羊骨木瓜蛋白酶水解物对小鼠免疫功能的影响

摘    要:目的探讨不同剂量的羊骨木瓜蛋白酶水解物对健康小鼠免疫功能的影响。方法按体质量将ICR小鼠分成对照组(0g/(kg·bw))及低(0.5g/(kg·bw))、中(1.0g/(kg·bw))、高(3.0g/(kg·bw))3个剂量组,分别以碳粒廓清试验和溶血试验评价水解物对非特异性免疫和体液免疫的影响;以噻唑蓝分光光度法评价低(0.01mg/mL)、中(0.1mg/mL)、高(1mg/mL)不同浓度水解物对细胞免疫功能的影响。结果只有中剂量水解物才能显著提高吞噬细胞的吞噬能力;3个剂量组小鼠脾细胞的抗体生成量均显著高于对照组,但高剂量组显著低于中、低剂量组;3个浓度的酶解物均能促进体外ConA诱导的T淋巴细胞增殖活性,但高浓度的促分化效果不如低浓度和中浓度。结论羊骨木瓜蛋白酶水解物能增强小鼠的特异性和非特异性免疫功能,但这种免疫促进作用与剂量并不呈线性关系。

Abstract:Objective To evaluate the effect of papain hydrolysates originated from sheep bone on the immunity of healthy mice.Methods ICR mice were divided by body mass into control (0 g/kg),low-dose (0.5 g/kg),mediumdose (1.0 g/kg) and high-dose (3.0 g/kg) groups.Carbon clearance test and quantitative SRBC hemolysis test were used to determine the impact of the hydrolysates on mice innate immunity and humoral immunity,respectively,and MTT assay was used to determine the effect of low (0.01 mg /mL),medium (0.1 mg /mL),high (1 mg /mL) concentration of the hydrolysates on the cell-mediated immunity (CMI).Results Only the hydrolysate of medium dose group significantly enhanced the phagocytic capacity of macrophages.The amounts of antibodies produced by splenic cells of all dosage groups were markedly higher than that of the control group,while antibodies produced by high-dose group were significantly lower than the other two groups.Supplementation of different concentrations of hydrolysates significantly enhanced lymphocyte proliferation in response to ConA stimulation in vitro,but high concentration was not as good as low and medium concentration.Conclusions Sheep bone collagen papain hydrolysates has a potential to enhance both the specific and innate immunity of normal mice,but this kind of immunomodulatory activity shows no linear relationship with the dosage administrated.
