荷瘤裸鼠暴露于PM2.5后肠道菌群的急性改变

摘    要:目的研究暴露于PM2.5后的肺癌荷瘤裸鼠肠道菌群发生的急性改变。方法 18只Bac裸鼠随机分为PM2.5暴露组和对照组,每组9只。全部裸鼠应用A549细胞腋下注射后,饲养1周让其适应环境,第2周开始于动式染尘暴露仓中暴露染尘,对照组暴露于生理盐水。每天暴露2h,每周暴露6d,共暴露3周。利用16S rDNA分析技术对粪便标本的PCR产物进行高通量测序。结果暴露于PM2.5的肺癌荷瘤裸鼠肠道菌群结构发生改变,与对照组荷瘤裸鼠肠道菌群相比,其厚壁菌门、拟杆菌门细菌显著减少,而致病菌变形菌门细菌增加。结论 PM2.5可以导致肺癌荷瘤裸鼠肠道菌群失衡。

Acute changes of intestinal flora after exposure to PM2.5 in nude mice bearing tumor
Abstract:Abstract: Objective To explore the acute changes of intestinal flora in tumor-bearing nude mice exposed to PM2.5. Methods 18 Bac nude mice were randomly divided into PM2.5 group or control group, 9 mice in each group. All the mice were injected subaxillarily A549 cells and fed for one week to make them adapt to the environment. From the second week, the mice were exposed to dust and normal saline for 3 weeks, 6 days a week, 2 hours a day. The high throughput sequencing of PCR products from fecal specimens was carried out by using 16S rDNA analysis. Results The structure of intestinal microbiota in tumor-bearing nude mice after exposure to PM2.5 was changed. Compared with the control group, the amounts of Streptomyces and Bacteroidetes significantly reduced, while that of pathogenic bacteria Proteobacteria phylum increased. Conclusion PM2.5 can cause intestinal flora imbalance in tumor-bearing nude mice.
