Abstract:Trichosanthin(TCS) is a single chain ribosome-inactivating protein isolated from the root tuber of Trichosanthes kirilowii Maxim and it has RNA N-glycosidase activity. Differencespectroscopic study revealed that Ciacron blue F_3GA bound to TCS (Kd= 0. 94 μmol/L),porducing a specific difference spectrum with a single maximum at 700 nm and two minima at 595 and 635 nm,which suggested that the binding of TCS with the dye was specific. MediTication of Lys residues in TCS with succinic anhydride caused the loss of difference absorbance preduced by the dye-TCS interaction. The modified TCS was much easily eluted from the column of BlueSepharose 4B than native TCS,showing that the affinity of TCS with the dye decreased after the modification of Lys residues.
