钜齿远蚓(Amynthas dancataa)纤溶酶的分离纯化及其若干性质

 以野生型钷齿远蚓为材料,组织匀浆后,经生理盐水抽提,硫酸铵分级沉淀,葡聚糖凝胶过滤和DEAE离子交换层析,得到两种纯的蚯蚓溶酶,具有强烈的溶解纤维蛋白的作用。它们都是糖蛋白,非寡聚酶,分子量分别为23,000、40,000。测定了一个酶的氨基酸组成,它对某些底物的作用,被一些抑制剂抑制的程度,说明它是练氨酸蛋白酶类、胰蛋白酶类酶

THE PURIFICATION AND CHARACTERIZATION OF FIBRINOLYTIC ENZYMES FROM AMYNTHAS DANCATALA

Two fibrinolytic enzymes were isolated and purified from earthworm Amynthas dancataal. The procedure of purification included saline extraction, ammonium sulfate precipitation, Sephadex gel filtration and ion-exchange chromatography. About ten components with fibrinolytic activity from earthworm could be separated. Two purified components were proved to be homogenous in SDS-PAGE electropho-resis with molecular weights 23,000 and 40,000. They have strong thrombolytic action on fibrin and thrombin. Some chemical and physical properties were characterized, they were confirmed to be glycoprotein and non-oligomeric enzymes. Their action on some substrates and the effect of various inhibitors on the enzymes have been studied. The enzymes seem to be serine type protease with trypsin-like activities.
