两种提取肠道微生物总DNA方法的比较

目的比较肠道微生物总DNA两种提取方法的优缺点。方法采用苯酚-氯仿抽提法和试剂盒法提取肠道微生物总DNA,比较其作为模板扩增细菌16S DNA的优缺点。结果两种方法提取的细菌DNA均能用于后续PCR扩增的模板。酚-氯仿抽提法经济可靠,但提取的DNA量及纯度较低试;剂盒法提取方便,操作简单,质量稳定,易标准化,但成本高。结论两种方法各具优缺点,应根据实验室条件和实验要求进行选择。

Two methods for total DNA extraction from intestinal microorganisms

Objective To compare two methods of total DNA extraction from intestinal microorganisms.Method Total DNA of intestinal microorganisms were extracted with two different methods： the phenol-chloroform extraction method and the extraction method with kit.Result The products by both the two methods were suitable for PCR amplification.The phenol-chloroform extraction method was relatively cheaper and more reliable,but the amount and purity of the product were much lower.Extraction with kit was convenient to operate and standardized,with the quality of product being stable,but the cost was more expensive.Conclusion Each of the two methods has its advantages and disadvantages and should be chosen based on the lab condition and research requirement.
