脊髓全横断损伤后差异表达蛋白的蛋白质组学分析

摘    要: 对脊髓全横断损伤前后的大鼠脊髓全蛋白质进行双向凝胶电泳,借助PDQuest软件从中找出差异表达蛋白质点.应用基质辅助激光解吸电离串联质谱,对差异表达的蛋白质点进行鉴定,成功鉴定出18种蛋白质.脊髓损伤3 d后表达上调的蛋白质有巨噬细胞游走抑制因子、S期激酶相关蛋白 1、热休克蛋白 27、多配体蛋白聚糖 3、T细胞受体β链可变区、膜联蛋白Ⅲ、腺苷酸激酶 1、半乳凝素 3、丙酮酸脱氢酶、磷脂酶 B、嗜铬粒蛋白 A、热休克蛋白70凝结蛋白 1;同时表达下调的蛋白质有磷酸丙糖异构酶、神经鞘氨醇磷酸化受体、热休克蛋白10、肽酰 脯氨酰 顺反式异构酶 A多数差异蛋白质涉及到神经细胞的增殖、凋亡、应激反应等过程,为进一步阐明中枢神经系统的损伤和修复机制提供了理论依据.摘要 对脊髓全横断损伤前后的大鼠脊髓全蛋白质进行双向凝胶电泳,借助PDQuest软件从中找出差异表达蛋白质点.应用基质辅助激光解吸电离串联质谱,对差异表达的蛋白质点进行鉴定,成功鉴定出18种蛋白质.脊髓损伤3 d后表达上调的蛋白质有巨噬细胞游走抑制因子、S期激酶相关蛋白 1、热休克蛋白 27、多配体蛋白聚糖 3、T细胞受体β链可变区、膜联蛋白Ⅲ、腺苷酸激酶 1、半乳凝素 3、丙酮酸脱氢酶、磷脂酶 B、嗜铬粒蛋白 A、热休克蛋白70凝结蛋白 1;同时表达下调的蛋白质有磷酸丙糖异构酶、神经鞘氨醇磷酸化受体、热休克蛋白10、肽酰 脯氨酰 顺反式异构酶 A多数差异蛋白质涉及到神经细胞的增殖、凋亡、应激反应等过程,为进一步阐明中枢神经系统的损伤和修复机制提供了理论依据.

Abstract:The total proteins of normal and(traumatic) injured spinal cord tissues from rats were extracted and separated by 2-DE,and the images analysis were performed with PDQuest software.The differentially expressed proteins were identified by matrix-assisted laser desorption/ionization quadrupole time –off-light mass spectrometry(MALDI-Q-TOF MS),and a total of 18 differential expression proteins were successfully identified.Three days after spinal cord injury,there were 13 up-regulated proteins such as macrophage migration inhibitory factor(MIF),heatshock protein 27(HSP27),syndecan-3(SDC3),T-cell receptor beta chain precursorVregion(CRTB4),annexi(ANXⅢ),galectin-3(LEG3),pyruvate dehydrogenase(ODPA),chromogranin A precursor(CgA), HSP70-bindingprotein1(HSPBP1).Meanwhile,triose-phosphate isomerase(TPIS),sphingosine 1-phosphate receptor 1(S1P1),heat-shock protein 10(HSP10),peptidyl-prolyl cis-trans isomerase A(PPIaseA) were down-regulated.Most of the differentially expressed proteins have beenreported to participate in neuron growth,apoptosis and stress-reaction.The data will be helpful to illuminate the mechanisms involved in the progress of injury and recovery in central nervous system.
