使用TDX进行血药浓度测定时缓冲溶液浓度对测定结果的影响;进一步分析缓冲溶液中γ-球蛋白成分在测定过程中的具体作用。方法利用原有的缓冲溶液及蒸馏水配制所需浓度的缓冲溶液,采用荧光偏振免疫法(TDX)分别在已配制的100%、75%、50%、25%和0%(蒸馏水)5个浓度的相同缓冲溶液条件下对同一种抗癫痫药物(卡马西平)血药浓度进行测定。结果在100%、75%、50%3个浓度的缓冲溶液条件下质控仍然在规定的范围内,经分析,75%浓度缓冲溶液的条件下血药浓度测定结果与原缓冲液测定结果差异无显著性。在25%和0%(蒸馏水)浓度的缓冲溶液条件下质控超出规定的范围,但仍具有可测性。结论在75%浓度的缓冲溶液条件下可以进行血药浓度的测定,测定结果有效可信,在应急的环境下可以使用75%浓度的缓冲溶液替代原液进行测定。除外特殊情况下,使用100%浓度的缓冲溶液进行测定为佳。

Abstract:Objective To study the effect of buffer concentration in Carbamazepine plasma sample determined by TDX(FPIA).Method Dilute the original concentration of the buffer solution by distilled water into 100%,75%,50%,25% and 0%.The carbamazepine plasma concentration was determined under different concentration of the buffer.Significance test was conduced.Result Compared with the group 100%,Carbamazepine concentration in group 75% had no significant difference while those in groups 50%,25% and 0% did.Conclusion While 75% buffer could be used for emergency cases,it is better to use the original buffer in determination of carbamazepine plasma concentration.
