摘    要:本文研究了用PEG修饰的McAb的某些理化性质。研究结果表明:修饰的McAb与天然的McAb相比有如下几点变化:1.随修饰度增高,分子量增大;2.随修饰度增高,PAGE与SDS-PAGE泳动速度减慢;3.修饰的McAb等电点发生酸移;4.等速电泳呈峰高降低与泳动速度稍快的变化;5.园二色性光谱无明显的构象变化。修饰的PcAb与酶,除修饰酶与天然酶的园二色性光谱有明显不同外,其它的变化与修饰的McAb相似。

Abstract:In this article,data on some physico-chemical properties of murine anti-TNP-human albumin monoclonal antibodies (McAb) modified by polyethylene gly-col (PEG-1900) were presented. As the result of modification,following changes were found in the PEG-modified McAb. (1) an increated molecular weight,(2) decreased mobilities both in PAGE and in SDS-PAGE,(3) the isoelectric point became more acidic in IEF (4) the single peak decreased in height and migrated more rapidly in isotachoporesis and (5) no spparant changes in circular dichroism spectra. Modification of polyclonal antibodies (PcAb,horse anti-human IgG) and an enzyme (bovine liver catalase) by PEG-1900 under the same condition brought about similar changes,except that there were striking changes in circular dichroism spectra for catalase. The possible mechanisms involved were discussed .
