同型半胱氨酸/半胱氨酸诱导血管平滑肌细胞中的新cDNA

摘    要:同型半胱氨酸/半胱氨酸是体内的含硫氨基酸,近年来提出它们可以诱导血管平滑肌细胞的增殖,是心血管病的一个新的独立的危险因子.但其机制尚不了解,本研究目的在于克隆同型半胱氨酸/半胱氨酸诱导的新基因.应用差异显示即RNAmapping的方法,从半胱氨酸诱导的大鼠血管平滑肌细胞中,寻找有差异的条带,克隆新的cDNA.结果分离出7种上调和4种下调cDNA,并且发现其中有4个是至今尚未发现的新的cDNA.应用Northernblot分析,这4个新的mRNA其长度分别为5.0、2.0、1.8和2.2kb.它们可能在血管平滑肌细胞增殖和心血管病的发病中具有重要意义.

Abstract:Recent years,a number of studies indicates that homocysteine/cysteine(HCY/CY),a sulfhydryl group containing amino acid,can induce the proliferation of vascular smooth muscle cells (VSMCs).Therefore,HCY/CY is a new independent risk factor for cardiovascular diseases.The mechanisms by which HCY/CY may promote vascular diseases,however,have not been illuminated.To understand the molecular basis of mechanisms,differential display,a mRNA mapping method was applid to identify and then clone novel cDNAs that are differentially expressed in normal versus HCY/CY treated rat VSMCs.Seven up regulated and four down regulated cDNAs were identified and cloned.Four of them does not have data base match and represent parts of novel cDNAs.Nothern blot analyses showed that the sizes of the four novel mRNAs are about 5 0,2 0,1 8 and 2 2 kb,respectively.They have been accepted by GenBank and the accession numbers of them are U94859,U94860,U94861,and U94862,respectively.Four novel cDNAs induced with CY were isolated.They may involve in the proliferation of VSMCs and in the occurrence of cardiovascular diseases.
