正常人卵巢绒毛膜促性腺激素(hCG)受体的研究

摘    要:23例正常卵巢制备之细胞膜,可与绒毛膜促性腺激素(hCG)发生特异性结合,最大结合率为13.2±2.24%,Scatchard作图分析,得一直线。23例正常卵巢之受体量为O.66±0.142×10~(-10)M/μg膜蛋白,Kd值为10.16±5.5×10~(-9)M。

Abstract:Human chorionic gonadotropin (hCG) is the major physiological luteotropic factor for the human corpus luteum. The observations strongly suggest that the human ovary possesses a goaadotropin receptor. Because the binging hCG to receptor is the first step in luteotropic. we studied the hCG receptor in normai human ovary.The plasma membranes of ovarian cell was prepared by sucrose density gradient centrifugation. hCG binding to receptors in human ovary from 23 normai subjects were studied. The highest binding activity was obtained with 40 μg membrane protein. The results demonstrated that a maximum of 13.2±2.24% of 125I-hCG was bound to plasma membrane in the abseuce of unlabeled hCG. The average receptor content in ovarian membrane was 0.66±0.142×10-10M per μg membrane protein. The Kd (dissociation constants) of hCG recep-lor in ovarian plasma membrane for 25I-hCG was 10.16±5.5×10-9M.In this studies, the average hCG content in blood from 13 normai persons was 5.14±2.59 IU and the average number of hCG receptor sites was 0.68±0.16×10-10M per μg protein.
