双歧杆菌对哮喘儿童DC表达CD86和HLA-DR的影响

摘    要:目的研究双歧杆菌对过敏性哮喘儿童外周血单个核细胞来源的树突状细胞(DC)表面表达CD86和HLA-DR的影响。方法从12例过敏性哮喘儿童和10例对照组儿童的外周血单个核细胞诱导生成未成熟DC,分别经双歧杆菌或细菌脂多糖(LPS)处理,用流式细胞仪检测各组DC表面CD86和HLA-DR分子表达。结果双歧杆菌刺激后,哮喘儿童DC表面CD86表达明显增高(P〈0.05),HLA-DR表达无明显变化(P〉0.05),对照组儿童CD86和HLA-DR表达无明显影响;LPS刺激可明显增加哮喘儿童和对照组儿童CD86和HLA-DR的表达。结论过敏性哮喘儿童DC表面CD86的表达可能存在缺陷,双歧杆菌能适度上调其表达,在DC的成熟过程中可能起调节作用。

Abstract:Objective To study the effects of Bifidobacterium on the expression of CD86 and HLA-DR in dendritic cells (DCs) derived from peripheral blood mononuclear ceils (PBMCs) of the children with allergic asthma. Method The PBMC-derived DCs were generated in the presence of granulocyte macrophage-colony stimulating factor (GM-CSF) and in- terleukin-4 (IL-4) from 12 patients with allergic asthma and 10 normal children,and then the immature DCs were eocul- tured with the Bifidobacterium adolescent or Lipopelysaccharide (LPS) for 48 h. The expression of CD86 and HLA-DR of DCs were measured by flow cytometer. Result After the pretreatment with Bifidobacterium adolescent, the expression of CD86 of I)Cs from the allergic patients was significantly increased ( P 〈 0.05 ) , but the expression of HLA-DR was not changed ( P 〉 0.05). The pretreatment with Bifidobacterium adolescent had no effects on the expression of CD86 and HLA- DR on the healthy children. The pretreatment with LPS may increase the expression of CD86 and HLA-DR from both the al- lergic patients and healthy children significantly ( P 〈 0.05 ). Conclusion Both Bifidobacterium adolescent and LPS could stimulate the mature of DCs. The expression of CD86 of DCs from the patients with allergic asthma may be impaired. The Bifidobacterium adolescent may up-regulate the expression of CD86 appropriately.
